Combinations of Independent Dominant Loci Conferring Clubroot Resistance in All Four Turnip Accessions (Brassica rapa) From the European Clubroot Differential Set

Clubroot disease is devastating to
crop production when susceptible cultivars are planted in infected fields. European turnips are the most resistant sources and their resistance genes have been introduced into other crops such oilseed rape (
L.), Chinese cabbage and other
vegetables. The European clubroot differential (ECD) set contains four turnip accessions (ECD1-4). These ECD turnips exhibited high levels of resistance to clubroot when they were tested under controlled environmental conditions with Canadian field isolates. Gene mapping of the clubroot resistance genes in ECD1-4 were performed and three independent dominant resistance loci were identified. Two resistance loci were mapped on chromosome A03 and the third on chromosome A08. Each ECD turnip accession contained two of these three resistance loci. Some resistance loci were homozygous in ECD accessions while others showed heterozygosity based on the segregation of clubroot resistance in 20 BC
families derived from ECD1 to 4. Molecular markers were developed linked to each clubroot resistance loci for the resistance gene introgression in different germplasm.

Multiomics analysis of a resistant European turnip ECD04 during clubroot infection reveals key hub genes underlying resistance mechanism

The clubroot disease has become a worldwide threat for crucifer crop production, due to its soil-borne nature and difficulty to eradicate completely from contaminated field. In this study we used an elite resistant European fodder turnip ECD04 and investigated its resistance mechanism using transcriptome, sRNA-seq, degradome and gene editing. A total of 1751 DEGs were identified from three time points after infection, among which 7 hub genes including XTH23 for cell wall assembly and two CPK28 genes in PTI pathways. On microRNA, we identified 17 DEMs and predicted 15 miRNA-target pairs (DEM-DEG). We validated two pairs (miR395- APS4 and miR160- ARF ) by degradome sequencing. We investigated the miR395- APS4 pair by CRISPR-Cas9 mediated gene editing, the result showed that knocking-out APS4 could lead to elevated clubroot resistance in B. napus . In summary, the data acquired on transcriptional response and microRNA as well as target genes provide future direction especially gene candidates for genetic improvement of clubroot resistance on Brassica species.

Residue analysis and risk assessment of pyrethrins in open field and greenhouse turnips

A sensitive and selective method was developed and validated for the determination of pyrethrin residues in turnips (turnip leaves, turnip tubers, and the whole of plant) and cultivated soil using gas chromatography coupled with mass spectrometry (GC-MS). Six major components of pyrethrins (pyrethrin I and II, cinerin I and II, and jasmolin I and II) were separated and identified. The method involving solid-phase extraction (SPE) cleanup led to satisfactory average recoveries (88.1–104%) with limits of quantification (LOQs) of 0.05 mg/kg. The dissipation and final residue of pyrethrins in six provinces (among these places, two experiments were conducted in greenhouse and other four experiments in open filed) in China were studied. The trial results suggested that the half-lives of pyrethrins in the whole of turnips and soil were 0.5–1.6 and 1.0–1.3 days, respectively, and the degradation of pyrethrins in the greenhouse was quicker than that in open fields. The final residues of pyrethrins in turnip leaves and tubers were all below the maximum residue limit (MRL) established by the EU (1.0 mg/kg). A pre-harvest interval of 2 days and MRL of 1.0 mg/kg are recommended to ensure food safety standards for pyrethrins in turnips. Long-term risk assessment and short-term risk assessment of turnip tubers were evaluated. Hazard quotient (HQ) and acute hazard index (aHI) were significantly less than 100%, indicating negligible risk for consumption of turnip tubers.

Efficiency and Persistence of Movento® Treatment against Myzus persicae and the Transmission of Aphid-Borne Viruses

Neonicotinoids are widely used to protect fields against aphid-borne viral diseases. The recent ban of these chemical compounds in the European Union has strongly impacted rapeseed and sugar beet growing practices. The poor sustainability of other insecticide families and the low efficiency of prophylactic methods to control aphid populations and pathogen introduction strengthen the need to characterize the efficiency of new plant protection products targeting aphids. In this study, the impact of Movento® (Bayer S.A.S., Leverkusen, Germany), a tetrameric acid derivative of spirotetramat, on Myzus persicae and on viral transmission was analyzed under different growing temperatures. The results show (i) the high efficiency of Movento® to protect rapeseed and sugar beet plants against the establishment of aphid colonies, (ii) the impact of temperature on the persistence of the Movento® aphicid properties and (iii) a decrease of approximately 10% of the viral transmission on treated plants. These observations suggest a beneficial effect of Movento® on the sanitary quality of treated crops by directly reducing primary infections and indirectly altering, through aphid mortality, secondary infections on which the spread of disease within field depends. These data constitute important elements for the future development of management strategies to protect crops against aphid-transmitted viruses.
