Phytochemical Profiles and Antioxidant Activities in Six Species of Ramie Leaves

Increased consumption of vegetables or plant food has been associated with decreased risk of developing major chronic diseases, such as cancers, diabetes, cardiovascular diseases, and age-related functional decline. Ramie leaves are rich in phenolics and flavonoids, which have been suggested for human health benefits. Phenolic contents, flavonoid contents, phenolic compounds, and anti-cancer properties in six species of ramie leaves were analyzed by Folin-reagent method, sodium borohydride/chloranil-based assay (SBC), HPLC method and antiproliferation, cytoxicity, respectively. Antioxidant activities were measured through peroxyl radical scavenging capacity (PSC) method, oxygen radical absorbance capacity (ORAC) method, and cellular antioxidant activity (CAA). Research indicated that Boehmeria penduliflora contained the highest total phenolic content (2313.7±27.28 mg GAE/100 g FW), and flavonoid content (1682.4±27.70 mg CAE/100 g FW). Boehmeria tricuspis showed the highest PSC value (9574.8±117.63 µM vit. C equiv./100 g FW), while Boehmeria penduliflora indicated the highest ORAC value (330.44±16.88 µmol Trolox equiv./g FW). The antioxidant activities were correlated with phenolic contents and flavonoid contents. Boehmeria tricuspis had the highest antiproliferative capacity with the lowest EC₅₀ (4.11±0.19 mg/mL). The results for the analyzed ramie for CAA were significantly different from each other (p<0.05), Boehmeria tricuspis had the highest CAA value (133.63±7.10 µmol QE/100 g). Benzoic acid, 4-coumaric acid, caffeic acid, and ferulic acid were the dominant phenolic ingredients in the ramie leaves according to HPLC analysis. Our research is the first report to study the phytochemical profiles and antioxidant activities in different species of ramie leaves for their health benefit.

Development and Characterization of 1,827 Expressed Sequence Tag-Derived Simple Sequence Repeat Markers for Ramie (Boehmeria nivea L. Gaud)

Ramie (Boehmeria nivea L. Gaud) is one of the most important natural fiber crops, and improvement of fiber yield and quality is the main goal in efforts to breed superior cultivars. However, efforts aimed at enhancing the understanding of ramie genetics and developing more effective breeding strategies have been hampered by the shortage of simple sequence repeat (SSR) markers. In our previous study, we had assembled de novo 43,990 expressed sequence tags (ESTs). In the present study, we searched these previously assembled ESTs for SSRs and identified 1,685 ESTs (3.83%) containing 1,878 SSRs. Next, we designed 1,827 primer pairs complementary to regions flanking these SSRs, and these regions were designated as SSR markers. Among these markers, dinucleotide and trinucleotide repeat motifs were the most abundant types (36.4% and 36.3%, respectively), whereas tetranucleotide, pentanucleotide, and hexanucleotide motifs represented <10% of the markers. The motif AG/CT was the most abundant, accounting for 28.74% of the markers. One hundred EST-SSR markers (97 SSRs located in genes encoding transcription factors and 3 SSRs in genes encoding cellulose synthases) were amplified using polymerase chain reaction for detecting 24 ramie varieties. Of these 100 markers, 98 markers were successfully amplified and 81 markers were polymorphic, with 2-6 alleles among the 24 varieties. Analysis of the genetic diversity of all 24 varieties revealed similarity coefficients that ranged from 0.51 to 0.80. The EST-SSRs developed in this study represent the first large-scale development of SSR markers for ramie. These SSR markers could be used for development of genetic and physical maps, quantitative trait loci mapping, genetic diversity studies, association mapping, and cultivar fingerprinting.

