The Utility of Proximity-Based Herbicide Exposure Assessment in Epidemiologic Studies of Vietnam Veterans
A fundamental challenge in past studies evaluating whether health problems experienced by Vietnam veterans might be linked to wartime use of Agent Orange or other herbicides has been a lack of information about the veterans' level of exposure to these herbicides.

Veterans and Agent Orange
In 2001, in response to a request by the U.S. Department of Veterans Affairs (DVA), the Institute of Medicine (IOM) called together a committee to conduct a review of the scientific evidence regarding the association between exposure to dioxin and other chemical compounds in herbicides used in Vietnam and acute myelogenous leukemia in the offspring of Vietnam veterans. Based on the scientific evidence reviewed in this report, the committee finds there is inadequate or insufficient evidence to determine if an association exists between exposure to the herbicides used in Vietnam or their contaminants and acute myelogenous leukemia (AML) in the children of Vietnam veterans. This is a change in classification from the recent Veterans and Agent Orange: Update 2000 report, which found limited/suggestive evidence for such an association.

Easy to use, effective, and safe: who wouldn't love synthetic pesticides? Apparently most Americans did--and apparently still do. Why--in the face of dire warnings, rising expense, and declining effectiveness--do we cling to our chemicals? Michelle Mart wondered. Her book, a cultural history of pesticide use in postwar America, offers an answer. America's embrace of synthetic pesticides began when they burst on the scene during World War II and has held steady into the 21st century--for example, more than 90% of soybeans grown in the US in 2008 are Roundup Ready GMOs, dependent upon generous use of the herbicide glyphosate to control weeds. Mart investigates the attraction of pesticides, with their up-to-the-minute promise of modernity, sophisticated technology, and increased productivity--in short, their appeal to human dreams of controlling nature. She also considers how they reinforced Cold War assumptions of Western economic and material superiority. Though the publication of Rachel Carson's Silent Spring and the rise of environmentalism might have marked a turning point in Americans' faith in pesticides, statistics tell a different story. Pesticides, a Love Story recounts the campaign against DDT that famously ensued; but the book also shows where our notions of Silent Spring's revolutionary impact falter--where, in spite of a ban on DDT, farm use of pesticides in the United States more than doubled in the thirty years after the book was published. As a cultural survey of popular and political attitudes toward pesticides, Pesticides, a Love Story tries to make sense of this seeming paradox. At heart, it is an exploration of the story we tell ourselves about the costs and benefits of pesticides--and how corporations, government officials, ordinary citizens, and the press shape that story to reflect our ideals, interests, and emotions\"--
The evidence of human exposure to glyphosate: a review
Background Despite the growing and widespread use of glyphosate, a broad-spectrum herbicide and desiccant, very few studies have evaluated the extent and amount of human exposure. Objective We review documented levels of human exposure among workers in occupational settings and the general population. Methods We conducted a review of scientific publications on glyphosate levels in humans; 19 studies were identified, of which five investigated occupational exposure to glyphosate, 11 documented the exposure in general populations, and three reported on both. Results Eight studies reported urinary levels in 423 occupationally and para-occupationally exposed subjects; 14 studies reported glyphosate levels in various biofluids on 3298 subjects from the general population. Average urinary levels in occupationally exposed subjects varied from 0.26 to 73.5 μg/L; environmental exposure urinary levels ranged from 0.16 to 7.6 μg/L. Only two studies measured temporal trends in exposure, both of which show increasing proportions of individuals with detectable levels of glyphosate in their urine over time. Conclusions The current review highlights the paucity of data on glyphosate levels among individuals exposed occupationally, para-occupationally, or environmentally to the herbicide. As such, it is challenging to fully understand the extent of exposure overall and in vulnerable populations such as children. We recommend further work to evaluate exposure across populations and geographic regions, apportion the exposure sources (e.g., occupational, household use, food residues), and understand temporal trends.
Enhancing tolerance of wheat cultivars to metribuzin stress through bioremediation with Pseudomonas spp
Pseudomonas spp. are bacteria known for their ability to degrade various pesticides, acting as natural biofilters. Despite their widespread use, the degradation of metribuzin—a herbicide commonly applied in agriculture—by these bacterial strains remains undocumented. This investigation aims to assess the capacity of Pseudomonas spp. to alleviate metribuzin-induced stress in different wheat cultivars under precisely controlled environmental conditions, offering insights into potential bioremediation strategies. This experiment was conducted as factorial in a completely randomized design with three replications. It included varying concentrations of metribuzin (0, 5, 10, 15, 20, and 30 mg/kg), bacterial treatments (control, P. fluorescens , and P. aeruginosa ), and wheat cultivars (Pishtaz, Chamran, and Sivand). The evaluation focused on plant growth and biochemical responses, monitored at ten-day intervals starting 14 days after planting (DAP). A strong dose-dependent inhibition of plant growth was observed with increasing metribuzin concentrations. Growth reduction intensified at concentrations above 20 mg/kg, but near-complete inhibition (EC90) generally required higher concentrations, depending on cultivar and bacterial treatment. Pishtaz demonstrated the highest level of herbicide tolerance, with EC90 values in the non-inoculated treatment ranging from 23 to 107 mg/kg across growth stages. Increases in antioxidant enzyme activities and decreases in malondialdehyde levels were observed, indicating the stress mitigation capabilities of Pseudomonas spp. through biodegradation, which also led to enhanced plant growth and increased bacterial colony numbers in the soil. Variability in metribuzin sensitivity was observed among the cultivars, with Pishtaz being notably tolerant, Chamran semi-sensitive, and Sivand sensitive. P. fluorescens , in particular, showed a marked ability to reduce metribuzin’s detrimental effects, underscoring the potential of employing microbial strategies for managing pesticide stress in wheat cultivation.
