Peanut seeds are currently widely used as source of human food ingredients in the United States of America and in European countries due to their high quality protein and oil content. This article describes the classification and molecular biology of peanut seed allergens with particular reference to their cross-reactivities. Currently, the IUIS allergen nomenclature subcommittee accepts 12 peanut allergens. Two allergens belong to the cupin and four to the prolamin superfamily, and six are distributed among profilins, Bet v 1-like proteins, oleosins, and defensins. Clinical observations frequently report an association of peanut allergy with allergies to legumes, tree nuts, seeds, fruits and pollen. Molecular cross-reactivity has been described between members of the Bet v 1-like proteins, the non-specific lipid transfer proteins, and the profilins. This review also addresses the less well-studied cross-reactivity between cupin and prolamin allergens of peanuts and of other plant food sources and the recently discovered cross-reactivity between peanut allergens of unrelated protein families.

Oral cytarabine ocfosfate pharmacokinetics and assessment of leukocyte biomarkers in normal dogs

Abstract
Background
Cytosine arabinoside (Ara-C) is a nucleoside analog prodrug utilized for immunomodulatory effects mediated by its active metabolite Ara-CTP. Optimal dosing protocols for immunomodulation in dogs have not been defined. Cytarabine ocfosfate (CO) is a lipophilic prodrug of Ara-C that can be administered PO and provides prolonged serum concentrations of Ara-C.
Objectives
Provide pharmacokinetic data for orally administered CO and determine accumulation and functional consequences of Ara-CTP within peripheral blood leukocytes.
Animals
Three healthy female hound dogs and 1 healthy male Beagle.
Methods
Prospective study. Dogs received 200 mg/m2 of CO PO q24h for 7 doses. Serum and cerebrospinal fluid (CSF) CO and Ara-C concentrations were measured by liquid chromatography-tandem mass spectroscopy (LC-MS/MS). Complete blood counts, flow cytometry, and leukocyte activation assays were done up to 21 days. Incorporation of Ara-CTP within leukocyte DNA was determined by LC-MS/MS.
Results
Maximum serum concentration (Cmax) for Ara-C was 456.1-724.0 ng/mL (1.88-2.98 μM) and terminal half-life was 23.3 to 29.4 hours. Cerebrospinal fluid: serum Ara-C ratios ranged from 0.54 to 1.2. Peripheral blood lymphocyte concentrations remained within the reference range, but proliferation rates poststimulation were decreased at 6 days. Incorporation of Ara-CTP was not saturated and remained >25% of peak concentration at 13 days.
Conclusions and Clinical Importance
Oral CO may produce prolonged serum Ara-C half-lives at concentrations sufficient to induce functional changes in peripheral leukocytes and is associated with prolonged retention of DNA-incorporated Ara-CTP. Application of functional and active metabolite assessment is feasible and may provide more relevant data to determine optimal dosing regimens for Ara-C-based treatments.

High-affinity allergen-specific human antibodies cloned from single IgE B cell transcriptomes

Immunoglobulin E (IgE) antibodies play a central role in immune responses against helminth and protozoan parasites; however, they also contribute to allergies. IgE antibodies (and the B cells generating them) are rare and thus poorly characterized. Croote et al. performed single-cell RNA sequencing of peripheral blood B cells from patients with peanut allergies and delineated each cell's gene expression, splice variants, and antibody sequences (see the Perspective by Gould and Ramadani). Unlike other isotypes, circulating IgE B cells were mostly immature plasmablasts. Surprisingly, certain IgE antibodies manifested identical gene rearrangements in unrelated individuals. These IgE antibodies showed high affinity and unexpected cross-reactivity to peanut allergens. Science , this issue p. 1306 ; see also p. 1247 Single-cell sequencing of IgE B cells from allergic individuals reveals surprising insights. Immunoglobulin E (IgE) antibodies protect against helminth infections but can also cause life-threatening allergic reactions. Despite their role in human health, the cells that produce these antibodies are rarely observed and remain enigmatic. We isolated single IgE B cells from individuals with food allergies and used single-cell RNA sequencing to elucidate the gene expression and splicing patterns unique to these cells. We identified a surprising example of convergent evolution in which IgE antibodies underwent identical gene rearrangements in unrelated individuals. Through the acquisition of variable region mutations, these IgE antibodies gained high affinity and unexpected cross-reactivity to the clinically important peanut allergens Ara h 2 and Ara h 3. These findings provide insight into IgE B cell transcriptomics and enable biochemical dissection of this antibody class.

Mitigation of Major Peanut Allergens by Pulsed Ultraviolet Light

Peanut allergy represents one of the most severe IgE-mediated reactions with food, but to date, the only effective way to prevent peanut allergy is total avoidance. If allergens could be mitigated during food processing before a product reaches the consumer, this would substantially lessen the food allergy problem. The efficacy of pulsed ultraviolet light (PUV), a novel food processing technology, on reducing peanut allergens, was examined. This study revealed for the first time that PUV was also capable of deactivating Ara h 2, the most potent allergenic protein of peanut. Protein extracts from raw and roasted peanuts were treated for 2, 4, and 6 min and peanut butter slurry was treated for 1, 2, and 3 min in a Xenon Steripulse XL 3000® PUV system. The distance from the central axis of the lamp was varied at 10.8, 14.6, and 18.2 cm. The SDS–PAGE showed a reduction in the protein band intensity for Ara h 1, Ara h 2, and Ara h 3 at the energy levels ranging from 111.6 to 223.2 J/cm
2
. Reduction of the protein band intensity for peanut allergens increased with treatment time but decreased with increased distance from the PUV lamp. The ELISA for peanut extracts and peanut butter slurry showed a reduction in IgE binding of up to 12.9- and 6.7-folds, respectively, compared to control.

