Background
Cinnamomum camphora
has been cultivated as an economically important tree for its medicinal and aromatic properties. Selective breeding has produced
Cinnamomum
plants for special uses, including spice strains with characteristic flavors and aromas and high-potency medicinal cultivars. The molecular biology underlying terpenoid biosynthesis is still unexplored.
Results
Gas chromatography-mass spectrometry was used to analyze the differences in contents and compositions of essential oil terpenoids in linalool- and borneol-type chemotypes of
C. camphora
. The data revealed that the essential oils consist primarily of monoterpenes with only very minor quantities of sesquiterpenes and diterpenes and that the essential oil differs in different chemotypes of
C. camphora
, with higher yields of (−)-borneol from the borneol-type than from the linalool-type. To study the terpenoid biosynthesis of signature compounds of the major monoterpenes, we performed RNA sequencing to profile the leaf transcriptomes of the two chemotypes of
C. camphora
. A total of 23.76 Gb clean data was generated from two chemotypes and assembled into 156,184 unigenes. The total length, average length, N50 and GC content of unigenes were 155,645,929 bp, 997 bp, 1430 bp, and 46.5%, respectively. Among them, 76,421 unigenes were annotated by publicly available databases, of which 67 candidate unigenes were identified to be involved in terpenoid biosynthesis in
C. camphora
. A total of 2863 unigenes were identified to be differentially expression between borneol-type and linalool-type, including 1714 up-regulated and 1149 down-regulated unigenes. Most genes encoding proteins involved in terpenoid precursor MVA and MEP pathways were expressed in similar levels in both chemotypes of
C. camphora
. In addition, 10 and 17 DEGs were significantly enriched in the terpene synthase activity and oxidoreductase activity terms of their directed acyclic graphs (DAG), respectively. Three monoterpene synthase genes,
TPS14-like1, TPS14-like2
and
TPS14-like3
were up-regulated in the borneol-type compared to the linalool-type, and their expression levels were further verified using quantitative real-time PCR.
Conclusions
This study provides a global overview of gene expression patterns related to terpenoid biosynthesis in
C. camphora
, and could contribute to a better understanding of the differential accumulation of terpenoids in different
C. camphora
chemotypes.

Chemical Variation and Environmental Influence on Essential Oil of Cinnamomum camphora

Cinnamomum camphora is a traditional aromatic plant used to produce linalool and borneol flavors in southern China; however, its leaves also contain many other unutilized essential oils. Herein, we report geographic relationships for the yield and compositional diversity of C. camphora essential oils. The essential oils of 974 individual trees from 35 populations in 13 provinces were extracted by hydrodistillation and analyzed qualitatively and quantitatively by gas chromatography-mass spectrometry and gas chromatography-flame ionization detection, respectively. Oil yields ranged from 0.01% to 3.46%, with a significantly positive correlation with latitude and a significantly negative correlation with longitude. In total, 41 compounds were identified, including 15 monoterpenoids, 24 sesquiterpenoids, and two phenylpropanoids. Essential oil compositions varied significantly among individuals and could be categorized into various chemotypes. The six main chemotypes were eucalyptol, nerolidol, camphor, linalool, selina, and mixed types. The other 17 individual plants were chemotypically rare and exhibited high levels of methyl isoeugenol, methyl eugenol, δ-selinene, or borneol. Eucalyptol-type plants had the highest average oil yield of 1.64%, followed in decreasing order by linalool-, camphor-, mixed-, selina-, and nerolidol-type plants. In addition, the five main compounds exhibited a clear geographic gradient. Eucalyptol and linalool showed a significantly positive correlation with latitude, while selina-6-en-4-ol was significantly and negatively correlated with latitude. trans-Nerolidol and selina-6-en-4-ol showed significantly positive correlations with longitude, whereas camphor was significantly and negatively correlated with longitude. Canonical correspondence analysis indicated that environmental factors could strong effect the oil yield and essential oil profile of C. camphora.