Urinary Biomarkers of Prenatal Atrazine Exposure and Adverse Birth Outcomes in the PELAGIE Birth Cohort
Background: Despite evidence of atrazine toxicity in developing organisms from experimental studies, few studies—and fewer epidemiologic investigations—have examined the potential effects of prenatal exposure. Objectives: We assessed the association between adverse birth outcomes and urinary biomarkers of prenatal atrazine exposure, while taking into account exposures to other herbicides used on corn crops (simazine, alachlor, metolachlor, and acetochlor). Methods: This study used a case-cohort design nested in a prospective birth cohort conducted in the Brittany region of France from 2002 through 2006. We collected maternal urine samples to examine pesticide exposure biomarkers before the 19th week of gestation. Results: We found quantifiable levels of atrazine or atrazine mercapturate in urine samples from 5.5% of 579 pregnant women, and dealkylated and identified hydroxylated triazine metabolites in 20% and 40% of samples, respectively. The presence versus absence of quantifiable levels of atrazine or a specific atrazine metabolite was associated with fetal growth restriction [odds ratio (OR) = 1.5; 95% confidence interval (CI), 1.0-2.2] and small head circumference for sex and gestational age (OR = 1.7; 95% CI, 1.0-2.7). Associations with major congenital anomalies were not evident with atrazine or its specific metabolites. Head circumference was inversely associated with the presence of quantifiable urinary metolachlor. Conclusions: Ulis study is the first to assess associations of birth outcomes with multiple urinary biomarkers of exposure to triazine and chloroacetanilide herbicides. Evidence of associations with adverse birth outcomes raises particular concerns for countries where atrazine is still in use.
Rotenone, Paraquat, and Parkinson's Disease
Background: Mitochondrial dysfunction and oxidative stress are pathophysiologic mechanisms implicated in experimental models and genetic forms of Parkinson's disease (PD). Certain pesticides may affect these mechanisms, but no pesticide has been definitively associated with PD in humans. Objectives: Our goal was to determine whether pesticides that cause mitochondrial dysfunction or oxidative stress are associated with PD or clinical features of parkinsonism in humans. Methods: We assessed lifetime use of pesticides selected by mechanism in a case–control study nested in the Agricultural Health Study (AHS). PD was diagnosed by movement disorders specialists. Controls were a stratified random sample of all AHS participants frequency-matched to cases by age, sex, and state at approximately three controlsrone case. Results: In 110 PD cases and 358 controls, PD was associated with use of a group of pesticides that inhibit mitochondrial complex I [odds ratio (OR) = 1.7; 95% confidence interval (CI), 1.0-2.8] including rotenone (OR = 2.5; 95% CI, 1.3-4.7) and with use of a group of pesticides that cause oxidative stress (OR = 2.0; 95% CI, 1.2-3.6), including paraquat (OR = 2.5; 95% CI, 1.4-4.7). Conclusions: PD was positively associated with two groups of pesticides defined by mechanisms implicated experimentally—those that impair mitochondrial function and those that increase oxidative stress—supporting a role for these mechanisms in PD pathophysiology.
Parallel and nonparallel genomic responses contribute to herbicide resistance in Ipomoea purpurea, a common agricultural weed
The repeated evolution of herbicide resistance has been cited as an example of genetic parallelism, wherein separate species or genetic lineages utilize the same genetic solution in response to selection. However, most studies that investigate the genetic basis of herbicide resistance examine the potential for changes in the protein targeted by the herbicide rather than considering genome-wide changes. We used a population genomics screen and targeted exome re-sequencing to uncover the potential genetic basis of glyphosate resistance in the common morning glory, Ipomoea purpurea, and to determine if genetic parallelism underlies the repeated evolution of resistance across replicate resistant populations. We found no evidence for changes in 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS), glyphosate's target protein, that were associated with resistance, and instead identified five genomic regions that showed evidence of selection. Within these regions, genes involved in herbicide detoxification-cytochrome P450s, ABC transporters, and glycosyltransferases-are enriched and exhibit signs of selective sweeps. One region under selection shows parallel changes across all assayed resistant populations whereas other regions exhibit signs of divergence. Thus, while it appears that the physiological mechanism of resistance in this species is likely the same among resistant populations, we find patterns of both similar and divergent selection across separate resistant populations at particular loci.
