A Small Family of Chloroplast Atypical Thioredoxins1CWOA
The reduction and the formation of regulatory disulfide bonds serve as a key signaling element in chloroplasts. Members of the thioredoxin (Trx) superfamily of oxidoreductases play a major role in these processes. We have characterized a small family of plant-specific Trxs in Arabidopsis (Arabidopsis thaliana) that are rich in cysteine and histidine residues and are typified by a variable noncanonical redox active site. We found that the redox midpoint potential of three selected family members is significantly less reducing than that of the classic Trxs. Assays of subcellular localization demonstrated that all proteins are localized to the chloroplast. Selected members showed high activity, contingent on a dithiol electron donor, toward the chloroplast 2-cysteine peroxiredoxin A and poor activity toward the chloroplast NADP-malate dehydrogenase. The expression profile of the family members suggests that they have distinct roles. The intermediate redox midpoint potential value of the atypical Trxs might imply adaptability to function in modulating the redox state of chloroplast proteins with regulatory disulfides.
A Framework for Efficient and Adaptable Post-Disaster Shelters: Streamlining Material Provisions through Modular Scaffolds
Providing shelter after disasters is a complex, multi-stage process. In the early aftermath, emergency shelters must be secured to meet basic physical needs of affected communities. However, the subsequent provision of various types of temporary accommodation requires new resources. Agencies and governments mobilize significant supplies during the phases of relief, often for a protracted period time, facing inefficiencies and challenges of wasted resources. This paper aims to review the persistent issues of post-disaster shelter processes to explore how the relief efforts could be more sustainable. The study derives a technical framework for shelter adaptability and efficiency based on architectural principles of flexibility to examine how material supply from early emergency stages could contribute to efficient transitional shelter processes. Following a review of literature, the study drew analogies with theories of Open Building and flexibility to propose guidelines for a design and building method. Through an analytical description, the paper draws up an actual shelter proposal using the derived principles. The analysis suggests that the early-stage provision of adaptable shelters, conducive to the concept of incremental transitional building processes, could optimize resources and logistics, reduce waste, enhancing the efficiency, adaptability, and overall sustainability of post-disaster shelter responses.

Exploring data-driven innovation: What’s missing in the relationship between big data analytics capabilities and supply chain innovation?
Data-driven innovations (DDI) have significantly impacted firms’ operations thanks to the massive exploitation of huge data. However, to leverage big data and achieve supply chain innovation, a variety of complementary resources are necessary. In this study, we hypothesise that supply chain innovation (SCI) is dependent on firms’ big data analytics capabilities (BAC). Furthermore, we propose that this relation is mediated by two crucial capabilities of agility and adaptability that enable firms to efficiently meet the challenges of supply chain ambidexterity. Finally, we also test the moderating role of technology uncertainty in our research model. We collected data from 386 manufacturing firms in Pakistan and tested our model using structural equation modelling. The results confirmed our initial hypotheses that agility and adaptability both mediated our baseline relationship of BAC and big data innovation in supply chains. We further found support for the moderating role of technology uncertainty. Furthermore, technology uncertainty moderates the relationship between BAC and SCI. This study extends the current literature on digital analytics capabilities and innovation along the supply chain. Practically, our research suggests that investment in big data can result in affirmative consequences, if firms cultivate capabilities to encounter supply chain ambidexterity through agility and adaptability. Accordingly, we suggest that managers belonging to manufacturing firms need to build up these internal capabilities and to monitor and assess technology uncertainty in the environment.