Analysis of bacterial and fungal communities in continuous-cropping ramie (Boehmeria nivea L. Gaud) fields in different areas in China

L. Gaud) suffers from long-term continuous cropping. Here, using Illumina high-throughput sequencing technology, we aimed to identify bacteria and fungi associated with continuous cropping in ramie fields in Yuanjiang, Xianning, Sichuan, and Jiangxi. The rarefaction results showed that Jiangxi had significantly lower bacterial α-diversity than that of the other areas. Firmicutes, Proteobacteria, and Acidobacteria were the dominant bacterial phyla, and Ascomycota, Basidiomycota, and Zygomycota were the dominant fungal phyla. In Jiangxi, Firmicutes accounted for 79.03% of all valid reads, which could have significant decreased microbial diversity and negative effects of continuous ramie cropping. We used traditional methods to examine soil nutrients. Sichuan had a relatively high pH and available P and K, but low total N; opposite findings were recorded in Jiangxi. The redundancy analysis revealed that the urease activity, PH, available K, and total N significantly correlated with bacterial community abundance, whereas only total N significantly correlated with fungal community abundance (P < 0.01). Overall, the effect of soil environmental factors on the bacterial diversity of continuous ramie cropping was greater than that on fungal diversity. In the future, we will focus on the effect of rhizosphere bacteria to solve the obstacle in continuous ramie cropping.

Boehmeria, as currently circumscribed, comprises 52 species and has a pantropical distribution. Liang et al. propose a sectional classification of Boehmeria based on the phylogenetic analysis of SNP data for 20 species and an additional 10 subspecific taxa of these at the rank of variety or form. They restrict their sampling to species documented in China. We found many shortcomings in the sampling and analyses which we feel have resulted in a misleading phylogeny for the genus and the economically important fibre-plant, Boehmeria nivea. By sampling only Chinese species of this genus for their in-group and using a single distantly related outgroup, Liang et al. have failed to capture the diversity of the genus and so erroneously concluded that it forms a monophyletic group. Previous published research clearly demonstrates that Boehmeria is paraphyletic and polyphyletic, comprising at least four monophyletic groupings most closely related to several genera within the Boehmerieae. For these reasons, the sections that Liang et al. (Ind Crops Prod 148:112092, 2020. https://doi.org/10.1016/j.indcrop.2020.112092) propose for Boehmeria are not effective tools for its classification. The important fibreplant, Boehmeria nivea, should therefore not be considered as part of the genus Boehmeria for the purposes of crop breeding, but as sister to Archiboehmeria. Breeding programmes for ramie should therefore focus on populations and germplasm of Archiboehmeria atrata. We conclude that poor taxon sampling, overlooking relevant molecular and taxonomic literature, internal conflict within their SNP data and the overinterpretation of low support values has resulted in the erroneous conclusion that Boehmeria represents a monophyletic or ‘natural’ genus.

Background
Ramie (
Boehmeria nivea
L.) is one of the most important natural fiber crops and an important forage grass in south China. Ramet number, which is a quantitative trait controlled by multigenes, is one of the most important agronomic traits in plants because the ramet number per plant is a key component of grain yield and biomass. However, the genetic variation and genetic architecture of ramie ramet number are rarely known.
Results
A genome-wide association study was performed using a panel of 112 core germplasms and 108,888 single nucleotide polymorphisms (SNPs) detected using specific-locus amplified fragment sequencing technology. Trait-SNP association analysis detected 44 significant SNPs that were associated with ramet number at
P
< 0.01. The favorable SNP Marker20170–64 emerged at least twice in the three detected stages and was validated to be associated with the ramie ramet number using genomic DNA polymerase chain reaction with an F
1
hybrid progeny population. Comparative genome analysis predicted nine candidate genes for ramet number based on Marker20170–64. Real-time quantitative polymerase chain reaction analysis indicated that six of the genes were specific to upregulation in the ramie variety with high ramet number. These results suggest that these genes could be considered as ramet number-associated candidates in ramie.
Conclusions
The identified loci or genes may be promising targets for genetic engineering and selection for modulating the ramet number in ramie. Our work improves understanding of the genetics of ramet number in ramie core germplasms and provides tools for marker-assisted selection for improvement of agricultural traits.

