Determining optimal mulching, planting density, and nitrogen application to increase maize grain yield and nitrogen translocation efficiency in Northwest China
Background The combination of mulch with N fertilizer application is a common agronomic technique used in the production of rainfed maize ( Zea mays L. ) to achieve higher yields under conditions of optimum planting density and adequate N supply. However, the combined effects of mulch, planting density, and N fertilizer application rate on plant N uptake and N translocation efficiency are not known. The objective of this study was to quantify the interaction effect of mulch, planting density, and N fertilizer application rate on maize grain yield, N uptake, N translocation, and N translocation efficiency. The experiment was arranged in a randomized complete block design with three factors (2 mulch levels × 2 planting densities × 4 N fertilizer application rates) replicated four times. Results There was a significant interaction among mulch, plant density, and N fertilizer on maize grain yield, kernel number per cob, N uptake, N translocation, and N translocation efficiency. Averaged over the 3 years of the study, total plant N uptake at silking ranged from 79 to 149 kg N ha − 1 with no mulch and from 76 to 178 kg N ha − 1 with mulch. The N uptake at silking in different plant organs ranked as leaf > grain > stem > cob. Averaged across all factors, the highest N translocation was observed in leaves, which was 59.4 and 88.7% higher than observed in stems and ears, respectively. The mean vegetative organ N translocation efficiency averaged over mulch, planting density, and N fertilizer application rate treatments decreased in the order of leaf > stem > cob. Conclusions Mulch, planting density, and N fertilizer application rate not only have significant effects on improving maize grain yield and NUE, but also on N uptake, N translocation, and N translocation efficiency. Our results showed clearly that under high planting density, the combination of mulch and moderate N fertilizer application rate was the optimal strategy for increasing maize grain yield and N use efficiency.
Increasing CO2 threatens human nutrition
Dietary deficiencies of zinc and iron are a major global public health problem. An estimated two billion people suffer these deficiencies causing a loss of 63 million life years annually. Most of these people depend upon grains and legumes as their primary dietary source of zinc and iron. This manuscript reports findings from the analysis of 540 pairs of crop samples grown at contemporary and elevated [CO2] from six different FACE experiments involving six food crops. We tested the nutrient concentrations of the edible portions of rice (Oryza sativa, 18 cultivars), wheat (Triticum aestivum, 8 cultivars), maize (Zea mays, 2 cultivars), soybeans (Glycine max, 7 cultivars), field peas (Pisum sativum, 4 cultivars) and sorghum (Sorghum bicolor, 1 cultivar). In all six experiments, the elevated [CO2] was in the range of 550-584 ppm. Each crop sample grown at elevated [CO2] was paired with an identical cultivar grown under the same conditions but at contemporary [CO2]. Our main outcomes were fractional changes in concentrations of the nutrients between samples grown at elevated and contemporary [CO2] levels, estimated using a linear mixed effects statistical model. We found that elevated [CO2] was associated with significant decreases in the concentrations of zinc and iron in all C3 grasses and legumes. For example, in wheat grains grown at elevated [CO2] compared with contemporary [CO2], zinc decreased 9.6% and iron decreased 5.2%. We also found that elevated [CO2] was associated with lower protein in C3 grasses with a 6.5% decrease in wheat grains and a 7.9% (95% CI: -8.9, -6.9) decrease in rice grains. Elevated [CO2] showed no significant effect on protein in C3 legumes or C4 crops. Response differences between cultivars suggest breeding crops for reduced sensitivity to elevations in atmospheric [CO2]. Such breeding efforts may partly address the new challenges to global health that these findings highlight.
Reducing brassinosteroid signalling enhances grain yield in semi-dwarf wheat
Modern green revolution varieties of wheat ( Triticum aestivum L.) confer semi-dwarf and lodging-resistant plant architecture owing to the Reduced height-B1b ( Rht-B1b ) and Rht-D1b alleles 1 . However, both Rht-B1b and Rht-D1b are gain-of-function mutant alleles encoding gibberellin signalling repressors that stably repress plant growth and negatively affect nitrogen-use efficiency and grain filling 2 – 5 . Therefore, the green revolution varieties of wheat harbouring Rht-B1b or Rht-D1b usually produce smaller grain and require higher nitrogen fertilizer inputs to maintain their grain yields. Here we describe a strategy to design semi-dwarf wheat varieties without the need for Rht-B1b or Rht-D1b alleles. We discovered that absence of Rht-B1 and ZnF-B (encoding a RING-type E3 ligase) through a natural deletion of a haploblock of about 500 kilobases shaped semi-dwarf plants with more compact plant architecture and substantially improved grain yield (up to 15.2%) in field trials. Further genetic analysis confirmed that the deletion of ZnF-B induced the semi-dwarf trait in the absence of the Rht-B1b and Rht-D1b alleles through attenuating brassinosteroid (BR) perception. ZnF acts as a BR signalling activator to facilitate proteasomal destruction of the BR signalling repressor BRI1 kinase inhibitor 1 (TaBKI1), and loss of ZnF stabilizes TaBKI1 to block BR signalling transduction. Our findings not only identified a pivotal BR signalling modulator but also provided a creative strategy to design high-yield semi-dwarf wheat varieties by manipulating the BR signal pathway to sustain wheat production. A strategy that depends on attenuated brassinosteroid signalling is described for the design of semi-dwarf wheat varieties with improved grain yield compared with that of green revolution varieties.
