Neurological effects of subchronic exposure to dioctyl phthalate (DOP), lead, and arsenic, individual and mixtures, in immature mice
Dioctyl phthalate (DOP) (200, 500, and 1000 mg kg −1 bw, i.g.), Pb (Ac) 2 (50 mg L −1 , p.o.), and NaAsO 2 (10 mg L −1 , p.o.) were administered individually and as mixtures to weanling male mice for 8 weeks. It was observed that Pb, As, and DOP exposure could significantly inhibit the growth and development of mice. Compared with the Pb, As, and Pb + As groups, the activities of iNOS and TNOS were significantly increased, the levels of AChE and SOD were significantly decreased, and the level of MDA was significantly increased in the Pb + DOP-H, As + DOP-H, and Pb + As + DOP-H groups. The factorial analysis shows that the iNOS, TNOS, and AChE present synergistic effects on Pb, As, and DOP. A significant increase of escape latency and a significant decrease of original platform quadrant stops were observed between Pb + As + DOP-H and Pb + As groups. The factorial analysis shows that there was a synergistic effect on Pb, As, and DOP. Compared with that of the control group, the expression levels of caspase-3 and Bax expression in Pb + As, DOP-H, Pb + DOP-H, As + DOP-H, and Pb + As + DOP-H groups were significantly increased in the hippocampus. The expression levels of Bcl-2 expression decreased significantly and the Bax/Bcl-2 ratio increased significantly. Pathological alterations on the hippocampus were found in exposed groups. This result shows that combined exposure of Pb, As, and DOP could induce neurotoxicity, of which possible mechanism is hippocampal neuronal apoptosis. Graphical abstract This study shows that there were three components with eigenvalues greater than 1, which together explained 89.40% of total variance. The first component (PC1) showed high loadings on B-SOD, L-SOD, B-MDA, L-MDA, K-MDA, iNOS, tNOS, and AChE and accounted for 46.55% of the total variance after Varimax rotation. PC2 accounted for 23.81% of the total variance with high loadings on B-As, L-As, K-As, and K-SOD, whereas PC3 showed high loadings on B-Pb, L-Pb, and K-Pb and accounted for 19.04% of the total variance.
Research on the Glass Transition Temperature and Mechanical Properties of Poly(vinyl chloride)/Dioctyl Phthalate (PVC/DOP) Blends by Molecular Dynamics Simulations
To effectively improve the performance and expand the applications of polymers, molecular dynamics (MD) simulations with the COMPASS force field have been applied to predict the miscibility, glass transition temperature (Tg), and mechanical properties of poly(vinyl chloride)/dioctyl phthalate (PVC/DOP) blends. The solubility parameter values obtained are in good agreement with the reference data and the little difference (∣Δδ∣ < 2.0 MPa0.5) between two components indicates that PVC/DOP is a miscible system. Tg is predicted by the slope of the free volume and density versus temperature simulation data based on density and free volume theory which is agree well with the experimental data. In addition, the analyses of mechanical properties results indicate that the values of Young’s modulus (E), bulk modulus (K), and shear modulus (G) decrease with the addition of DOP, demonstrating that the rigidity of material is weakened and the ductility is improved. The mechanical properties can also be effectively improved by increasing the temperature, which may provide a more flexible mixture, with lower E, K, G but an increased ductility.
Effect of Oily Aerosol Charge Characteristics on the Filtration Efficiency of an Electrostatically Enhanced Fibrous Filter System
The synergistic effect of electrostatically enhanced fibrous filtration originates from the charging characteristics of aerosol particles and electret fibers in an electric field. Two electrostatically enhanced fibrous filter systems are designed in this study to investigate the mechanism of the effects of the charging characteristics of oily aerosol on the filtration efficiency. We investigate the charging characteristics and their effects on the filtration efficiency of dioctyl-phthalate (DOP) aerosol particles of various sizes by setting different filter systems and electric field intensities. The experimental results show that the charge of DOP particles increases with the strength of the electric field, and the average charge increases with the particle size. The maximum charge of DOP particles reaches 4760 eC/P, and the filtration efficiency of the coupled system improves when DOP particles are amply charged. For 0.25 μm DOP particles as the most penetrating particle size, the system had good long-term stability, and the filtration efficiency is approximately 72% higher than that of the fiber acting alone. Meanwhile, the problem of oily aerosol deposition reducing the electret filtration efficiency is solved, providing a basis for long-term filtration and oily aerosol purification by electret fiber.