Cryphodera sinensis n. sp. (Nematoda: Heteroderidae), a non-cyst-forming parasitic nematode from the root of ramie Boehmeria nivea in China

Cryphodera sinensis n. sp. is described from ramie (Boehmeria nivea) based on the morphology and molecular analyses of rRNA small subunit (SSU), D2D3 expansion domains of large subunit (LSU D2D3) and internal transcribed spacer (ITS). This new species is characterized by oval females with a distinct subcrystalline layer and pronounced and protruding vulval lip, distinctly concave vulva–anus profile and a vulva–anus distance of 29.5–35.8 μm. Males possess two annuli in the lip region, a stylet 27–32.5 μm in length with round knobs sloping slightly posteriorly, lateral fields with three lines, spicules 20–28 μm long and the presence of a short cloacal tube. Second-stage juveniles possess three lip annuli, a stylet 28–31 μm in length with well-developed knobs projected anteriorly and three lines along the lateral field. The pointed tail, 52–65 μm long, possesses a mucro-like tip and a hyaline region, 24.5–35 μm long. Large phasmids with a lens-like structure are located 2–6 annuli posterior to the anus. Phylogenetic analysis shows that the species has unique SSU, LSU D2D3 and ITS rRNA sequences. Phylogenetic relationships of the three rDNA sequences of C. sinensis n. sp. and other cystoid/cyst nematodes are analysed together with a comparison of other species within the genus Cryphodera.

Characteristics of Polygalacturonate Lyase C from Bacillus subtilis 7-3-3 and Its Synergistic Action with PelA in Enzymatic Degumming

An alkaline polygalacturonate lyase (PGL) from Bacillus subtilis 7-3-3, PelC, with diverse depolymerization abilities for different pectin substrates was found. The PGL activity of PelC decreased with increasing degree of methyl esterification of the substrate. PelA and PelC displayed notable synergistic effects in the enzymatic degumming of ramie fibers. Gum loss rates increased by 62% when PelC was used to replace up to three-eighths of the PelA dose (PelC, 60 U g(-1) ramie fibers). To the best of our knowledge, this study is the first to report the synergistic action of members of polysaccharide lyase families 1 and 3, represented by PelA and PelC, respectively. The present paper provides new insights into the improvement and production of enzymes used in enzymatic degumming.

Identification of small auxin-up RNA (SAUR) genes in Urticales plants: mulberry (Morus notabilis), hemp (Cannabis sativa) and ramie (Boehmeria nivea)

Small auxin-up RNA (SAUR) genes are important gene families in auxin signalling transduction and are commonly used as early auxin responsive markers. Till date, no SAUR gene is identified in Urticales plants despite of the published bioinformation of mulberry, hemp and ramie. In this study, we used Arabidopsis sequences as query to search against mulberry, hemp genomes and ramie transcriptome database. In total, we obtained 62, 56 and 71 SAUR genes in mulberry, hemp and ramie, respectively. Phylogenetic analysis revealed the Urticales specific expansion of SAUR genes. Expression analysis showed 15 randomly selected ramie SAUR genes that were diversely functioned in ramie tissues and revealed a series of IAA-responsive, drought-responsive and high temperature-responsive genes. Moreover, comparison of qRT-PCR data and previous RNA-Seq data suggested the reliability of our work. In this study, we first report the identification of SAUR genes in Urticales plants. These results will provide a foundation for their function validation in Urticales plant growth and development.

A New Species of Andesipolis Whitfield & Choi from the Eastern Andes of Ecuador with Notes on Biology and Classification (Hymenoptera: Braconidae: Rhysipolinae)

A new species of braconid wasp, Andesipolis yanayacu, is described from the eastern Andes of Ecuador. Andesipolis yanayacu was reared as a gregarious koinobiont parasitoid of shelter building Pyralidae (Lepidoptera) larvae feeding on Urticaceae (Phenax rugosus and Boehmeria bullata). These are the first biological observations for the genus Andesipolis and the first species recorded from Ecuador. This is also the northern-most record for the genus as previously described species are from Chile. Based on morphological attributes and the newly discovered biology, Andesipolis is re-classified from the subfamily Mesostoinae into the subfamily Rhysipolinae.