Analysis of a Robot Selection Problem Using Two Newly Developed Hybrid MCDM Models of TOPSIS-ARAS and COPRAS-ARAS

Traditional Multi-Criteria Decision Making (MCDM) methods have now become outdated; therefore, most researchers are focusing on more robust hybrid MCDM models that combine two or more MCDM techniques to address decision-making problems. The authors attempted to create two novel hybrid MCDM systems in this paper by integrating Additive Ratio ASsessment (ARAS) with Technique for Order of Preference by Similarity to Ideal Solution (TOPSIS) and Complex PRoportional ASsessment (COPRAS). To demonstrate the ability and effectiveness of these two hybrid models i.e., TOPSIS-ARAS and COPRAS-ARAS were applied to solve a real-time robot selection problem with 12 alternative robots and five selection criteria, while evaluating the parametric importance using the CRiteria Importance Through Inter criteria Correlation (CRITIC) objective weighting estimation tool. The rankings of the robot alternatives gained from these two hybrid models were also compared to the obtained results from eight other solo MCDM tools. Although the rankings by the applied methods slightly differ from each other, the final outcomes from all of the adopted techniques are consistent enough to suggest that robot 12 is the best choice followed by robot 11, and robot 4 is the worst one among these 12 alternatives. Spearman Correlation Coefficient (SCC) also reveals that the proposed rankings derived from various methods have a strong ranking relationship with one another. Finally, sensitivity analysis was performed to investigate the effects of weight variation and to validate the robustness of the implemented MCDM approaches.

In this study, we aim to investigate certain triple integral transform and its application to a class of partial differential equations. We discuss various properties of the new transform including inversion, linearity, existence, scaling and shifting, etc. Then, we derive several results enfolding partial derivatives and establish a multi-convolution theorem. Further, we apply the aforementioned transform to some classical functions and many types of partial differential equations involving heat equations, wave equations, Laplace equations, and Poisson equations as well. Moreover, we draw some figures to illustrate 3-D contour plots for exact solutions of some selected examples involving different values in their variables.

Response and Toxicity to Cytarabine Therapy in Leukemia and Lymphoma: From Dose Puzzle to Pharmacogenomic Biomarkers

Cytarabine is a pyrimidine nucleoside analog, commonly used in multiagent chemotherapy regimens for the treatment of leukemia and lymphoma, as well as for neoplastic meningitis. Ara-C-based chemotherapy regimens can induce a suboptimal clinical outcome in a fraction of patients. Several studies suggest that the individual variability in clinical response to Leukemia & Lymphoma treatments among patients, underlying either Ara-C mechanism resistance or toxicity, appears to be associated with the intracellular accumulation and retention of Ara-CTP due to genetic variants related to metabolic enzymes. Herein, we reported (a) the latest Pharmacogenomics biomarkers associated with the response to cytarabine and (b) the new drug formulations with optimized pharmacokinetics. The purpose of this review is to provide readers with detailed and comprehensive information on the effects of Ara-C-based therapies, from biological to clinical practice, maintaining high the interest of both researcher and clinical hematologist. This review could help clinicians in predicting the response to cytarabine-based treatments.

The Fourth Industrial Revolution is a complex phenomenon characterized by rapid technological changes that significantly transform social and economic landscapes, particularly in the workplaces. This transformation creates a gap between university curricula and the skill sets needed in the job market. If educational institutions do not adapt, they risk obsolescence, making effective curriculum revision essential. This study investigates the impact of the Fourth Industrial Revolution on education in engineering, focusing on its implications for human resource training. The Fuzzy SWARA approach was employed to assess the severity, likelihood, and detectability of educational challenges, while the Fuzzy ARAS method prioritized these issues. Key challenges identified include weaknesses in instructional content development and university incompatibility, which require immediate attention. The proposed model’s effectiveness was compared to the FMEA method, demonstrating its superiority. Strategies to address these challenges include enhancing curriculum quality, promoting exchange programs with leading universities, and fostering local technology manufacturing. The findings aim to improve methods for bridging the skills gap, ensuring students acquire the necessary skills for their current and future roles.

Applications on Double ARA–Sumudu Transform in Solving Fractional Partial Differential Equations

In this article, we apply the double ARA–Sumudu transformation (DARA-ST) to the nonlocal fractional Caputo derivative operator. We achieve interesting results and implement them to solve certain classes of fractional partial differential equations (FPDEs). Several physical applications are discussed and analyzed, such as telegraph, Klein–Gordon and Fokker–Planck equations. The new technique with DARA-ST is efficient and accurate in examining exact solutions of FPDEs. In order to show the applicability of the presented method, some numerical examples and figures are illustrated. A symmetry analysis is used to verify the results.
