Turning rice meiosis into mitosis
Introduction of clonal reproduction through seeds (apomixis) in crops has the potential to revolutionize agricul- ture by allowing self-propagation of any elite variety, in particular F1 hybrids. In the sexual model plant Arabidopsis thaliana synthetic clonal reproduction through seeds can be artificially implemented by (i) combining three muta- tions to turn meiosis into mitosis (MiMe) and (ii) crossing the obtained clonal gametes with a line expressing modified CENH3 and whose genome is eliminated in the zygote. Here we show that additional combinations of mutations can turn Arabidopsis meiosis into mitosis and that a combination of three mutations in rice (Oryza sativa) efficiently turns meiosis into mitosis, leading to the production of male and female clonal diploid gametes in this major crop. Suc- cessful implementation of the MiMe technology in the phylogenetically distant eudicot Arabidopsis and monocot rice opens doors for its application to any flowering plant and paves the way for introducing apomixis in crop species.
Mimetics: Towards Understanding Human Actions Out of Context
Recent methods for video action recognition have reached outstanding performances on existing benchmarks. However, they tend to leverage context such as scenes or objects instead of focusing on understanding the human action itself. For instance, a tennis field leads to the prediction playing tennis irrespectively of the actions performed in the video. In contrast, humans have a more complete understanding of actions and can recognize them without context. The best example of out-of-context actions are mimes, that people can typically recognize despite missing relevant objects and scenes. In this paper, we propose to benchmark action recognition methods in such absence of context and introduce a novel dataset, Mimetics, consisting of mimed actions for a subset of 50 classes from the Kinetics benchmark. Our experiments show that (a) state-of-the-art 3D convolutional neural networks obtain disappointing results on such videos, highlighting the lack of true understanding of the human actions and (b) models leveraging body language via human pose are less prone to context biases. In particular, we show that applying a shallow neural network with a single temporal convolution over body pose features transferred to the action recognition problem performs surprisingly well compared to 3D action recognition methods.
Recent Progress on Plant Apomixis for Genetic Improvement
Apomixis is a reproductive process that produces clonal seeds while bypassing meiosis (or apomeiosis) without undergoing fertilization (or pseudo-fertilization). The progenies are genetically cloned from their parents, retaining the parental genotype, and have great potential for the preservation of genes of interest and the fixing of heterosis. The hallmark components of apomixis include the formation of female gametes without meiosis, the development of fertilization-independent embryos, and the formation of functional endosperm. Understanding and utilizing the molecular mechanism of apomixis has far-reaching implications for plant genetic breeding and agricultural development. Therefore, this study focuses on the classification, influencing factors, genetic regulation, and molecular mechanism of apomixis, as well as progress in the research and application of apomixis-related genes in plant breeding. This work will elucidate the molecular mechanisms of apomixis and its application for plant genetic improvement.
Genetic manipulation of the genes for clonal seeds results in sterility in cotton
Background Heterosis is a common phenomenon in plants and has been extensively applied in crop breeding. However, the superior traits in the hybrids can only be maintained in the first generation but segregate in the following generations. Maintaining heterosis in generations has been challenging but highly desirable in crop breeding. Recent study showed that maternally produced diploid seeds could be achieved in rice by knocking out three meiosis related genes, namely REC8 , PAIR1 , OSD1 to create MiMe in combination with egg cell specific expression of BBM transcription factor, a technology called clonal seeds. Interestingly, there has been very limited reports indicating the feasibility of this approach in other crops. Results In this study, we aimed to test whether clonal seeds could be created in cotton. We identified the homologs of the three meiosis related genes in cotton and used the multiplex CRISPR/Cas9 gene editing system to simultaneously knock out these three genes in both A and D sub-genomes. More than 50 transgenic cotton plants were generated, and fragment analysis indicated that multiple gene knockouts occurred in the transgenic plants. However, all the transgenic plants were sterile apparently due to the lack of pollen. Pollination of the flowers of the transgenic plants using the wild type pollens could not generate seeds, an indication of defects in the formation of female sexual cells in the transgenic plants. In addition, we generated transgenic cotton plants expressing the cotton BBM gene driven by the Arabidopsis egg cell specific promoter pDD45 . Two transgenic plants were obtained, and both showed severely reduced fertility. Conclusions Overall, our results indicate that knockout of the clonal seeds related genes in cotton causes sterility and how to manipulate genes to create clonal seeds in cotton requires further research.
Imperial Nuptials at Pompeii: CIL IV.1261, an Obscene Take on the Marriage of Nero and Pythagoras
Among Pompeian wall inscriptions CIL IV.1261 is well known for its brisk obscenities and problems of text and interpretation. According to the text as it is currently printed, the graffito is generally understood to make a political statement of some kind about the mistreatment of the Roman citizen body; in what circumstances and by whom the collective of ciues Romani has been mistreated cannot be determined from the evidence of the graffito. In this paper, I present a revised text of the graffito, and I argue that the phrase ciuium Romanorum cunnus refers not to the Roman citizen body but to the emperor Nero and that the scene described in the text is the earliest extant testimony to an event that took place in 64 CE and was witnessed by persons unknown, Nero's marriage as bride to his freedman Pythagoras.
Staging Rachel: Rabbinic Midrash, Theatrical Mime, and Christian Martyrdom in Late Antiquity
Lamentations Rabbah Proem 24, a late ancient rabbinic midrash, is in many ways a unique text within the entire rabbinic corpus. It presents an extraordinary array of characters (including Abraham, Moses, the Torah, and even the alphabet) who are called upon to placate God, but fail. As their quest proves inconclusive, the biblical Rachel jumps into the fray to tell her story: how out of sisterly compassion she allowed Leah to take her own place in the conjugal bed on “her” wedding night. Disclosing to her sister the secretive “signs” she had shared with Jacob, Rachel crawled under the nuptial bed to respond to Jacob whenever he spoke. This scandalous autobiography transforms an apparent instance of illicit sex, the ideal material for theatrical stage mimes, into an act of martyrdom and sublime compassion. This article argues that the performance culture of the late ancient Mediterranean world provides the key for assessing this text’s originality. We begin with an analysis of the text, drawing attention to its theatrical qualities and its relationships with contemporary visual imagery (mosaics) and texts from outside the rabbinic milieu (Christian Apocrypha). We then examine the casting of midrashic Rachel as a response to both the mimic adulteress and the Christian martyr. Finally, we consider rabbinic familiarity with mime, particularly with its usefulness as a social mediator and agent of collective catharsis. It is precisely these aspects of mimic performance, we argue, that Rachel’s vignette appropriates in this fascinating rabbinic text.
ACTUAL AND INTELLECTUAL ARROWSHOTS IN SOPHRON (FRR. 50 AND 86 KASSEL–AUSTIN)
After some notes on the remarkable linguistic difficulties encountered by ancient Athenian readers of Sophron's text, this article will deal with the textual and interpretative problems posed by two fragments of the mimes that are more significant than it may appear at first sight. It is argued that a verbal form transmitted in fr. 86 K–A, whose correctness has systematically been doubted, is in fact linguistically unassailable and used in the description of an ambush that might be either military or metaphorical. As to fr. 50, once the correct reading of the principal verb has been established, we recover a subtle metaphor used by a character in a mime to criticise those who make hasty speculations about things unknown –something that reveals a sophistication rarely encountered in other fragments, and provides us with a glimpse into the reasons why Sophron's mimes enjoyed such respect in antiquity.