Exposure to di-2-ethylhexyl phthalate (DEHP) increases the risk of cancer
Cancer is a major socioeconomic burden that seriously affects the life and spirit of patients. However, little is known about the role of environmental toxicant exposure in diseases, especially ubiquitous di-(2-ethylhexyl) phthalate (DEHP) which is one of the most widely used plasticizers. Hence, the objective of this study was to assess the potential association between cancer and DEHP. The data were collected using the 2011–2018 National Health and Nutrition Examination Survey (NHANES) data ( n  = 6147), and multiple logistic regression was conducted to evaluate the association. The concentrations of DEHP were calculated by each metabolite and split into quartiles for analysis. After adjusting for confounding factors, DEHP was significantly associated with an increased risk of cancer prevalence, and the metabolites of DEHP showed similar results (OR > 1.0, p  < 0.05). Simultaneously, the association remained when the analyses were stratified by age and sex, and the risk of cancer appeared to be higher in male patients. In addition, further analysis suggested that DEHP exposure obviously increased the risk of female reproductive system cancer, male reproductive system cancer, and other cancers (OR > 1.0, p  < 0.05) but not skin and soft tissue cancer. DEHP exposure is associated with the risk of cancer, especially female reproductive system cancer, male reproductive system cancer and other cancers.
A Review of Biomonitoring of Phthalate Exposures
Phthalates (diesters of phthalic acid) are widely used as plasticizers and additives in many consumer products. Laboratory animal studies have reported the endocrine-disrupting and reproductive effects of phthalates, and human exposure to this class of chemicals is a concern. Several phthalates have been recognized as substances of high concern. Human exposure to phthalates occurs mainly via dietary sources, dermal absorption, and air inhalation. Phthalates are excreted as conjugated monoesters in urine, and some phthalates, such as di-2-ethylhexyl phthalate (DEHP), undergo secondary metabolism, including oxidative transformation, prior to urinary excretion. The occurrence of phthalates and their metabolites in urine, serum, breast milk, and semen has been widely reported. Urine has been the preferred matrix in human biomonitoring studies, and concentrations on the order of several tens to hundreds of nanograms per milliliter have been reported for several phthalate metabolites. Metabolites of diethyl phthalate (DEP), dibutyl- (DBP) and diisobutyl- (DiBP) phthalates, and DEHP were the most abundant compounds measured in urine. Temporal trends in phthalate exposures varied among countries. In the United States (US), DEHP exposure has declined since 2005, whereas DiNP exposure has increased. In China, DEHP exposure has increased since 2000. For many phthalates, exposures in children are higher than those in adults. Human epidemiological studies have shown a significant association between phthalate exposures and adverse reproductive outcomes in women and men, type II diabetes and insulin resistance, overweight/obesity, allergy, and asthma. This review compiles biomonitoring studies of phthalates and exposure doses to assess health risks from phthalate exposures in populations across the globe.
