Effects of Heat Stress on Plant-Nutrient Relations: An Update on Nutrient Uptake, Transport, and Assimilation
As a consequence of global climate change, the frequency, severity, and duration of heat stress are increasing, impacting plant growth, development, and reproduction. While several studies have focused on the physiological and molecular aspects of heat stress, there is growing concern that crop quality, particularly nutritional content and phytochemicals important for human health, is also negatively impacted. This comprehensive review aims to provide profound insights into the multifaceted effects of heat stress on plant-nutrient relationships, with a particular emphasis on tissue nutrient concentration, the pivotal nutrient-uptake proteins unique to both macro- and micronutrients, and the effects on dietary phytochemicals. Finally, we propose a new approach to investigate the response of plants to heat stress by exploring the possible role of plant peroxisomes in the context of heat stress and nutrient mobilization. Understanding these complex mechanisms is crucial for developing strategies to improve plant nutrition and resilience during heat stress.
Effects of Drought on Nutrient Uptake and the Levels of Nutrient-Uptake Proteins in Roots of Drought-Sensitive and -Tolerant Grasses
Climate change will increase drought in many regions of the world. Besides decreasing productivity, drought also decreases the concentration (%) of nitrogen (N) and phosphorous (P) in plants. We investigated if decreases in nutrient status during drought are correlated with decreases in levels of nutrient-uptake proteins in roots, which has not been quantified. Drought-sensitive (Hordeum vulgare, Zea mays) and -tolerant grasses (Andropogon gerardii) were harvested at mid and late drought, when we measured biomass, plant %N and P, root N- and P-uptake rates, and concentrations of major nutrient-uptake proteins in roots (NRT1 for NO3, AMT1 for NH4, and PHT1 for P). Drought reduced %N and P, indicating that it reduced nutrient acquisition more than growth. Decreases in P uptake with drought were correlated with decreases in both concentration and activity of P-uptake proteins, but decreases in N uptake were weakly correlated with levels of N-uptake proteins. Nutrient-uptake proteins per gram root decreased despite increases per gram total protein, because of the larger decreases in total protein per gram. Thus, drought-related decreases in nutrient concentration, especially %P, were likely caused, at least partly, by decreases in the concentration of root nutrient-uptake proteins in both drought-sensitive and -tolerant species.
Root effects on soil organic carbon
From recent developments on how roots affect soil organic carbon (SOC) an apparent paradox has emerged where roots drive SOC stabilization causing SOC accrual, but also SOC destabilization causing SOC loss. We synthesize current results and propose the new Rhizo-Engine framework consisting of two linked components: microbial turnover and the soil physicochemical matrix. The Rhizo-Engine is driven by rhizodeposition, root turnover, and plant uptake of nutrients and water, thereby accelerating SOC turnover through both stabilization and destabilization mechanisms. This Rhizo-Engine framework emphasizes the need for a more holistic approach to study root-driven SOC dynamics. This framework would provide better understanding of plant root effects on soil carbon sequestration and the sensitivity of SOC stocks to climate and land-use changes.
Field inoculation by arbuscular mycorrhizal fungi with contrasting life-history strategies differently affects tomato nutrient uptake and residue decomposition dynamics
Purpose Plant microbial biostimulants, such as arbuscular mycorrhizal fungi (AMF), enhance nutrient concentration in fruits, including tomato. However, field studies on tomato AMF inoculation are scarce. AMF species belonging to Gigasporaceae and Glomeraceae families known to vary in life-history strategies may determine differential effects on plant nutrient benefits and residue decomposition. Despite this, the effect of different life-history strategies on nutrient acquisition of tomato fruits has not been investigated yet. Methods We studied the effect of inoculation of two tomato varieties with four AMF species belonging to Glomeraceae and Gigasporaceae. Fungal colonization, yield, fruit nutrient concentration, litter decomposition, and bacterial and fungal abundances in soil were assessed in the field under organic agriculture. Results Overall Gigasporaceae promoted the concentration of nutrients in tomato fruits compared to Glomeraceae. A variability in AM fungal colonization and fruit nutrient concentration was detected within Glomeraceae. Scutellospora pellucida increased the yield (+ 27%) of var. Rio Grande with respect to Gigaspora gigantea . In var. Rio Grande, inoculation with Funneliformis mosseae did not change litter decomposition as compared to non-inoculated controls, whereas it was lower than in Sclerocystis sinuosa and Gigasporaceae species, which showed the highest decomposition rates. AMF inoculation promoted soil total bacterial and fungal abundance and fungal:bacterial (F:B) ratio compared to controls, and members of Gigasporaceae had the highest F:B ratio. Conclusion These findings pointed at the inclusion of AM fungal life-history strategy within the selection criteria for the development of biofertilizers able to enhance the nutritional value of vegetables under organic farming systems.
