Filamentous fungi isolated from Tunisian olive mill wastes: use of solid-state fermentation for enzyme production

Olive mill wastewaters and olive cake are effluents generated during olive oil production process. They represent a major disposal and potentially severe pollution problem for the industry, also promising source of substances of high value. The aim of this study is the valorization of olive mill wastes (OMWW, olive cake, olive twigs and leaves) to produce enzymes with high industrial and biotechnological potential, by the solid-state fermentation technique (SSF), from isolated fungi present in olive mill wastewater and olive cake. A total of 47 strains were isolated and purified from these two residues. The metabolic potential of isolated strains was study by testing the hydrolytic enzymes activities of lipase, protease, amylase, cellulase, invertase, phytase and tannase on agar plate media containing different substrate. The monitoring of SSF has shown that the metabolic activity of these strains is extremely rapid using this technique. Our fungi collection contains a diversity of strains capable to producing a variety of enzymes of biotechnological interest.

Green synthesis, biomedical and biotechnological applications of carbon and graphene quantum dots. A review

Carbon and graphene quantum dots are prepared using top-down and bottom-up methods. Sustainable synthesis of quantum dots has several advantages such as the use of low-cost and non-toxic raw materials, simple operations, expeditious reactions, renewable resources and straightforward post-processing steps. These nanomaterials are promising for clinical and biomedical sciences, especially in bioimaging, diagnosis, bioanalytical assays and biosensors. Here we review green methods for the fabrication of quantum dots, and biomedical and biotechnological applications.

Endophytic microbes: biodiversity, plant growth-promoting mechanisms and potential applications for agricultural sustainability

Endophytic microbes are known to live asymptomatically inside their host throughout different stages of their life cycle and play crucial roles in the growth, development, fitness, and diversification of plants. The plant–endophyte association ranges from mutualism to pathogenicity. These microbes help the host to combat a diverse array of biotic and abiotic stressful conditions. Endophytic microbes play a major role in the growth promotion of their host by solubilizing of macronutrients such as phosphorous, potassium, and zinc; fixing of atmospheric nitrogen, synthesizing of phytohormones, siderophores, hydrogen cyanide, ammonia, and act as a biocontrol agent against wide array of phytopathogens. Endophytic microbes are beneficial to plants by directly promoting their growth or indirectly by inhibiting the growth of phytopathogens. Over a long period of co-evolution, endophytic microbes have attained the mechanism of synthesis of various hydrolytic enzymes such as pectinase, xylanases, cellulase, and proteinase which help in the penetration of endophytic microbes into tissues of plants. The effective usage of endophytic microbes in the form of bioinoculants reduce the usage of chemical fertilizers. Endophytic microbes belong to different phyla such as Actinobacteria, Acidobacteria, Bacteroidetes, Deinococcus–thermus, Firmicutes, Proteobacteria, and Verrucomicrobia. The most predominant and studied endophytic bacteria belonged to Proteobacteria followed by Firmicutes and then by Actinobacteria. The most dominant among reported genera in most of the leguminous and non-leguminous plants are Bacillus, Pseudomonas, Fusarium, Burkholderia, Rhizobium, and Klebsiella. In future, endophytic microbes have a wide range of potential for maintaining health of plant as well as environmental conditions for agricultural sustainability. The present review is focused on endophytic microbes, their diversity in leguminous as well as non-leguminous crops, biotechnological applications, and ability to promote the growth of plant for agro-environmental sustainability.

Surfactants are a broad category of tensio-active biomolecules with multifunctional properties applications in diverse industrial sectors and processes. Surfactants are produced synthetically and biologically. The biologically derived surfactants (biosurfactants) are produced from microorganisms, with Pseudomonas aeruginosa, Bacillus subtilis Candida albicans, and Acinetobacter calcoaceticus as dominant species. Rhamnolipids, sophorolipids, mannosylerithritol lipids, surfactin, and emulsan are well known in terms of their biotechnological applications. Biosurfactants can compete with synthetic surfactants in terms of performance, with established advantages over synthetic ones, including eco-friendliness, biodegradability, low toxicity, and stability over a wide variability of environmental factors. However, at present, synthetic surfactants are a preferred option in different industrial applications because of their availability in commercial quantities, unlike biosurfactants. The usage of synthetic surfactants introduces new species of recalcitrant pollutants into the environment and leads to undesired results when a wrong selection of surfactants is made. Substituting synthetic surfactants with biosurfactants resolves these drawbacks, thus interest has been intensified in biosurfactant applications in a wide range of industries hitherto considered as experimental fields. This review, therefore, intends to offer an overview of diverse applications in which biosurfactants have been found to be useful, with emphases on petroleum biotechnology, environmental remediation, and the agriculture sector. The application of biosurfactants in these settings would lead to industrial growth and environmental sustainability.

