A New Isospora (Apicomplexa: Eimeriidae) from the Summer Tanager, Piranga rubra (Aves: Passeriformes: Cardinalidae), in Oklahoma
The summer tanager, Piranga rubra (L., 1758) is a medium-sized songbird formerly belonging to the tanager family Thraupidae but now has been placed within the family Cardinalidae. Nothing is known about the coccidian parasites of this stunningly colorful bird. Feces from 2 P. rubra found dead in McCurtain County, Oklahoma were collected and examined for coccidia; 1 was found to be passing a new species of Isospora. Oocysts of Isospora mccurtainensis n. sp. are subspheroidal to ovoidal with a smooth bilayered wall, measure (length 3 width [L 3 W]) 21.7 3 19.5 µm, and have a L/W ratio of 1.1; a micropyle and oocyst residuum were absent but a bilobed and refractile polar granule is present. Sporocysts are ellipsoidal and measure 13.9 3 8.6 µm, L/W 1.6; a knoblike Stieda body is present as well as a distinct sub-Stieda body. The sporocyst residuum is composed of a granular compact cluster with a dense, irregular mass of granules lying between and dispersed among the sporozoites. This is the first coccidian reported from P. rubra and, most important, only the first known from the Cardinalidae in the mainland of the United States.
A New Isosporan (Apicomplexa: Eimeriidae) from Wood Thrush, Hylocichla mustelina (Aves: Passeriformes: Turdidae), in Oklahoma
The wood thrush, Hylocichla mustelina, is a North American passerine bird closely related to other thrushes and is widely distributed across North America. Nothing is known of the coccidian parasites of this bird. Feces from a single H. mustelina found dead in McCurtain County, Oklahoma were collected and examined for coccidia; it was found to be passing a new species of Isospora. Oocysts of Isospora gmelini n. sp. are subspheroidal to ellipsoidal with a smooth bilayered wall, measure (length [L] × width [W]) 19.5 × 16.5 µm, and have a L/W ratio of 1.2; a micropyle and oocyst residuum were absent but polar granule(s) are present. Sporocysts are ellipsoidal to elongate and measure 13.4 × 8.9 µm, L/W 1.5; a buttonlike Stieda body is present, but sub-Stieda and para-Stieda bodies are absent. The sporocyst residuum is composed of a compact spheroid with a dense, irregular mass of finer granules lying between and dispersed among the sporozoites. Although several isosporans have been reported from other turdid birds, mainly from Brazil and Costa Rica, this is the initial coccidian reported from H. mustelina and only the second known from the Turdidae in the mainland of the United States.
Polymorphism and genetic diversity of Isospora parnaitatiaiensis Silva, Rodrigues, Lopes, Berto, Luz, Ferreira & Lopes, 2015 (Eimeriidae) from antbirds (Thamnophilidae) in Brazil
Isospora parnaitatiaiensis Silva, Rodrigues, Lopes, Berto, Luz, Ferreira & Lopes, 2015 was identified from a new host, the plain antvireo Dysithamnus mentalis (Temminck), and also from the white-shouldered fire-eye Pyriglena leucoptera Vieillot, in its type-locality, the Itatiaia National Park in the southeastern Brazil, and a preliminary genotypic characterisation by sequencing the mitochondrial cytochrome c oxidase subunit 1 gene is provided. The oöcysts recovered from P. leucoptera and D. mentalis were polymorphic and have genotypic differences that were not considered sufficient for the description of new species, but only different genotypes and morphotypes of I. parnaitatiaiensis related to each host. These morphological and molecular variations were associated with a process of ongoing speciation and in adaptive development to their respective host species.