Contaminant occurrence, distribution and ecological risk assessment of phthalate esters in the Persian Gulf
Due to the increasing population of the world, the presence of harmful compounds, especially phthalate esters (PAEs), are one of the important problems of environmental pollution. These compounds are known as carcinogenic compounds and Endocrine-disrupting chemicals (EDCs) for humans. In this study, the occurrence of PAEs and the evaluation of its ecological risks were carried out in the Persian Gulf. Water samples were collected from two industrial sites, a rural site and an urban site. Samples were analyzed using magnetic solid phase extraction (MSPE) and gas chromatography-mass spectrometry (GC/MS) technique to measure seven PAEs including Di(2-ethylhexyl) phthalate (DEHP), butyl benzyl phthalate (BBP), diethyl phthalate (DEP), dibutyl phthalate (DBP), Dimethyl phthalate (DMP), di-n-octyl phthalate (DNOP), and Di-iso-butyl phthalate (DIBP). The BBP was not detected in any of the samples. The total concentration of six PAEs (Σ6PAEs) ranged from 7.23 to 23.7 μg/L, with a mean concentration of 13.7μg/L. The potential ecological risk of each target PAEs was evaluated by using the risk quotient (RQ) method in seawater samples, and the relative results declined in the sequence of DEHP >DIBP > DBP > DEP > DMP in examined water samples. DEHP had a high risk to algae, crustaceans and fish at all sites. While DMP and DEP showed lower risk for all mentioned trophic levels. The results of this study will be helpful for the implementation of effective control measures and remedial strategies for PAEs pollution in the Persian Gulf.
Plastics Derived Endocrine Disruptors (BPA, DEHP and DBP) Induce Epigenetic Transgenerational Inheritance of Obesity, Reproductive Disease and Sperm Epimutations
Environmental compounds are known to promote epigenetic transgenerational inheritance of adult onset disease in subsequent generations (F1-F3) following ancestral exposure during fetal gonadal sex determination. The current study was designed to determine if a mixture of plastic derived endocrine disruptor compounds bisphenol-A (BPA), bis(2-ethylhexyl)phthalate (DEHP) and dibutyl phthalate (DBP) at two different doses promoted epigenetic transgenerational inheritance of adult onset disease and associated DNA methylation epimutations in sperm. Gestating F0 generation females were exposed to either the \"plastics\" or \"lower dose plastics\" mixture during embryonic days 8 to 14 of gonadal sex determination and the incidence of adult onset disease was evaluated in F1 and F3 generation rats. There were significant increases in the incidence of total disease/abnormalities in F1 and F3 generation male and female animals from plastics lineages. Pubertal abnormalities, testis disease, obesity, and ovarian disease (primary ovarian insufficiency and polycystic ovaries) were increased in the F3 generation animals. Kidney and prostate disease were only observed in the direct fetally exposed F1 generation plastic lineage animals. Analysis of the plastics lineage F3 generation sperm epigenome previously identified 197 differential DNA methylation regions (DMR) in gene promoters, termed epimutations. A number of these transgenerational DMR form a unique direct connection gene network and have previously been shown to correlate with the pathologies identified. Observations demonstrate that a mixture of plastic derived compounds, BPA and phthalates, can promote epigenetic transgenerational inheritance of adult onset disease. The sperm DMR provide potential epigenetic biomarkers for transgenerational disease and/or ancestral environmental exposures.
Mono-(2-ethyl-5-hydroxyhexyl) phthalate promotes uterine leiomyoma cell survival through tryptophan-kynurenine-AHR pathway activation
Uterine leiomyoma is the most common tumor in women and causes severe morbidity in 15 to 30% of reproductive-age women. Epidemiological studies consistently indicate a correlation between leiomyoma development and exposure to endocrine-disrupting chemical phthalates, especially di-(2-ethylhexyl) phthalate (DEHP); however, the underlying mechanisms are unknown. Here, among the most commonly encountered phthalate metabolites, we found the strongest association between the urine levels of mono(2-ethyl-5-hydroxyhexyl) phthalate (MEHHP), the principal DEHP metabolite, and the risk of uterine leiomyoma diagnosis (n = 712 patients). The treatment of primary leiomyoma and smooth muscle cells (n = 29) with various mixtures of phthalate metabolites, at concentrations equivalent to those detected in urine samples, significantly increased cell viability and decreased apoptosis. MEHHP had the strongest effects on both cell viability and apoptosis. MEHHP increased cellular tryptophan and kynurenine levels strikingly and induced the expression of the tryptophan transporters SLC7A5 and SLC7A8, as well as, tryptophan 2,3-dioxygenase (TDO2), the key enzyme catalyzing the conversion of tryptophan to kynurenine that is the endogenous ligand of aryl hydrocarbon receptor (AHR). MEHHP stimulated nuclear localization of AHR and up-regulated the expression of CYP1A1 and CYP1B1, two prototype targets of AHR. siRNA knockdown or pharmacological inhibition of SLC7A5/SLC7A8, TDO2, or AHR abolished MEHHP-mediated effects on leiomyoma cell survival. These findings indicate that MEHHP promotes leiomyoma cell survival by activating the tryptophan-kynurenine-AHR pathway. This study pinpoints MEHHP exposure as a high-risk factor for leiomyoma growth, uncovers a mechanism by which exposure to environmental phthalate impacts leiomyoma pathogenesis, and may lead to the development of novel druggable targets.