Green Synthesis of Metallic Nanoparticles and Their Prospective Biotechnological Applications: an Overview

The green synthesis of nanoparticles (NPs) using living cells is a promising and novelty tool in bionanotechnology. Chemical and physical methods are used to synthesize NPs; however, biological methods are preferred due to its eco-friendly, clean, safe, cost-effective, easy, and effective sources for high productivity and purity. High pressure or temperature is not required for the green synthesis of NPs, and the use of toxic and hazardous substances and the addition of external reducing, stabilizing, or capping agents are avoided. Intra- or extracellular biosynthesis of NPs can be achieved by numerous biological entities including bacteria, fungi, yeast, algae, actinomycetes, and plant extracts. Recently, numerous methods are used to increase the productivity of nanoparticles with variable size, shape, and stability. The different mechanical, optical, magnetic, and chemical properties of NPs have been related to their shape, size, surface charge, and surface area. Detection and characterization of biosynthesized NPs are conducted using different techniques such as UV–vis spectroscopy, FT-IR, TEM, SEM, AFM, DLS, XRD, zeta potential analyses, etc. NPs synthesized by the green approach can be incorporated into different biotechnological fields as antimicrobial, antitumor, and antioxidant agents; as a control for phytopathogens; and as bioremediative factors, and they are also used in the food and textile industries, in smart agriculture, and in wastewater treatment. This review will address biological entities that can be used for the green synthesis of NPs and their prospects for biotechnological applications.

Harnessing Bacterial Endophytes for Promotion of Plant Growth and Biotechnological Applications: An Overview

Endophytic bacteria colonize plants and live inside them for part of or throughout their life without causing any harm or disease to their hosts. The symbiotic relationship improves the physiology, fitness, and metabolite profile of the plants, while the plants provide food and shelter for the bacteria. The bacteria-induced alterations of the plants offer many possibilities for biotechnological, medicinal, and agricultural applications. The endophytes promote plant growth and fitness through the production of phytohormones or biofertilizers, or by alleviating abiotic and biotic stress tolerance. Strengthening of the plant immune system and suppression of disease are associated with the production of novel antibiotics, secondary metabolites, siderophores, and fertilizers such as nitrogenous or other industrially interesting chemical compounds. Endophytic bacteria can be used for phytoremediation of environmental pollutants or the control of fungal diseases by the production of lytic enzymes such as chitinases and cellulases, and their huge host range allows a broad spectrum of applications to agriculturally and pharmaceutically interesting plant species. More recently, endophytic bacteria have also been used to produce nanoparticles for medical and industrial applications. This review highlights the biotechnological possibilities for bacterial endophyte applications and proposes future goals for their application.

The Plant Invertase/Pectin Methylesterase Inhibitor Superfamily

Invertases (INVs) and pectin methylesterases (PMEs) are essential enzymes coordinating carbohydrate metabolism, stress responses, and sugar signaling. INVs catalyzes the cleavage of sucrose into glucose and fructose, exerting a pivotal role in sucrose metabolism, cellulose biosynthesis, nitrogen uptake, reactive oxygen species scavenging as well as osmotic stress adaptation. PMEs exert a dynamic control of pectin methylesterification to manage cell adhesion, cell wall porosity, and elasticity, as well as perception and signaling of stresses. INV and PME activities can be regulated by specific proteinaceous inhibitors, named INV inhibitors (INVIs) and PME Inhibitors (PMEIs). Despite targeting different enzymes, INVIs and PMEIs belong to the same large protein family named “Plant Invertase/Pectin Methylesterase Inhibitor Superfamily.” INVIs and PMEIs, while showing a low aa sequence identity, they share several structural properties. The two inhibitors showed mainly alpha-helices in their secondary structure and both form a non-covalent 1:1 complex with their enzymatic counterpart. , there are now several characterized INVI/PMEIs in different plant species. This review provides an integrated and updated overview of this fascinating superfamily, from the specific activity of characterized isoforms to their specific functions in plant physiology. We also highlight INVI/PMEIs as biotechnological tools to control different aspects of plant growth and defense. Some isoforms are discussed in view of their potential applications to improve industrial processes. A review of the nomenclature of some isoforms is carried out to eliminate confusion about the identity and the names of some INVI/PMEI member. Open questions, shortcoming, and opportunities for future research are also presented.

Alcohol dehydrogenases (ADHs), which belong to the oxidoreductase superfamily, catalyze the interconversion between alcohols and aldehydes or ketones with high stereoselectivity under mild conditions. ADHs are widely employed as biocatalysts for the dynamic kinetic resolution of racemic substrates and for the preparation of enantiomerically pure chemicals. This review provides an overview of biotechnological applications for ADHs in the production of chiral pharmaceuticals and fine chemicals.

Phycobiliproteins are photosynthetic light-harvesting pigments isolated from microalgae with fluorescent, colorimetric and biological properties, making them a potential commodity in the pharmaceutical, cosmetic and food industries. Hence, improving their metabolic yield is of great interest. In this regard, the present review aimed, first, to provide a detailed and thorough overview of the optimization of culture media elements, as well as various physical parameters, to improve the large-scale manufacturing of such bioactive molecules. The second section of the review offers systematic, deep and detailed data about the current main features of phycobiliproteins. In the ultimate section, the health and nutritional claims related to these bioactive pigments, explaining their noticeable potential for biotechnological uses in various fields, are examined.

Flavonols are a subclass of natural flavonoids characterized by a remarkable number of biotechnological applications and health-promoting properties. They attract researchers’ attention due to many epidemiological studies supporting their usage. They are phytochemicals commonly present in our diet, being ubiquitous in the plant kingdom and, in particular, relatively very abundant in fruits and vegetables. All these aspects make flavonols candidates of choice for the valorization of products, based on the presence of a remarkable number of different chemical structures, each one characterized by specific chemical features capable of influencing biological targets inside the living organisms in very different manners. In this review, we analyzed the biochemical and physiological characteristics of flavonols focalizing our attention on the most promising compounds to shed some light on their increasing utilization in biotechnological applications in processing industries, as well as their suitable employment to improve the overall wellness of the humankind.