Phytochemicals and antioxidant activity of leaf extract and callus cultures of Cinnamomum camphora L

Cinnamomum camphora L. is highly significant landscape tree known for its medicinal values and presence of secondary metabolites that have antioxidant, antimicrobial, anticancer, anti-inflammatory effects and widely utilized in pharmaceutical and cosmetic industry. Callus cultures of C. camphora have better antioxidant activity than extracts of naturally grown tree leaves. While there is significant lack of research about the potential of its in vitro callus cultures as a controlled and sustainable biotechnological alternative for mass production of bioactive compounds. The present work was aimed at comparative analysis of phytochemicals (phenolic and flavonoid) and antioxidant activities of calli and leaf extract of field grown camphor plant. To get in vitro germplasm, callus formation and direct shoot initiation was carried out and it was observed that MS medium supplemented with 0.1 mg L -1 thidiazuron (TDZ) + 0.5 mg L -1 2, 4-dichlorophenoxyacetic acid (2, 4-D) proved best for shoot initiation from nodal explant. MS medium fortified with various plant growth regulators was used for callus formation and best callus induction response (100%) from nodal and leaf explants was observed on 0.5 mg L -1 2, 4-D) + 2.0 mg L -1 6-Benzylaminopurine (BAP). Callus was successfully sub-cultured and this in vitro proliferated calli and fresh leaf extract of field grown plant were used for comparative study of phytochemicals. Results revealed that callus culture exhibited highest antioxidant enzyme activities compared to leaf extract and hence there was statistically significant ( P ≤ 0.05 ) difference. Similarly, peroxidase (31.12 UmL -1 of enzyme), superoxide dismutase (35.24 Umg -1 of protein), and catalase (58.6 UmL -1 of enzyme) also showing highest vales. In contrast, glutathione peroxidase activity (0.552 Umg -1 ) was comparatively higher in leaf extract. Additionally, callus cultures accumulated higher phenolic contents (1.106 mg GAE g -1 of FW) while flavonoid contents (7.87 mg CatE g -1 of FW) were higher in leaf extract. This investigation showed that in vitro conditions and the use of plant growth regulators in various combinations might be work as elicitors to enhance the phytochemicals and antioxidant enzymes in callus culture as compared to the leaf extract.

To investigate the chemical composition and insecticidal activity of the essential oils of certain Chinese medicinal herbs and spices, the essential oils were extracted from the stem barks, leaves, and fruits of Cinnamomum camphora (L.) Presl, which were found to possess strong fumigant toxicity against Tribolium castaneum and Lasioderma serricorne adults. The essential oils of the plants were extracted by the method of steam distillation using a Clavenger apparatus. Their composition was determined by gas chromatography/mass spectrometric (GC-MS) analyses (HP-5MS column), and their insecticidal activity was measured by seal-spaced fumigation. D-camphor (51.3%), 1,8-cineole (4.3%), and α-terpineol (3.8%), while D-camphor (28.1%), linalool (22.9%), and 1,8-cineole (5.3%) were the main constituents of its fruits. The essential oils of the C. camphora all showed fumigant and contact toxicity. Other compounds exhibited various levels of bioactivities. The results indicate that the essential oils of C. camphora and its individual compounds can be considered a natural resource for the two stored-product insect management.

GC×GC-TOFMS Analysis of Essential Oils Composition from Leaves, Twigs and Seeds of Cinnamomum camphora L. Presl and Their Insecticidal and Repellent Activities