Characterization and Distribution of Isospora sabiai N. SP. (Apicomplexa: Eimeriidae) from Thrushes Turdus spp. (Passeriformes: Turdidae) from Brazil
A new coccidian species (Protozoa: Apicomplexa: Isospora) is described parasitizing white-necked thrushes Turdus albicollis Vieillot, 1818; rufous-bellied thrushes Turdus rufiventris Vieillot, 1818; pale-breasted thrushes Turdus leucomelas Vieillot, 1818; and yellow-legged thrushes Turdus flavipes Vieillot, 1818 from 3 different localities in Brazil. Isospora sabiai n. sp. has oocysts that are subspherical to ellipsoidal, 20.9 × 18.6 μm, with smooth, delicate, bilayered wall, ∼1.1 μm thick. Micropyle inconspicuous or imperceptible. Oocyst residuum absent, but small polar granules rounded or comma-shaped are present. Sporocysts are elongate ellipsoidal to reniform, 16.5 × 9.2 μm. The Stieda body is knob-like. Sub-Stieda body rounded to conical, sometimes homogeneous with the Stieda body. Sporocyst residuum is present, usually as a cluster of numerous granules. Sporozoites are vermiform with 2 refractile bodies. The oocysts and sporocysts of I. sabiai n. sp. are uniform in the proportionality of width on length, but exhibited different patterns of size associated with each host species; therefore, an ecological discussion is introduced aimed at associating these morphometrical patterns of the oocysts with the habits of the different species of thrushes. This is the seventh isosporoid coccidian reported from New World turdids.
Coccidia of New World passerine birds (Aves: Passeriformes): a review of Eimeria Schneider, 1875 and Isospora Schneider, 1881 (Apicomplexa: Eimeriidae)
In the New World, the avian order Passeriformes comprises 47 families and 2,453 species, yet to date only 21 (45%) of the families and 58 (2%) of the species have been examined for coccidia, and from these only two species of Eimeria Schneider, 1875 and 81 species of Isospora Schneider, 1881 have been described. This review contributes to our understanding of the morphology and systematics of coccidian parasites of passeriforms, providing a scientific basis for the identification of sporulated oöcysts recovered from the faeces of passerine birds from North, Central and South America. To this end, the coccidia were organised and grouped according to the family of the host, following the widely recognised concept of family-specificity and the updated systematics of the class Aves. Details of 83 eimeriid species are presented along with an illustration and tabulated data.
Individual Oocysts of Isospora (Apicomplexa: Coccidia) Parasites From Avian Feces: From Photo to Sequence
Numerous microscopic studies of coccidian oocysts from avian feces have become the basis for species identification. In contrast, molecular studies of wild birds' Coccidia are still in their infancy and are mostly based on DNA extracted from the blood stages of these parasites. Linking microscopic and molecular data requires a method that reliably extracts DNA from single oocysts with parallel detailed morphological examination of the same cell. We offer a thorough manual of isolating, photographing, and trapping single oocysts from avian feces, followed by extraction of parasite DNA and amplification of mitochondrial DNA from the same cells. In 39 single oocysts from 6 wild blackcaps, we combined microscopic studies of individual cells with studies on their mitochondrial haplotype. In 72% of the single oocysts sampled, we detected unambiguous sequences. From feces and blood of investigated birds, we obtained 6 different haplotypes of Isospora sp. (iSAT1-iSAT 6), finding both the same haplotype in different host individuals and various haplotypes in the same host individual. Our described methodology enables linking the huge amount of morphological data with innovative gene analysis. This method expands the scope of genetic studies conducted on Isospora species, including routine molecular analysis of single oocysts isolated from fecal samples.
Some remarks on the distribution and dispersion of Coccidia from icterid birds in South America: Isospora guaxi n. sp. and Isospora bellicosa Upton, Stamper & Whitaker, 1995 (Apicomplexa: Eimeriidae) from the red-rumped cacique Cacicus haemorrhous (L.) (Passeriformes: Icteridae) in southeastern Brazil
A new species of coccidian, Isospora guaxi n. sp., and Isospora bellicosa Upton, Stamper & Whitaker, 1995 (Protozoa: Apicomplexa: Eimeriidae) are recorded from red-rumped caciques Cacicus haemorrhous (L.) in the Parque Nacional do Itatiaia, Brazil. Isospora guaxi n. sp. has sub-spheroidal oöcysts, measuring on average 30.9 × 29.0 μm, with smooth, bi-layered wall c. 1.9 μm thick. Micropyle and oöcyst residuum are absent, but a polar granule is present. Sporocysts are ellipsoidal, measuring on average 19.3 × 13.8 μm. Stieda body is knob-like and sub-Stieda body is prominent and compartmentalized. Sporocyst residuum is composed of scattered granules. Sporozoites are vermiform, with one refractile body and a nucleus. Isospora bellicosa has sub-spheroidal to ovoidal oöcysts, measuring on average 27.1 × 25.0 μm, with smooth, bi-layered wall c. 1.5 μm thick. Micropyle and oöcyst residuum are absent, but one or two polar granules are present. Sporocysts are ellipsoidal, measuring on average 18.1 × 10.9 μm. Stieda body is knob-like and sub-Stieda body is rounded to rectangular. Sporocyst residuum is composed of a cluster of compact or diffuse granules. Sporozoites are vermiform, with one refractile body and a nucleus. Isospora bellicosa was originally described from the Peruvian meadowlark Sturnella bellicosa deFilippi, a trans -Andean icterid which is allopatric with the cis- Andean C. haemorrhous . Therefore, in conclusion, this current study reveals the dispersion of coccidia from Icteridae across the Andes Mountains, besides describing the sixth isosporoid coccidium infecting an icterid bird.
