LAD1 promotes malignant progression by diminishing ubiquitin-dependent degradation of vimentin in gastric cancer
Background Ladinin-1 (LAD1), an anchoring filament protein, has been associated with several cancer types, including cancers of the colon, lungs, and breast. However, it is still unclear how and why LAD1 causes gastric cancer (GC). Methods Multiple in vitro and in vivo, functional gains and loss experiments were carried out in the current study to confirm the function of LAD1. Mass spectrometry was used to find the proteins that interact with LAD1. Immunoprecipitation analyses revealed the mechanism of LAD1 involved in promoting aggressiveness. Results The results revealed that the LAD1 was overexpressed in GC tissues, and participants with increased LAD1 expression exhibited poorer disease-free survival (DFS) and overall survival (OS). Functionally, LAD1 promotes cellular invasion, migration, proliferation, and chemoresistance in vivo and in vitro in the subcutaneous patient-and cell-derived xenograft (PDX and CDX) tumor models. Mechanistically, LAD1 competitively bound to Vimentin, preventing it from interacting with the E3 ubiquitin ligase macrophage erythroblast attacher (MAEA), which led to a reduction in K48-linked ubiquitination of Vimentin and an increase in Vimentin protein levels in GC cells. Conclusions In conclusion, the current investigation indicated that LAD1 has been predicted as a possible prognostic biomarker and therapeutic target for GC due to its ability to suppress Vimentin–MAEA interaction.
The E3 ubiquitin ligase MAEA promotes macrophage phagocytosis and inhibits gastrointestinal cancer progression by mediating PARP1 ubiquitination and degradation
While a role for the E3 ubiquitin ligase MAEA (macrophage erythroblast attacher) has been reported in several cancer types, its importance and mechanistic functions in gastrointestinal cancer (GIC) have yet to be established. The functions of MAEA in GIC were explored through and experiments, including loss- and gain-of-function analyses. Mass spectrometry was used to identify proteins that interact with MAEA. The mechanisms through which MAEA influences tumor aggression were examined through immunoprecipitation analyses. GIC patients exhibiting reduced expression of MAEA were found to exhibit worse disease-free and overall survival outcomes. MAEA was found to impair the proliferation and chemoresistance of GIC tumors and in subcutaneous xenograft model systems. The combination of MAEA and the PARP1 inhibitor veliparib resulted in enhanced oxaliplatin treatment efficacy From a mechanistic perspective, MAEA was found to mediate the K48-linked ubiquitination and degradation of PARP1, in addition to suppressing the M2 polarization of macrophages and enhancing macrophage phagocytic activity. These data suggest that MAEA offers value as a prognostic biomarker and target for the treatment of GIC owing to its ability to degrade PARP1 and augment the phagocytic activity of macrophages.
Magelonidae Cunningham & Ramage, 1888 (Annelida, Polychaeta)—Worldwide Identification Keys and the Re-Establishment of Maea Johnston, 1865
Shovel head worms (Annelida: Magelonidae) have long presented challenges to polychaete systematists due to their unique morphology and relative uniformity. This has been compounded by a lack of taxonomic work across key biogeographic regions. However, over the past few decades, a series of studies has progressively addressed these issues, refining our understanding of magelonid taxonomy and making taxonomic revisions of several regions. Whilst a standardised framework for morphological characters across the family has been produced, a worldwide review of taxonomic knowledge has been warranted. The information is herein presented alongside the first worldwide identification key in over 50 years. The much-needed key is presented in two formats: a dichotomous key divided by marine realms, and a pictorial key based on putative morphological groups. The key is additionally supplemented by the provision of an interactive map providing type locality data and links to taxonomic works. The pictorial key provides additional support for the unique terminology historically applied to the group. The genus Maea Johnston, 1865 is herein re-established for fifteen species possessing long rounded prostomia without horns, but possessing mucronate chaetae of the ninth chaetiger, and lateral abdominal pouches.

The reform of the nuclear safeguards and verification of nuclear materials as a condition of nuclear safety in the 21 Century The condition of maintaining nuclear safety in the twentieth century is to ensure a global system of nuclear materials safeguards. In practice, this means the need for a global system of verification of compliance with non‑proliferation requirements equipped with mechanisms of international control, accounting and physical protection of nuclear materials and facilities, as means of preventing the usage of nuclear dual‑use technology for non‑peaceful purposes (construction of nuclear weapons or other nuclear explosive devices). The track record of non‑proliferation regime discovered the weakness of the nuclear safeguards and verification system, in particular, limits of the IAEA control functions. Maintaining the “Nuclear Peace” can be assured only by a global system of physical protection, accounting, and security of nuclear materials, which means the need to improve the internationally comprehensive system of verification of compliance with non‑proliferation. The aim of the article is to present the principles of the international system of nuclear safeguards and the role of the International Atomic Energy Agency in maintaining nuclear peace and nuclear challenges of non‑proliferation regime reform. The analysis leads, among others, to the conclusion that the Agency should develop a procedure to standardize the rules, structure, and terminology of reporting non‑compliance, promote the internationalization of the fuel cycle while the UN Security Council should react faster in the case of non‑compliance detection.
