Expression and Function of the Protein Tyrosine Phosphatase Receptor J (PTPRJ) in Normal Mammary Epithelial Cells and Breast Tumors
The protein tyrosine phosphatase receptor J, PTPRJ, is a tumor suppressor gene that has been implicated in a range of cancers, including breast cancer, yet little is known about its role in normal breast physiology or in mammary gland tumorigenesis. In this paper we show that PTPRJ mRNA is expressed in normal breast tissue and reduced in corresponding tumors. Meta-analysis revealed that the gene encoding PTPRJ is frequently lost in breast tumors and that low expression of the transcript associated with poorer overall survival at 20 years. Immunohistochemistry of PTPRJ protein in normal human breast tissue revealed a distinctive apical localisation in the luminal cells of alveoli and ducts. Qualitative analysis of a cohort of invasive ductal carcinomas revealed retention of normal apical PTPRJ localization where tubule formation was maintained but that tumors mostly exhibited diffuse cytoplasmic staining, indicating that dysregulation of localisation associated with loss of tissue architecture in tumorigenesis. The murine ortholog, Ptprj, exhibited a similar localisation in normal mammary gland, and was differentially regulated throughout lactational development, and in an in vitro model of mammary epithelial differentiation. Furthermore, ectopic expression of human PTPRJ in HC11 murine mammary epithelial cells inhibited dome formation. These data indicate that PTPRJ may regulate differentiation of normal mammary epithelia and that dysregulation of protein localisation may be associated with tumorigenesis.

Jeff David has been appointed managing director and chief executive officer of Greencross Ltd, which today completed its acquisition of Mammoth Pet Holdings Pty Ltd, creating \"Australasia's largest integrated consumer-facing pet care company\". He replaces Glen Richards, who will remain on the Greencross board. Mr David is a founder of Mammoth, served as a non-executive director of Greencross since 2007 and is a non-executive director of the Skin Cancer Foundation Australia. Cossack Energy Ltd today announced the appointment of Robert Bensh and Stuart Brown as non-executive directors. Mr Bensh and Mr Brown are highly regarded and significantly experienced, with Mr Bensh having specific Ukraine political and capital raising experience and Mr Brown having specific technical experience. As foreshadowed on November 14, Mirvac Group non-executive director James MacKenzie has resigned from the board. The effective date for Mr MacKenzie's resignation was 5pm on January 30. He has also resigned from the board's Audit, Risk and Compliance Committee.

It is already possible to use biodiesel (in various percentages) in all Case IH equipment, and the company is continually working on engine technology so that future biodiesel engines will be as gutsy and productive as their conventional diesel equivalents. Stuart says Case IH is interested in any project that would advance the biodiesel cause. \"The grant-winning application might be for direct research into how to produce different types of biodiesel, or it could be broader, perhaps looking at developing Australian standards for biodiesel, or working on retro-fit filtration systems for fuel storage tanks to make them suitable for biodiesel. It's a relatively new area of research and there are many avenues to be explored.\"
Centro looks for 6mo extension to pay off debt, Pelorus postpone
It is understood that all eightf of Centro's Australian lenders agree the best way forward is to extend the April 30 deadline to September 30, giving Centro more time to repay its Australian-based debt. However, no formal agreement has been reached. Centro's new chief executive, Glenn Rufrano, has previously said he does not want to offload assets, but would prefer to receive an equity injection to help pay down part of Centro's debt obligations. \"We have gone through due diligence with [CENTRO]'s syndicates and the major problem is the lack of transparency in relation to the perceived conflicts of interest between the various roles Centro has undertaken in the syndicates.\"
