The goal of this work was to design mixed vegetable (vegetable-fruit) juices which are preserved by high pressure, have an increased content of bioactive substances, are made from locally available produce, and offer excellent nutrition and appealing taste. The new products were prepared on laboratory scale units and underwent nutritional, microbiological, and sensory evaluations. The basic composition, total polyphenol content, ascorbic acid content, and total antioxidant activity were determined.

Oranges, carrot, red beet, and parsley were used for the study of the influence of packing nets on the quality of fruit colour. Different nets were tested. The measurements of lightness and chromaticity parameters were performed according to the L*a*b* system. The red net used for the packing of oranges caused a decrease of the lightness parameter L* for all stages of fruits maturity. However, some unripe and bright oranges, through the use of a red net, looked more mature. The chromaticity parameter a* is the most influential factor affecting the human perception of the fruit colour. The red net improves this parameter significantly, thus unripe yellow oranges become more saturated, changing the perception of colour to the predicted range.

Changes in the level and activation state of the plasma membrane H+-ATPase during aging of red beet slices

The effect of aging on the plasma membrane (PM) H+-ATPase of red beet (Beta vulgaris L.) parenchyma discs was analyzed in PM purified by aqueous two-phase partitioning. Aging increased both the activity and the amount of immunodetectable H+-ATPase in the PM. The activity assayed at slightly alkaline pH values increased earlier and more strongly than that assayed at acidic pH values, so that the pH curve of the enzyme from aged beet discs was shifted toward more alkaline values. Aging decreased the stimulation of the PM H+-ATPase activity by controlled trypsin treatments or by lysophosphatidylcholine. After trypsin treatment the pH dependence of H+-ATPase from dormant or aged beet discs became equal. These results indicate that aging not only increases the level of H+-ATPase in the PM, but also determines its activation, most likely by modifying the interaction between the autoinhibitory carboxyl-terminal domain and the catalytic site. When the PM H+-ATPase activity was assayed at a slightly alkaline pH, the tyrosine modifier N-acetylimidazole inhibited the H+-ATPase in the PM from dormant beet discs much less than in the PM from aged discs, suggesting that modification of a tyrosine residue may be involved in the activation of the PM H+-ATPase induced by aging. The results are discussed with regard to aging-induced development of transmembrane transport activities

The effect of different antioxidants on stability of betalains was measured in solution at 70, 80 and 90 deg C, pH 5.0. Ascorbic acid, extracts from balm (Melissa officinalis) and oregano (Origanum) had similar effects in improving betalains stability.

An enriched glucan synthase fraction was obtained from red beet root microsomes by sequential extraction with the detergents 3-[(3-cholamidopropyl)dimethylammonio]-1-propanesulfonate and digitonin. The digitonin suspension was centrifuged on a glycerol gradient, where a glucan synthase peak with a specific activity of 30- to 40-fold over microsomes was recovered. Most protein contaminants were found in the gradient pellet. The glucan synthase-containing fraction was largely free of plasma membrane and tonoplast-derived ATPase activity and was enriched with a protein subunit of 68 kilodaltons.