RANBP9 and RANBP10 cooperate in regulating non-small cell lung cancer proliferation
RANBP9 and RANBP10, also called Scorpins, are essential components of the C-terminal to LisH (CTLH) complex, an evolutionarily conserved poorly investigated multisubunit E3 ligase. Their role in non-small cell lung cancer (NSCLC) is unknown. In this study, first we used stable loss-of function and overexpression inducible cell lines to investigate the ability of either RANBP9 or RANBP10 to form their own functional CTLH complex. Then, we probed lysates from patient tumors and analyzed data from publicly available repositories to investigate the expression of RANBP9 and RANBP10. Finally, we used inducible cell lines in vitro to recapitulate the expression observed in patients and investigate the changes of the proteome and the ubiquitylome associated with either RANBP9 or RANBP10 in NSCLC. Here, we show that the two Scorpins are both expressed in NSCLC cells and that either of them can independently support the formation of the CTLH complex. Short-term experiments revealed that the RANBP9 and RANBP10 proteins balance each other in terms of expression, and the acute overexpression of one or the other results in significant reshaping of the NSCLC cell proteome and ubiquitylome. A higher RANBP9/RANBP10 ratio is associated with greater proliferation in both NSCLC cell lines and patients. Acute increased expression of RANBP10 slows NSCLC cell proliferation and decreases the level of proliferation-associated proteins, including key players in DNA replication. We present evidence that the Scorpins act as partial antagonists and work together as one sophisticated rheostat to modulate the CTLH complex ubiquitylation output, which regulates cell proliferation and other key biological processes in NSCLC. These results suggest that the two Scorpins can be considered as targets for the treatment of NSCLC.
Cristo esperando la crucifixión: el hallazgo de una pintura de José Maea copiando a Alonso Cano y las versiones gráficas de Álvarez de Mon
Alonso Cano lleva a término a mitad del siglo XVII un lienzo con la representación de Cristo esperando la crucifixión influido por el naturalismo severo. No debió de pasar desapercibida esta pintura en el neoclasicismo, en tanto que dos autores como José Maea (1759-1826) y Manuel Álvarez de Mon (f. 1809) utilizaron este modelo según dos fórmulas diferentes: el primero, a través de una copia al óleo como objeto de aprendizaje y el segundo, con un dibujo y posterior grabado para optar a un premio en forma de concurso de la Academia de San Fernando.En el presente trabajo se estudian los aspectos artísticos, estéticos, iconográficos, de análisis de fuentes gráficas, así como de la historicidad propia de las obras.Con la exposición de una pintura inédita de Maea y las versiones gráficas de Álvarez de Mon, podemos analizar una convergencia hacia el estudio de una pintura barroca bajo los postulados del academicismo.
Discriminative stimulus functions in rats of AM1346, a high-affinity CB1R selective anandamide analog
Objective To characterize in vivo the high-affinity CB 1 cannabinoid receptor (CB 1 R) selective anandamide analog AM1346 [alkoxyacid amide of N -eicosa-tetraenylamine] using drug discrimination. Substitution tests involved Δ 9 -tetrahydrocannabinol (Δ 9 -THC) and R -(+)-methanandamide (mAEA), a metabolically stable analog of anandamide (AEA), as well as the CB 1 R antagonist/inverse agonist rimonabant; d -amphetamine and morphine were also examined to assess pharmacological specificity. Materials and methods Rats were initially trained to discriminate between i.p.-injected vehicle and 3 mg/kg AM1346 (group 3 mg/kg; t ′ = 20 min); subsequently, the rats were retrained with 5.6 mg/kg AM1346 (group 5.6 mg/kg; t ′ = 20 min). Results Dose-generalization curves of AM1346, Δ 9 -THC, and mAEA suggested the following order of potency: Δ 9 -THC > AM1346 > mAEA both for rats discriminating between 3 and 5.6 mg/kg AM1346 from vehicle. In group 3 mg/kg, challenge by 1 mg/kg rimonabant resulted in parallel shifts to the right of the dose-generalization curves for Δ 9 -THC and AM1346, suggesting surmountable antagonism. Surmountable antagonism was not demonstrated with rimonabant–mAEA combinations. A long duration of effect was indicated when 3 mg/kg AM1346 was examined after different time intervals following i.p. administration (group 3 mg/kg). The in vivo half-life was close to 5 h. Neither d -amphetamine nor morphine generalized in either of groups 3 mg/kg and 5.6 mg/kg, suggesting pharmacological specificity. Conclusion Unlike mAEA, the surmountable antagonism between rimonabant and AM1346 showed that the structural features of AEA can be modified to produce novel ligands that reduce the dissociation between the discriminative stimulus and rate decreasing effects of CB 1 R agonists derived from an AEA template.

Already, street thugs trained by American street gangs such as the Sons of Samoa and the Tongan Crip Gang have been blamed for the violence that engulfed Tonga's capital, Nuku'alofa, and a political assassination in Samoa. Salt Lake City has turned into a gang melting pot and Taufe'ulungaki claims Mormon children, who joined TCG, have since been deported back to Tonga by US authorities. Joined by disaffected youth from New Zealand they destroyed Nuku'alofa. [Teri Sommers] said the Tongan Crip Gang was involved in a long list of serious crime. The public felt its violence as did rival gangs. TCG's main enemy was fellow Tongan gang the Baby Regulators, which was big in the methamphetamine trade.
