A SARS-CoV-2 protein interaction map reveals targets for drug repurposing
A newly described coronavirus named severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), which is the causative agent of coronavirus disease 2019 (COVID-19), has infected over 2.3 million people, led to the death of more than 160,000 individuals and caused worldwide social and economic disruption1,2. There are no antiviral drugs with proven clinical efficacy for the treatment of COVID-19, nor are there any vaccines that prevent infection with SARS-CoV-2, and efforts to develop drugs and vaccines are hampered by the limited knowledge of the molecular details of how SARS-CoV-2 infects cells. Here we cloned, tagged and expressed 26 of the 29 SARS-CoV-2 proteins in human cells and identified the human proteins that physically associated with each of the SARS-CoV-2 proteins using affinity-purification mass spectrometry, identifying 332 high-confidence protein–protein interactions between SARS-CoV-2 and human proteins. Among these, we identify 66 druggable human proteins or host factors targeted by 69 compounds (of which, 29 drugs are approved by the US Food and Drug Administration, 12 are in clinical trials and 28 are preclinical compounds). We screened a subset of these in multiple viral assays and found two sets of pharmacological agents that displayed antiviral activity: inhibitors of mRNA translation and predicted regulators of the sigma-1 and sigma-2 receptors. Further studies of these host-factor-targeting agents, including their combination with drugs that directly target viral enzymes, could lead to a therapeutic regimen to treat COVID-19.
Influence of COVID-19 on lifestyle behaviors in the Middle East and North Africa Region: a survey of 5896 individuals
Background Coronavirus disease (COVID-19) pandemic has affected health and lifestyle behaviors of people globally. This project aims to identify the impact of COVID-19 on lifestyle behavior of individuals in the Middle East and North Africa (MENA) region during confinement. Methods We conducted an online survey in 17 countries (Egypt, Jordan, United Arab Emirates, Kuwait, Bahrain, Saudi Arabia, Oman, Qatar, Yemen, Syria, Palestine, Algeria, Morocco, Libya, Tunisia, Iraq, and Sudan) from the MENA region on August and September 2020. The questionnaire included self-reported information on lifestyle behaviors, including physical activity, eating habits, smoking, watching television, social media use and sleep before and during the pandemic. Logistic regression was performed to analyze the impact of COVID-19 on lifestyle behaviors. Results A total of 5896 participants were included in the final analysis and 62.8% were females. The BMI of the participants was 25.4 ± 5.8 kg/m 2 . Around 38.4% of the participants stopped practicing any physical activities during the confinement (P < 0.001), and 57.1% reported spending more than 2 h on social media (P < 0.001). There were no significant changes in smoking habits. Also, 30.9% reported an improvement in their eating habits compared with 24.8% reported worsening of their eating habits. Fast-food consumption decreased significantly in 48.8% of the study population. This direct/indirect exposure to COVID-19 was associated with an increased consumption of carbohydrates (OR = 1.09; 95% CI = 1.02–1.17; P = 0.01), egg (OR = 1.08; 95% CI = 1.02–1.16; P = 0.01), sugar (OR = 1.09; 95% CI = 1.02–1.16; P = 0.02), meat, and poultry (OR = 1.13; 95% CI = 1.06–1.20; P < 0.01). There was also associated increase in hours spent on watching television (OR = 1.07; 95% CI = 1.02–1.12; P < 0.01) and social media (OR = 1.09; 95% CI = 1.01–1.18; P = 0.03). However, our results showed a reduction in sleeping hours among those exposed to COVID-19 infection (OR = 0.85; 95% CI = 0.77–0.94; P < 0.01). Conclusions The COVID-19 pandemic was associated with an increase in food consumption and sedentary life. Being exposed to COVID-19 by direct infection or through an infected household is a significant predictor of amplifying these changes. Public health interventions are needed to address healthy lifestyle behaviors during and after the COVID-19 pandemic.
Health, social, and economic characteristics of patients enrolled in a COVID-19 recovery program
At least one in five people who recovered from acute COVID-19 have persistent clinical symptoms, however little is known about the impact on quality-of-life (QOL), socio-economic characteristics, fatigue, work and productivity. We present a cross-sectional descriptive characterization of the clinical symptoms, QOL, socioeconomic characteristics, fatigue, work and productivity of a cohort of patients enrolled in the MedStar COVID Recovery Program (MSCRP). Our participants include people with mental and physical symptoms following recovery from acute COVID-19 and enrolled in MSCRP, which is designed to provide comprehensive multidisciplinary care and aid in recovery. Participants completed medical questionnaires and the PROMIS-29, Fatigue Severity Scale, Work and Productivity Impairment Questionnaire, and Social Determinants of Health surveys. Participants (n = 267, mean age 47.6 years, 23.2% hospitalized for COVID-19) showed impaired QOL across all domains assessed with greatest impairment in physical functioning (mean 39.1 ± 7.4) and fatigue (mean 60.6 ±. 9.7). Housing or “the basics” were not afforded by 19% and food insecurity was reported in 14% of the cohort. Participants reported elevated fatigue (mean 4.7 ± 1.1) and impairment with activity, work productivity, and on the job effectiveness was reported in 63%, 61%, and 56% of participants, respectively. Patients with persistent mental and physical symptoms following initial illness report impairment in QOL, socioeconomic hardships, increased fatigue and decreased work and productivity. Our cohort highlights that even those who are not hospitalized and recover from less severe COVID-19 can have long-term impairment, therefore designing, implementing, and scaling programs to focus on mitigating impairment and restoring function are greatly needed.
