BACKGROUND: We report the discovery of a nematode parasite egg (Nemata: Oxyurida) from a coprolite closely associated with the remains of several species of Cynodontia, dated to 240 million years old. This finding is particularly significant because this is the oldest record of an oxyurid nematode yet discovered, and because the cynodonts are considered a stem-group of the mammals. METHODS: We extracted material from a fully mineralized coprolite by both scraping the surface, and removing fragments from its interior with clean dental instruments used a single time. A single drop of glycerol from a new vial was added as a clearing reagent. Each slide was sealed with wax and examined with an optical microscope at 100× to 400× magnification. RESULTS: From one coprolite, 550 slides were examined; from 275 of these slides, sediment was examined that was scraped from the surface of the coprolite, and from the other 275 slides, material was examined that was extracted from the interior of the coprolite. All microscopic structures encountered were photographed, measured, and identified when possible. CONCLUSIONS: From the coprolite examined, we discovered an egg representing a new species of pinworm that, based on the egg structure, clearly places it in the family Heteroxynematidae. Nematodes of the order Oxyurida have very constrained life-histories, occurring only in animals that are not strictly carnivorous and also ingest large amounts of plant material. This fact enabled us to determine which species of cynodont, from several collected at the site in Brazil, are most likely the depositors of the coprolite, and therefore were the putative host of the parasite.

is a genus of oxyurid nematodes which parasitize both omnivorous and herbivorous hosts.
Thelandros chalcidiae
sp. nov. is a new taxa described from the large intestine of the ocellated skink,
Chalcides ocellatus
, from the South Sinai Desert in Egypt. The recovered parasite species was examined at both morphological and molecular levels in order to determine the exact taxonomic position within Pharyngodonidae family. The current pharyngodonid species is characterized by a mouth opening bounded by three bilobed lips; male worms are characterized by the presence of two pairs of cloacal papillae (pre- and adcloacal) and one single caudal papillae (postcloacal) and caudal alae in an auricular form and females with post-equatorial vulva, amphidelphic ovary in which anterior ovary extended to level of excretory pore and posterior ovary extended posteriorly up anal opening. The recovered nematodes were compared with other known species from different hosts and it was found to be morphologically different from them. Molecular characterization based on the partial 28S rRNA nuclear ribosomal gene sequence showed sequence identities ≥ 83.15% with taxa under family Pharyngodonidae, 74.84–87.37% with Oxyuridae, 80.54% with Heteroxynematidae, and 75.98–77.72% with Thelastomatidea. Phylogenetic analysis showed that parasite sequence in conjunction with existing data facilitate placement of this species within Oxyurida. The present species is deeply embedded in genus
Thelandros
with close relationships to previously described
Thelandros
sp. and
T. galloti
in same taxon. This study highlighted importance of combining morphological and genetic data with taxonomy in pharyngodonid species.

Why Museums Matter: A Tale of Pinworms (Oxyuroidea: Heteroxynematidae) Among Pikas (Ochotona princeps and O. collaris) in the American West

Permanent and well-supported museum or natural history collections provide a solid foundation for the process of systematics research through creation of an empirical record which validates our understanding of the biosphere. We explore the role of museums in ongoing studies of the complex helminth fauna characteristic of pikas (Ochotona spp.) in the American west. These studies address the taxonomy for pinworms of the Labiostomatinae and the problems associated with the absence of adequate type series and vouchers and with misidentifications in original descriptions. We demonstrate that the types for Labiostomum (Labiostomum) coloradensis are identical to some specimens in the syntype series representing L. (Eugenuris) utahensis, although the published descriptions are in disagreement. Both are identical to L. (Eugenuris) talkeetnaeuris and, as a consequence, are reduced as junior synonyms. Only 2 species of large pinworms, namely L. (Labiostomum) rauschi and L. (Eugenuris) talkeetnaeuris, are widely distributed in Ochotona collaris and O. princeps. Although this serves to clarify the taxonomy for species in these genera, prior records remain confused, as representative voucher specimens from all major surveys in North America were never submitted to museum collections. We strongly suggest that type and voucher series should not be held in private or personal collections, where such are eventually lost, discarded, or destroyed through neglect due to inattention and the absence of curation. The potential to accumulate meaningful baselines for assessment of environmental change is jeopardized if materials from survey and inventory are not routinely submitted to museum collections. The capacity of museum repositories, as a focus for systematics, ecology, and evolutionary studies and for the development of resources for biodiversity informatics, continues to be undervalued and poorly utilized by a cadre of scientists who are dependant on accurate and definitive information that transcends specific disciplines.

Molecular phylogeny of the pinworms of mice, rats and rabbits, and its use to develop molecular beacon assays for the detection of pinworms in mice

Though pinworm infestation has been prevalent since the early years of laboratory animal medicine, the genomes of these parasites have not yet been sequenced. The authors used high-fidelity polymerase chain reaction to amplify a large portion of the ribosomal gene complex of four pinworm species commonly found in lab rodents and rabbits (
Aspiculuris tetraptera, Passalurus ambiguus, Syphacia muris
and
Syphacia obvelata
). They determined DNA sequences for these complexes and carried out phylogenetic analysis. Using this information, the authors developed real-time molecular beacon assays for pinworm detection, comparing the new diagnostic approach with traditional methods such as perianal tape testing, fecal flotation and direct examination of intestinal content.

. Gerbils in the wild are known as hosts of this parasite in Asia, pet gerbils — in the USA and Brazil. Our record is the first in Europe. Complete descriptions of male and female individuals are given.