Interest in essential oils with pesticidal activity against insects and pests is growing. In this study, essential oils from different parts (leaves, twigs and seeds) of Cinnamomum camphora L. Presl were investigated for their chemical composition, and insecticidal and repellent activities against the cotton aphid. The essential oils, obtained by hydrodistillation, were analyzed by GC×GC-TOFMS. A total of 96 components were identified in the essential oils and the main constituents found in the leaves and twigs were camphor, eucalyptol, linalool and 3,7-dimethyl-1,3,7-octatriene. The major components found in the seeds were eucalyptol (20.90%), methyleugenol (19.98%), linalool (14.66%) and camphor (5.5%). In the contact toxicity assay, the three essential oils of leaves, twigs and seeds exhibited a strong insecticidal activity against cotton aphids with LC50 values of 245.79, 274.99 and 146.78 mg/L (after 48 h of treatment), respectively. In the repellent assay, the highest repellent rate (89.86%) was found in the seed essential oil at the concentration of 20 μL/mL after 24 h of treatment. Linalool was found to be a significant contributor to the insecticidal and repellent activities. The results indicate that the essential oils of C. camphora might have the potential to be developed into a natural insecticide or repellent for controlling cotton aphids.

Many essential oils (EOs) regulate the quorum-sensing (QS) system of pathogens and inhibit the virulence expression. Interference with QS can potentially reduce bacterial multidrug resistance and aid the biological control of bacterial disease. In the present work, the antibacterial and anti-QS activities of Cinnamomum camphora leaf EO were investigated. A total of 23 chemical components with relative levels ≥0.11%, including a large number of terpene compounds, were identified in C. camphora leaf EO by gas chromatography–mass spectrometry (GC-MS). The principal component was linalool, followed by eucalyptol, with relative levels of 51.57% and 22.07%, respectively. The minimum inhibitory concentration (MIC) and antibacterial activity of C. camphora EO were examined, and P. aeruginosa and E. coli ATCC25922 showed the highest and lowest sensitivity to C. camphora EO, respectively. Tests of QS inhibitory activity revealed that C. camphora EO significantly decreased the production of violacein and biofilm biomass in C. violaceum, with the maximum inhibition rates of 63% and 77.64%, respectively, and inhibited the biofilm formation and swarming movement, independent of affecting the growth of C. violaceum. Addition of C. camphora EO also resulted in downregulation of the expression of the acyl-homoserine lactones (AHL) synthesis gene (cviI) and transcription regulator (cviR), and inhibited the expression of QS-regulated virulence genes, including vioA, vioB, vioC, vioD, vioE, lasA, lasB, pilE3, and hmsHNFR. Collectively, the prominent antibacterial activity and anti-QS activities clearly support that C. camphora EO acts as a potential antibacterial agent and QS inhibitor in the prevention of bacterial contamination.

Much previous research has indicated most composts of pruning waste are characterized by potential phytotoxicity, it is highly correlated with the chemical compounds of raw materials. Cinnamomum camphora, a common kind of pruning waste in Southeast Asia and East Asia, is characterized by intense bioactivities due to complex chemical components. This study investigated the potential phytotoxicity of C. camphora pruning waste in light of germination and higher plant growth. C. camphora extracted from leaves completely inhibited seed germination and still showed suppression of root elongation at an extremely low dosage. C. camphora extract also displayed significant inhibition of nutrient absorption in tomato seedlings, including moisture, available nutrients (N, P and K) and key microelements (Fe, Mn, Zn and S). The gene expression of aquaporins and transporters of nitrate and phosphate was significantly up-regulated in roots. This could be regarded as a positive response to C. camphora extract for enhancing nutrient absorption. Moreover, the severe damage to the plasma membrane in roots caused by C. camphora extract might seriously affect nutrient absorption. Camphor is the main component of the C. camphora extract that may induce the phytotoxicity of plasma membrane damage, resulting in the inhibition of nutrient absorption and low biomass accumulation. This study provided a new understanding of the ecotoxicological effects of C. camphora pruning waste, indicating that the harmless disposal of pruning waste requires much attention and exploration in the future.

