We are told that under the pressure of increasing globalisation, it is India and China that are the new world manufacturing powerhouses. Rapid and Sensitive Detection of IVerticillium dahliae/I from Soil Using LAMP-CRISPR/Cas12a Technology

Cotton Verticillium wilt is mainly caused by the fungus Verticillium dahliae, which threatens the production of cotton. Its pathogen can survive in the soil for several years in the form of microsclerotia, making it a destructive soil-borne disease. The accurate, sensitive, and rapid detection of V. dahliae from complex soil samples is of great significance for the early warning and management of cotton Verticillium wilt. In this study, we combined the loop-mediated isothermal amplification (LAMP) with CRISPR/Cas12a technology to develop an accurate, sensitive, and rapid detection method for V. dahliae. Initially, LAMP primers and CRISPR RNA (crRNA) were designed based on a specific DNA sequence of V. dahliae, which was validated using several closely related Verticillium spp. The lower detection limit of the LAMP-CRISPR/Cas12a combined with the fluorescent visualization detection system is approximately ~10 fg/μL genomic DNA per reaction. When combined with crude DNA-extraction methods, it is possible to detect as few as two microsclerotia per gram of soil, with the total detection process taking less than 90 min. Furthermore, to improve the method’s user and field friendliness, the field detection results were visualized using lateral flow strips (LFS). The LAMP-CRISPR/Cas12a-LFS system has a lower detection limit of ~1 fg/μL genomic DNA of the V. dahliae, and when combined with the field crude DNA-extraction method, it can detect as few as six microsclerotia per gram of soil, with the total detection process taking less than 2 h. In summary, this study expands the application of LAMP-CRISPR/Cas12a nucleic acid detection in V. dahliae and will contribute to the development of field-deployable diagnostic productions.

Two-dimensional (2D) and three-dimensional (3D) batch electrochemical degradation (ECD) of raw cotton industry wastewater (CIWW) was adopted using stainless steel (SS) and aluminium (Al) electrodes. ECD as a treatment option was aimed at removing priority quality parameters, viz. chemical oxygen demand (COD), colour, chloride, nitrate, etc. COD removal of 85 and 80% were achieved by using 3D SS and 2D SS electrodes operated at 6 V (0.9 A) for a maximum electrolysis time (ET) of 30 min. Similarly, 76 and 70% COD removal were achieved for 3D Al and 2D Al electrodes, respectively. Simultaneous colour removal in the 2D ECD system using SS and Al electrodes was low by 12 and 11% compared to the 3D ECD system. Water quality parameters, viz. total dissolved solids, chloride, nitrate, phosphates, and sulphate were also removed by 3D (SS and Al) and 2D (SS and Al) electrodes. Higher pollutant removal efficiencies were observed at 30 min ET for 3D SS electrodes compared to 2D SS, 3D Al, and 2D Al. Post-ECD slurry showed good settling characteristics for SS electrodes generating dense and sturdy flocs giving a low sludge volume index values for 2D SS electrodes compared to other electrode options.

Genomic Dynamics and Functional Insights under Salt Stress in IGossypium hirsutum/I L

The changing climate is intensifying salt stress globally. Salt stress is a menace to cotton crop quality and yield. The seedling, germination, and emergence phases are more prone to the effects of salt stress than other stages. Higher levels of salt can lead to delayed flowering, a reduced number of fruiting positions, shedding of fruits, decreased boll weight, and yellowing of fiber, all of which have an adverse effect on the yield and quality of the seed cotton. However, sensitivity toward salt stress is dependent on the salt type, cotton growth phase, and genotype. As the threat of salt stress continues to grow, it is crucial to gain a comprehensive understanding of the mechanisms underlying salt tolerance in plants and to identify potential avenues for enhancing the salt tolerance of cotton. The emergence of marker-assisted selection, in conjunction with next-generation sequencing technologies, has streamlined cotton breeding efforts. This review begins by providing an overview of the causes of salt stress in cotton, as well as the underlying theory of salt tolerance. Subsequently, it summarizes the breeding methods that utilize marker-assisted selection, genomic selection, and techniques for identifying elite salt-tolerant markers in wild species or mutated materials. Finally, novel cotton breeding possibilities based on the approaches stated above are presented and debated.