Heat Stress Decreases Levels of Nutrient-Uptake and -Assimilation Proteins in Tomato Roots
Global warming will increase root heat stress, which is already common under certain conditions. Effects of heat stress on root nutrient uptake have rarely been examined in intact plants, but the limited results indicate that heat stress will decrease it; no studies have examined heat-stress effects on the concentration of nutrient-uptake proteins. We grew Solanum lycopersicum (tomato) at 25 °C/20 °C (day/night) and then transferred some plants for six days to 35 °C /30 °C (moderate heat) or 42 °C/37 °C (severe heat) (maximum root temperature = 32 °C or 39 °C, respectively); plants were then moved back to control conditions for seven days to monitor recovery. In a second experiment, plants were grown for 15 days at 28 °C/23 °C, 32 °C/27 °C, 36 °C/31 °C, and 40 °C/35 °C (day/night). Concentrations of nutrient-uptake and -assimilation proteins in roots were determined using protein-specific antibodies and ELISA (enzyme-linked immunosorbent assay). In general, (1) roots were affected by heat more than shoots, as indicated by decreased root:shoot mass ratio, shoot vs. root %N and C, and the level of nutrient metabolism proteins vs. less sensitive photosynthesis and stomatal conductance; and (2) negative effects on roots were large and slow-to-recover only with severe heat stress (40 °C–42 °C). Thus, short-term heat stress, if severe, can decrease total protein concentration and levels of nutrient-uptake and -assimilation proteins in roots. Hence, increases in heat stress with global warming may decrease crop production, as well as nutritional quality, partly via effects on root nutrient relations.
Dynamic model of flexible phytoplankton nutrient uptake
The metabolic machinery of marine microbes can be remarkably plastic, allowing organisms to persist under extreme nutrient limitation. With some exceptions, most theoretical approaches to nutrient uptake in phytoplankton are largely dominated by the classic Michaelis–Menten (MM) uptake functional form, whose constant parameters cannot account for the observed plasticity in the uptake apparatus. Following seminal ideas by earlier researchers, we propose a simple cell-level model based on a dynamic view of the uptake process whereby the cell can regulate the synthesis of uptake proteins in response to changes in both internal and external nutrient concentrations. In our flexible approach, the maximum uptake rate and nutrient affinity increase monotonically as the external nutrient concentration decreases. For low to medium nutrient availability, our model predicts uptake and growth rates larger than the classic MM counterparts, while matching the classic MM results for large nutrient concentrations. These results have important consequences for global coupled models of ocean circulation and biogeochemistry, which lack this regulatory mechanism and are thus likely to underestimate phytoplankton abundances and growth rates in oligotrophic regions of the ocean.
Magnesium application improves the morphology, nutrients uptake, photosynthetic traits, and quality of tobacco (Nicotiana tabacum L.) under cold stress
Cold stress is one of the major constraints limiting the productivity of many important crops, including tobacco ( Nicotiana tabacum L.) production and quality worldwide. However, the role of magnesium (Mg) nutrition in plants has been frequently overlooked, especially under cold stress, and Mg deficiency adversely affects plant growth and development. Here, we evaluated the influence of Mg under cold stress on tobacco morphology, nutrient uptake, photosynthetic and quality attributes. The tobacco plants were grown under different levels of cold stress, i.e., 8°C, 12°C, 16°C, including with a controlled temperature of 25°C, and evaluated their effects with Mg (+Mg) and without Mg (–Mg) application. Cold stress resulted in reduced plant growth. However, the +Mg alleviated the cold stress and significantly increased the plant biomass on an average of 17.8% for shoot fresh weight, 20.9% for root fresh weight, 15.7% for shoot dry weight, and 15.5% for root dry weight. Similarly, the nutrients uptake also increased on average for shoot-N (28.7%), root-N (22.4%), shoot-P (46.9%), root-P (7.2%), shoot-K (5.4%), root-K (28.9%), shoot-Mg (191.4%), root-Mg (187.2%) under cold stress with +Mg compared to –Mg. Mg application significantly boosted the photosynthetic activity (Pn 24.6%) and increased the chlorophyll contents ( Chl-a (18.8%), Chl-b (25%), carotenoids (22.2%)) in the leaves under cold stress in comparison with –Mg treatment. Meanwhile, Mg application also improved the quality of tobacco, including starch and sucrose contents, on an average of 18.3% and 20.8%, respectively, compared to –Mg. The principal component analysis revealed that tobacco performance was optimum under +Mg treatment at 16°C. This study confirms that Mg application alleviates cold stress and substantially improves tobacco morphological indices, nutrient absorption, photosynthetic traits, and quality attributes. In short, the current findings suggest that Mg application may alleviate cold stress and improve tobacco growth and quality.