Genome-Scale Identification, Classification, and Expression Profiling of MYB Transcription Factor Genes in Cinnamomum camphora

The camphor tree (Cinnamomum camphora (L.) Presl.) is the representative species of subtropical evergreen broadleaved forests in eastern Asia and an important raw material for essential oil production worldwide. Although MYBs have been comprehensively characterized and their functions have been partially resolved in many plants, it has not been explored in C. camphora. In this study, 121 CcMYBs were identified on 12 chromosomes in the whole genome of C. camphora and found that CcMYBs were mainly expanded by segmental duplication. They were divided into 28 subgroups based on phylogenetic analysis and gene structural characteristics. In the promoter regions, numerous cis-acting elements were related to biological processes. Analysis of RNA sequencing data from seven tissues showed that CcMYBs exhibited different expression profiles, suggesting that they have various roles in camphor tree development. In addition, combined with the correlation analysis of structural genes in the flavonoid synthesis pathway, we identified CcMYBs from three subgroups that might be related to the flavonoid biosynthesis pathway. This study systematically analyzed CcMYBs in C. camphora, which will set the stage for subsequent research on the functions of CcMYBs during their lifetime and provide valuable insights for the genetic improvement of camphor trees.

In this study, the efficiency of microwave-assisted hydro-distillation (MAHD) to extract essential oil from Cinnamomum camphora leaf, and the recovery of polyphenols from extract fluid were investigated. The effects of microwave power, liquid-to-material ratio, and extraction time on the extraction efficiency were studied by a single factor test as well as the response surface methodology (RSM) based on the central composite design method. The optimal extraction conditions were a microwave power of 786.27 W, liquid-to-material ratio of 7.47:1 mL/g, and extraction time of 35.57 min. The yield of essential oil was 3.26 ± 0.05% (w/w), and the recovery of polyphenols was 4.97 ± 0.02 mg gallic acid equivalent/g dry weight under the optimal conditions. Furthermore, the comprehensive two-dimensional gas chromatography-time-of-flight mass spectrometry (GC×GC-TOFMS) was used to characterize the essential oils of fresh and fallen leaves, and 159 individual compounds were tentatively identified, accounting for more than 89.68 and 87.88% of the total contents, respectively. The main ingredients include sabinene, l-β-pinene, β-myrcene, α-terpineol, 3-heptanone, and β-thujene, as well as δ-terpineol and 3-heptanone, which were first identified in C. camphora essential oil. In conclusion, the MAHD method could extract essential oil from C. camphora with high efficiency, and the polyphenols could be obtained from the extract fluid at the same time, improving the utilization of C. camphora leaf.

The Key Phytochemical Cue Camphor Is a Promising Lure for Traps Monitoring the New Monophagous Camphor Tree Borer Pagiophloeus tsushimanus (Coleoptera: Curculionidae)
The landscape plant, Cinnamomum camphora, is a broad-spectrum insect-repelling tree species, mainly due to a diversity of terpenoids, such as camphor. Despite its formidable chemical defenses, C. camphora is easily attacked and invaded by a monophagous weevil pest, Pagiophloeus tsushimanus. Deciphering the key olfactory signal components regulating host preference could facilitate monitoring and control strategies for this pest. Herein, two host volatiles, camphor and ocimene, induced GC-EAD/EAG reactions in both male and female adult antennae. Correspondingly, Y-tube olfactometer assays showed that the two compounds were attractive to both male and female adults. In field assays, a self-made trap device baited with 5 mg dose d(+)-camphor captured significantly more P. tsushimanus adults than isopropanol solvent controls without sexual bias. The trunk gluing trap device baited with bait can capture adults, but the number was significantly less than that of the self-made trap device and adults often fell after struggling. The cross baffle trap device never trapped adults. Neither ocimene nor isopropanol solvent control captured adults. When used in combination, ocimene did not enhance the attraction of d(+)-camphor to both female and male adults. These results indicate that d(+)-camphor is a key active compound of P. tsushimanus adults for host location. The combination of the host-volatile lure based on d(+)-camphor and the self-made trapping device is promising to monitor and provide an eco-friendly control strategy for this novel pest P. tsushimanus in C. camphora plantations.