In-situ positive electrode-electrolyte interphase construction enables stable Ah-level Zn-MnO2 batteries
Engineering the formulation of an Mn-based positive electrode is a viable strategy for producing an efficient aqueous zinc-ion battery. However, Mn dissolution and the byproducts result in capacity fading, thus limiting its electrochemical performances. To solve the undesirable issues, the concept of in-situ forming the positive electrode/electrolyte interface on the commercial MnO 2 is designed, with the help of introducing the Dioctyl Phthalate into the ZS-based electrolyte (2 M ZnSO 4  + 0.2 M MnSO 4 ), designated as ZS-DOP electrolyte. An advanced three-dimensional chemical and imaging analysis on a model material reveals the dynamic formation of positive electrode/electrolyte interface. The formed organic interface effectively suppresses the corrosion of the electrolytes with its hydrophobicity, and adjusts the pH value according to Le Chatelier’s Principle to inhibit the production of by-products. Specifically, the pouch cell assembled with the ZS-DOP electrolyte attains a reversible capacity of ~2.5 Ah and powers the unmanned aerial vehicle. Furthermore, photovoltaic energy storage applications deliver a stable capacity of 0.5 Ah and realize the power supply for mobile phones and other electronic devices. Our results facilitate the development of in-situ surface protection on the positive electrode in aqueous zinc-ion battery, providing insights into its practical application. Mn dissolution and unwanted byproducts result in capacity fading of MnO 2 -based aqueous zinc batteries. Here, authors report an in situ-formed interphase on commercial MnO 2 that inhibits dissolution and generation of byproducts, resulting in improved battery performance.
Measurement of phthalate acid esters in non-alcoholic malt beverages by MSPE-GC/MS method in Tehran city: chemometrics
Phthalic acid esters (PAEs) are compounds that are used in the bottle as the main plasticizers. Therefore, the possibility of releasing phthalate esters into beverages is very high and there is a concern to consumer health and monitoring organizations. The aim of this research was to assess the phthalic acid esters (di-n-octyl phthalate (DNOP), butyl benzyl phthalate (BBP), dimethyl phthalate (DMP), diethyl phthalate (DEP) and dibutyl phthalate (DBP), bis(di-ethylhexyl) phthalate (DEHP), and total PAEs) in bottled non-alcoholic malt beverages ( n = 120) by multi-walled carbon nanotubes were magnetized with iron (MWCNT-Fe 3 O 4 ) using gaschromatography/mass spectrometry (GC-MS). The results showed that the highest and the lowest levels of total phthalate esters in samples were 9483.93 and 2412.50 ng/L, respectively. The mean of DEHP which has also been found to be carcinogenic in all samples was lower than 5944.73 ng/L. The highest concentration of DEHP in four samples was upper than 8957.87 ng/L. Perceived limit of detection (LOD) ranged from 13 to 30 ng/L and the limit of quantification (LOQ) ranged from 39 to 90 ng/L. Multivariate techniques and heat map visualization were used to assess the correlation among the type and levels of PAEs with the brand, color, product date, pH, sugar, volume, and gas pressure. Therefore, based on heat map and principal component analysis (PCA) results, the DEHP and total PAEs were the closest accessions, indicating that these variables had similar trends. Based on the results, it can be stated that due to the low average of total phthalate esters in non-alcoholic malt beverages, there is no serious health hazard of these compounds for humans.