Temporal Differentiation of Resource Capture and Biomass Accumulation as a Driver of Yield Increase in Intercropping
Intercropping, i.e., the simultaneous cultivation of different crops on the same field, has demonstrated yield advantages compared to monoculture cropping. These yield advantages have often been attributed to complementary resource use, but few studies quantified the temporal complementarity of nutrient acquisition and biomass production. Our understanding of how nutrient uptake rates of nitrogen (N) and phosphorous (P) and biomass accumulation change throughout the growing season and between different neighbors is limited. We conducted weekly destructive harvests to measure temporal trajectories of N and P uptake and biomass production in three crop species (oat, lupin, and camelina) growing either as isolated single plants, in monocultures or as intercrops. Additionally, we quantified organic acid exudation in the rhizosphere and biological N 2 -fixation of lupin throughout the growing season. Logistic models were fitted to characterize nutrient acquisition and biomass accumulation trajectories. Nutrient uptake and biomass accumulation trajectories were curtailed by competitive interactions, resulting in earlier peak rates and lower total accumulated nutrients and biomass compared to cultivation as isolated single plants. Different pathways led to overyielding in the two mixtures. The oat–camelina mixture was characterized by a shift from belowground temporal niche partitioning of resource uptake to aboveground competition for light during the growing season. The oat–lupin mixture showed strong competitive interactions, where lupin eventually overyielded due to reliance on atmospheric N and stronger competitiveness for soil P compared to oat. Synthesis: This study demonstrates temporal shifts to earlier peak rates of plants growing with neighbors compared to those growing alone, with changes in uptake patterns suggesting that observed temporal shifts in our experiment were driven by competitive interactions rather than active plant behavior to reduce competition. The two differing pathways to overyielding in the two mixtures highlight the importance of examining temporal dynamics in intercropping systems to understand the underlying mechanisms of overyielding.
An efficient three-dimensional rhizosphere modeling capability to study the effect of root system architecture on soil water and reactive transport
Aims The objective of this research was to develop a three-dimensional (3D) rhizosphere modeling capability for plant-soil interactions by integrating plant biophysics, water and ion uptake and release from individual roots, variably saturated flow, and multicomponent reactive transport in soil. Methods We combined open source software for simulating plant and soil interactions with parallel computing technology to address highly-resolved root system architecture (RSA) and coupled hydrobiogeochemical processes in soil. The new simulation capability was demonstrated on a model grass, Brachypodium distachyon . Results In our simulation, the availability of water and nutrients for root uptake is controlled by the interplay between 1) transpiration-driven cycles of water uptake, root zone saturation and desaturation; 2) hydraulic redistribution; 3) multicomponent competitive ion exchange; 4) buildup of ions not taken up during kinetic nutrient uptake; and 5) advection, dispersion, and diffusion of ions in the soil. The uptake of water and ions by individual roots leads to dynamic, local gradients in ion concentrations. Conclusion By integrating the processes that control the fluxes of water and nutrients in the rhizosphere, the modeling capability we developed will enable exploration of alternative RSAs and function to advance the understanding of the coupled hydro-biogeochemical processes within the rhizosphere.
Cell wall-bound silicon optimizes ammonium uptake and metabolism in rice cells
Abstract Background and Aims Turgor-driven plant cell growth depends on cell wall structure and mechanics. Strengthening of cell walls on the basis of an association and interaction with silicon (Si) could lead to improved nutrient uptake and optimized growth and metabolism in rice (Oryza sativa). However, the structural basis and physiological mechanisms of nutrient uptake and metabolism optimization under Si assistance remain obscure. Methods Single-cell level biophysical measurements, including in situ non-invasive micro-testing (NMT) of NH4+ ion fluxes, atomic force microscopy (AFM) of cell walls, and electrolyte leakage and membrane potential, as well as whole-cell proteomics using isobaric tags for relative and absolute quantification (iTRAQ), were performed. Key Results The altered cell wall structure increases the uptake rate of the main nutrient NH4+ in Si-accumulating cells, whereas the rate is only half in Si-deprived counterparts. Conclusions Rigid cell walls enhanced by a wall-bound form of Si as the structural basis stabilize cell membranes. This, in turn, optimizes nutrient uptake of the cells in the same growth phase without any requirement for up-regulation of transmembrane ammonium transporters. Optimization of cellular nutrient acquisition strategies can substantially improve performance in terms of growth, metabolism and stress resistance.