Isospora toxostomai n. sp. (Apicomplexa: Eimeriidae) from the curved-billed thrasher Toxostoma curvirostre (Swainson) (Passeriformes: Mimidae) at the Central highlands of Mexico
Isospora toxostomai n. sp. (Apicomplexa: Eimeriidae) is described based on material from the curved-billed thrasher Toxostoma curvirostre (Swainson) in the Central Highlands of Mexico. The new species possesses subspherical oöcysts, with a smooth, bi-layered wall. Sporulated oöcysts measure 22-25 × 21-24 (23.4 × 22.3) µm; length/width (L/W) ratio of 1.0-1.1 (1.1). Sporocysts are ellipsoidal, 15-17 × 10-11 (15.8 × 10.5); L/W ratio of 1.3-1.6 (1.5). Micropyle and oöcyst residuum are both absent, and a polar granule present (many fibrils). Mean dimensions of both sporulated oöcysts and sporocysts of I. toxostomai n. sp. appear to be considerably larger than those of Isospora mimusi Coelho, Berto, Neves, Oliveira Flausino & Lopes, 2011 from the tropical mockingbird Mimus gilvus (Vieilot) in Brazil. This is the second species of Isospora Schneider, 1881 infecting a host of the Mimidae in the Americas.
Coprology of a single Northern white-breasted hedgehog (Erinaceus roumanicus): first report of Isospora rastegaievae in Poland
Coprological analysis of a sample from one free-living hedgehog was done with the use of a direct flotation method with additional incubation of fecal material. The study revealed three types of eggs and oocysts in the feces. The most commonly diagnosed were oocysts of Isospora rastegaievae (543/3g), while oocysts of Monocystis sp. (267/3g) and eggs of Aonchotheca/Eucoleus spp. (52/3g) were seen less often. This is the first report of coccidia I. rastegaievae (Apicomplexa: Eimeriida) and acephaline gregarine Monocystis sp. (Apicomplexa: Eugregarinida) infection in a hedgehog in Poland.
Isospora phainopepla n. sp. (Apicomplexa: Eimeriidae) from Phainopepla nitens (Swainson) (Passeriformes: Ptiliogonatidae) in the Joshua Tree National Park, USA
Isospora phainopepla n. sp. is described from the faeces of Phainopepla nitens (Swainson) collected in Joshua Tree National Park, California, USA. To our knowledge, there are no metazoan or protist parasites described yet from any species in the Ptiliogonatidae of the Americas. Sporulated oöcysts of the isosporan are ellipsoidal, 30-35 × 22-29 (32.9 × 25.4) μm, with a length/width (L/W) ratio 1.20-1.35 (1.30); one rice-shaped polar granule is present, but both oöcyst residuum and micropyle are absent. Sporocyts are elongate-ellipsoidal, 22-27 × 11-14 (23.9 × 12.0) μm, with L/W ratio 1.97-2.02 (2.00); prominent Stieda and sub-Stieda bodies are both present, but para-Stieda body is absent. Sporocyst residuum of many scattered granules is present and the sporozoites have prominent anterior and posterior refractile bodies. This parasite is clearly new to science and represents the first species of Isospora hosted by a New World bird species from this small family of silky flycatchers (Ptiliogonatidae).