UDP-glucosyltransferase regulates grain size and abiotic stress tolerance associated with metabolic flux redirection in rice
Grain size is an important component trait of grain yield, which is frequently threatened by abiotic stress. However, little is known about how grain yield and abiotic stress tolerance are regulated. Here, we characterize GSA1 , a quantitative trait locus (QTL) regulating grain size and abiotic stress tolerance associated with metabolic flux redirection. GSA1 encodes a UDP-glucosyltransferase, which exhibits glucosyltransferase activity toward flavonoids and monolignols. GSA1 regulates grain size by modulating cell proliferation and expansion, which are regulated by flavonoid-mediated auxin levels and related gene expression. GSA1 is required for the redirection of metabolic flux from lignin biosynthesis to flavonoid biosynthesis under abiotic stress and the accumulation of flavonoid glycosides, which protect rice against abiotic stress. GSA1 overexpression results in larger grains and enhanced abiotic stress tolerance. Our findings provide insights into the regulation of grain size and abiotic stress tolerance associated with metabolic flux redirection and a potential means to improve crops. Increasing grain yield needs to be put in the context of environmental stress. Here, the authors reveal that a UDP-glucosyltransferase is associated with regulation of rice grain size, abiotic stress tolerance, flavonoid-mediated auxin signaling, and redirection of carbon flux to flavonoid glycosides synthesis.
Molecular Genetics and Breeding for Nutrient Use Efficiency in Rice
In the coming decades, rice production needs to be carried out sustainably to keep the balance between profitability margins and essential resource input costs. Many fertilizers, such as N, depend primarily on fossil fuels, whereas P comes from rock phosphates. How long these reserves will last and sustain agriculture remains to be seen. Therefore, current agricultural food production under such conditions remains an enormous and colossal challenge. Researchers have been trying to identify nutrient use-efficient varieties over the past few decades with limited success. The concept of nutrient use efficiency is being revisited to understand the molecular genetic basis, while much of it is not entirely understood yet. However, significant achievements have recently been observed at the molecular level in nitrogen and phosphorus use efficiency. Breeding teams are trying to incorporate these valuable QTLs and genes into their rice breeding programs. In this review, we seek to identify the achievements and the progress made so far in the fields of genetics, molecular breeding and biotechnology, especially for nutrient use efficiency in rice.
Future warming increases probability of globally synchronized maize production shocks
Meeting the global food demand of roughly 10 billion people by the middle of the 21st century will become increasingly challenging as the Earth’s climate continues to warm. Earlier studies suggest that once the optimum growing temperature is exceeded, mean crop yields decline and the variability of yield increases even if interannual climate variability remains unchanged. Here, we use global datasets of maize production and climate variability combined with future temperature projections to quantify how yield variability will change in the world’s major maize-producing and -exporting countries under 2 °C and 4 °C of global warming. We find that as the global mean temperature increases, absent changes in temperature variability or breeding gains in heat tolerance, the coefficient of variation (CV) of maize yields increases almost everywhere to values much larger than present-day values. This higher CV is due both to an increase in the SD of yields and a decrease in mean yields. For the top four maize-exporting countries, which account for 87% of global maize exports, the probability that they have simultaneous production losses greater than 10% in any given year is presently virtually zero, but it increases to 7% under 2 °C warming and 86% under 4 °C warming. Our results portend rising instability in global grain trade and international grain prices, affecting especially the ∼800 million people living in extreme poverty who are most vulnerable to food price spikes. They also underscore the urgency of investments in breeding for heat tolerance.