Dying of starvation if not from bombs: assessing measurement properties of the Food Insecurity Experiences Scale (FIES) in Gaza’s civilian population experiencing the world’s worst hunger crisis
Background Since October 2023, the humanitarian crisis in Gaza has dramatically escalated and food insecurity (FI) has rapidly deteriorated. In the face of such situation, it has been recommended that accurate information on FI should be integrated in emergency reporting systems in order to better target and assess the impact of humanitarian aid. To achieve this, field practitioners, researchers and decision-makers should be equipped with user-friendly and psychometrically sound measures tailored to real-life war situations and the Palestinian context. Therefore, the present study aimed to test the psychometric properties of the Food Insecurity Experiences Scale (FIES) in a sample of Arabic-speaking community adults currently living in the Gaza Strip during the war. Methods This study used a cross-sectional design, snowball sampling as a recruiting strategy, and a self-administered online questionnaire to collect data from participants. It was performed during the period from September 1st to 30th, 2024. A total of 534 valid responses were received and were included in the final analysis. Results Approximately one out of two people in Gaza were experiencing any FI. Fit indices in confirmatory factor analysis indicated that a one-factor solution fit the data with acceptable factor loadings. Furthermore, our findings indicated that the Arabic version of the FIES presented an excellent level of reliability, with both Cronbach’s alpha and McDonald omega coefficients of 0.94. Cross-sex measurement invariance in the FIES was established at scalar and metric levels. Finaly, concurrent validity was evidenced by FIES’s positive correlations with water insecurity experiences, post-traumatic stress, depression and anxiety symptoms. Conclusion Altogether, our findings showed that the FIES fulfils requirements of validity and reliability. It may thus be considered as appropriate tool for assessing FI in Gazan war-affected populations currently suffering from acute FI emergency and requiring urgent action to meet their food needs. The FIES is simple, short, economic and time-effective. As a FI indicator, the Arabic FIES is useful in providing information that enables actions by policy makers and guides humanitarian-aid actors’ efforts aimed at decreasing, mitigating, or preventing severe FI, and saving lives or livelihoods. Adaptation to a changing climate in the Arab Countries
Adapting to climate change is not a new phenomenon for the Arab world. For thousands of years, the people in Arab countries have coped with the challenges of climate variability by adapting their survival strategies to changes in rainfall and temperature. Their experience has contributed significantly to the global knowledge on climate change and adaptation. But over the next century global climatic variability is predicted to increase, and Arab countries may well experience unprecedented extremes in climate. Temperatures may reach new highs, and in most places there may be a risk of less rainfall. Under these circumstances, Arab countries and their citizens will once again need to draw on their long experience of adapting to the environment to address the new challenges posed by climate change. This report prepared through a consultative process with Government and other stakeholders in the Arab world assesses the potential effects of climate change on the Arab region and outlines possible approaches and measures to prepare for its consequences. It offers ideas and suggestions for Arab policy makers as to what mitigating actions may be needed in rural and urban settings to safeguard key areas such as health, water, agriculture, and tourism. The report also analyzes the differing impacts of climate change, with special attention paid to gender, as a means of tailoring strategies to address specific vulnerabilities. The socioeconomic impact of climate change will likely vary from country to country, reflecting a country's coping capacity and its level of development. Countries that are wealthier and more economically diverse are generally expected to be more resilient. The report suggests that countries and households will need to diversify their production and income generation, integrate adaptation into all policy making and activities, and ensure a sustained national commitment to address the social, economic, and environmental consequences of climate variability. With these coordinated efforts, the Arab world can, as it has for centuries, successfully adapt and adjust to the challenges of a changing climate.
The Food Question in the Middle East
In recent years, the food question has been a central concern for politicians, economists, international organizations, activists and NGOs alike, as well as social scientists at large. This interest has emerged from the global food crisis and its impact on the environment and the political economy and security of the global south, as well as the expansion of scholarly studies relating food issues to agrarian questions with the objective of developing theoretical frameworks that would allow for a critical analysis of the current food issues at historical, cultural, social, political and economic levels. In this context, Cairo Papers organized its 2016 symposium around the food question in the Middle East. Papers in this collection address the food question from both its food and agricultural aspects, and approach it as the site of political and economic conflicts, as the means of sociocultural control and distinction, and as the expression of national and ethnic identities. Climate change impacts in the Middle East and Northern Africa (MENA) region and their implications for vulnerable population groups
The Middle East and North Africa (MENA) region emerges as one of the hot spots for worsening extreme heat, drought and aridity conditions under climate change. A synthesis of peer-reviewed literature from 2010 to date and own modeling work on biophysical impacts of climate change on selected sectors shows that the region is highly affected by present and future climate change. These biophysical impacts paired with other pressures and a lack of resilience in some countries cause high vulnerabilities within these sectors and for social dimensions in the MENA region. The agricultural sector, of which 70 percent is rain-fed, is highly exposed to changing climatic conditions. This is of critical importance as the agriculture sector is the largest employer in many Arab countries and contributes significantly to national economies. Impacts will be high in a 2 °C world, as, e.g., annual water discharge, already critically low, is projected to drop by another 15–45% (75% in a 4 °C world) and unusual heat extremes projected to affect about one-third of the land area with likely consequences for local food production. As a consequence, deteriorating rural livelihoods associated with declining agricultural productivity will continue to contribute to migration flows, often to urban areas as already observed. The region could be heavily challenged by both rising food and water demand given its projected increase in population that may double by 2070. As a result, the regions already substantial import dependency could increase and thus its vulnerability to agricultural impacts well beyond its country borders. A severe and sustained pressure on resources could contribute to further social unrest in the already unstable political environment that currently characterizes parts of the region. While the particular societal responses to such changes are hard to foresee, it is clear that extreme impacts would constitute unprecedented challenges to the social systems affected.