Effects of high temperature stress during anthesis and grain filling periods on photosynthesis, lipids and grain yield in wheat
Background Short episodes of high temperature (HT) stress during reproductive stages of development cause significant yield losses in wheat ( Triticum aestivum L.). Two independent experiments were conducted to quantify the effects of HT during anthesis and grain filling periods on photosynthesis, leaf lipidome, and yield traits in wheat. In experiment I, wheat genotype Seri82 was exposed to optimum temperature (OT; 22/14 °C; day/night) or HT (32/22 °C) for 14 d during anthesis stage. In experiment II, the plants were exposed to OT or HT for 14 d during the grain filling stage. During the HT stress, chlorophyll index, thylakoid membrane damage, stomatal conductance, photosynthetic rate and leaf lipid composition were measured. At maturity, grain yield and its components were quantified. Results HT stress during anthesis or grain filling stage decreased photosynthetic rate (17 and 25%, respectively) and grain yield plant − 1 (29 and 44%, respectively), and increased thylakoid membrane damage (61 and 68%, respectively) compared to their respective control (OT). HT stress during anthesis or grain filling stage increased the molar percentage of less unsaturated lipid species [36:5- monogalactosyldiacylglycerol (MGDG) and digalactosyldiacylglycerol (DGDG)]. However, at grain filling stage, HT stress decreased the molar percentage of more unsaturated lipid species (36:6- MGDG and DGDG). There was a significant positive relationship between photosynthetic rate and grain yield plant − 1 , and a negative relationship between thylakoid membrane damage and photosynthetic rate. Conclusions The study suggests that maintaining thylakoid membrane stability, and seed-set per cent and individual grain weight under HT stress can improve the photosynthetic rate and grain yield, respectively.
Multiple wheat genomes reveal global variation in modern breeding
Advances in genomics have expedited the improvement of several agriculturally important crops but similar efforts in wheat ( Triticum spp.) have been more challenging. This is largely owing to the size and complexity of the wheat genome 1 , and the lack of genome-assembly data for multiple wheat lines 2 , 3 . Here we generated ten chromosome pseudomolecule and five scaffold assemblies of hexaploid wheat to explore the genomic diversity among wheat lines from global breeding programs. Comparative analysis revealed extensive structural rearrangements, introgressions from wild relatives and differences in gene content resulting from complex breeding histories aimed at improving adaptation to diverse environments, grain yield and quality, and resistance to stresses 4 , 5 . We provide examples outlining the utility of these genomes, including a detailed multi-genome-derived nucleotide-binding leucine-rich repeat protein repertoire involved in disease resistance and the characterization of Sm1 6 , a gene associated with insect resistance. These genome assemblies will provide a basis for functional gene discovery and breeding to deliver the next generation of modern wheat cultivars. Comparison of multiple genome assemblies from wheat reveals extensive diversity that results from the complex breeding history of wheat and provides a basis for further potential improvements to this important food crop.
AMT1;1 transgenic rice plants with enhanced NH4+ permeability show superior growth and higher yield under optimal and suboptimal NH4+ conditions
The major source of nitrogen for rice (Oryza sativa L.) is ammonium (NH4(+)). The NH4(+) uptake of roots is mainly governed by membrane transporters, with OsAMT1;1 being a prominent member of the OsAMT1 gene family that is known to be involved in NH4(+) transport in rice plants. However, little is known about its involvement in NH4(+) uptake in rice roots and subsequent effects on NH4(+) assimilation. This study shows that OsAMT1;1 is a constitutively expressed, nitrogen-responsive gene, and its protein product is localized in the plasma membrane. Its expression level is under the control of circadian rhythm. Transgenic rice lines (L-2 and L-3) overexpressing the OsAMT1;1 gene had the same root structure as the wild type (WT). However, they had 2-fold greater NH4(+) permeability than the WT, whereas OsAMT1;1 gene expression was 20-fold higher than in the WT. Analogous to the expression, transgenic lines had a higher NH4(+) content in the shoots and roots than the WT. Direct NH4(+) fluxes in the xylem showed that the transgenic lines had significantly greater uptake rates than the WT. Higher NH4(+) contents also promoted higher expression levels of genes in the nitrogen assimilation pathway, resulting in greater nitrogen assimilates, chlorophyll, starch, sugars, and grain yield in transgenic lines than in the WT under suboptimal and optimal nitrogen conditions. OsAMT1;1 also enhanced overall plant growth, especially under suboptimal NH4(+) levels. These results suggest that OsAMT1;1 has the potential for improving nitrogen use efficiency, plant growth, and grain yield under both suboptimal and optimal nitrogen fertilizer conditions.
The PLATZ Transcription Factor GL6 Affects Grain Length and Number in Rice
Grain size is one of the key determinants of grain yield. Although a number of genes that control grain size in rice (Oryza sativa) have been identified, the overall regulatory networks behind this process remain poorly understood. Here, we report the map-based cloning and functional characterization of the quantitative trait locus GL6, which encodes a plant-specific plant AT-rich sequence- and zinc-binding transcription factor that regulates rice grain length and spikelet number. GL6 positively controls grain length by promoting cell proliferation in young panicles and grains. The null gl6 mutant possesses short grains, whereas overexpression of GL6 results in large grains and decreased grain number per panicle. We demonstrate that GL6 participates in RNA polymerase III transcription machinery by interacting with RNA polymerase III subunit C53 and transcription factor class C1 to regulate the expression of genes involved in rice grain development. Our findings reveal a further player involved in the regulation of rice grain size that may be exploited in future rice breeding.
