Comparative proteomics analysis of adult Haemonchus contortus isolates from Ovis ammon
Haemonchus contortus is an important parasite that causes disease that seriously endangers ruminant animals cattle, sheep, goat, and camel. Here, we compared the proeomics analysis of three adult Haemonchus contortus isolates from mouflons ( Ovis ammon ). A total of 1,299 adult worm proteins were identified, and 461 proteins were quantified, of which 82 (108), 83 (97), and 97 (86) significantly upregulated (downregulated) differentially expressed proteins (DEPs) were detected among pairwise comparisons (1-vs.-3, 2-vs.-3, and 2-vs.-1). Liquid chromatography–tandem mass spectrometry (LC−MS/MS) and bioinformatic analysis indicated that these DEPs are mainly concentrated in cellular composition, molecular function, biological function, and catabolism pathways. In addition, Gene Ontology (GO) classification and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses were carried out to screen the DEPs. The main biological processes involved were nucleotide, nucleotide phosphate, ribonucleotide, purine-containing compound, purine ribonucleotide, single-organism, oxoacid, organic, carboxylic, oxoacid metabolic processes and single-organism catabolic processes. The majority of KEGG pathways were found to be related to metabolic pathways, biosynthesis of secondary metabolites, biosynthesis of antibiotics, carbon metabolism, and microbial metabolism in diverse environments. Moreover, we also found differences in the expression of some important or novel regulatory proteases, such as serine hydroxymethyl transferase (SHMT), dihydrolipoyl dehydrogenase (DLD), and transket pyr domain-containing protein (TKPD). In summary, label-free proteomic analysis of adult H. contortus worms displayed significant differences in three different individual isolates, which helps to improve our understanding of the growth and metabolic mechanisms of H. contortus in different individuals and relative natural environments and provides novel drug targets for the treatment of parasitic diseases.
Investigating the potential impact of trophy hunting of wild ungulates on snow leopard Panthera uncia conservation in Tajikistan
Declining availability of prey is potentially a major factor limiting snow leopard Panthera uncia populations in Tajikistan and neighbouring states. Conservation initiatives to develop community-based trophy hunting programmes for ibex Capra sibirica and argali Ovis ammon polii aim to provide financial incentives for communities to limit poaching of wild ungulates. Such programmes could help to reverse local declines in ibex and argali populations, and consequently snow leopard populations, while simultaneously improving the economic status of local people. However, in practice the desired effect may not materialize. To investigate the premise, we estimated the population density of the snow leopard, using a spatial capture–recapture model based on camera trapping in two study areas (each c. 1,000 km2) in the Tajik Pamirs: a well-managed trophy hunting concession and an otherwise similar area where grazing and poaching are unmanaged. We used distance-truncated counts to assess relative densities of wild and domestic ungulates between sites, and faecal analyses to compare the dietary habits of snow leopards. Our data were limited in scope but suggested that the density of snow leopards and the relative density of wild and domestic ungulates may have been greater in the hunting concession, where wild ungulates accounted for a greater proportion of prey items. Our results provide preliminary evidence that trophy hunting of ungulates may be a viable tool for achieving snow leopard conservation goals; however, we conclude that further investigation is necessary to adequately address the question.
Do Marco Polo argali Ovis ammon polii persist in Pakistan?
The distribution range of the Near Threatened Marco Polo argali, or Marco Polo sheep, Ovis ammon polii is restricted to the Pamir Mountains, spanning Afghanistan, Tajikistan, China and Pakistan. Until the early 1970s the Marco Polo argali was abundant in northern areas of Pakistan, particularly in the Khunjerab and Misgar Valleys around the Pamir Knot, bordering China, Afghanistan and Tajikistan. In Pakistan the subspecies now occurs only in one small watershed, in Khunjerab National Park, bordering China, which it visits sporadically during summer. We used map-based questionnaire surveys, double-observer surveys and camera trapping in a search for Marco Polo argali in the Pakistani Pamirs. We observed a herd of 19 individuals in Karachanai Nallah, in Khunjerab National Park, in 2012. The population that was formerly present in the Misgar Valley appears to have been extirpated, presumably as a result of anthropogenic factors such as poaching, competition with livestock, habitat disturbance, and the construction of a fence along the international border with China. Transboundary migration and range expansion into Pakistan could be facilitated by removal of the border fence adjacent to the Kilik–Mintika area and by limiting livestock grazing in former lambing areas.
Status assessment of the Endangered snow leopard Panthera uncia and other large mammals in the Kyrgyz Alay, using community knowledge corrected for imperfect detection
The Endangered snow leopard Panthera uncia occurs in the Central Asian Mountains, which cover c. 2 million km2. Little is known about its status in the Kyrgyz Alay Mountains, a relatively narrow stretch of habitat connecting the southern and northern global ranges of the species. In 2010 we gathered information on current and past (1990, the last year of the Soviet Union) distributions of snow leopards and five sympatric large mammals across 14,000 km2 of the Kyrgyz Alay. We interviewed 95 key informants from local communities. Across 49 400-km2 grid cells we obtained 1,606 and 962 records of species occurrence (site use) in 1990 and 2010, respectively. The data were analysed using the multi-season site occupancy framework to incorporate uncertainty in detection across interviewees and time periods. High probability of use by snow leopards in the past was recorded in > 70% of the Kyrgyz Alay. Between the two sampling periods 39% of sites showed a high probability of local extinction of snow leopard. We also recorded high probability of local extinction of brown bear Ursus arctos (84% of sites) and Marco Polo sheep Ovis ammon polii (47% of sites), mainly in regions used intensively by people. Data indicated a high probability of local colonization by lynx Lynx lynx in 41% of the sites. Although wildlife has declined in areas of central and eastern Alay, regions in the north-west, and the northern and southern fringes appear to retain high conservation value.
Seasonal changes in the sexual segregation patterns of Marco Polo sheep in Taxkorgan Nature Reserve
Sexual segregation is observed widely among ruminants, but remains poorly understood in argali. Since sexual segregation is affected by habitat, and in different environments may differ among species and subspecies, we studied this behavioral pattern in Marco Polo sheep (Ovis ammon polii) in their natural habitat at high elevations of the Pamir Plateau using three distinct measurements. We found that, similar to other wild sheep, female and male Marco Polo sheep were highly segregated most of the year and gathered into mixed-sex groups mostly during the rut, which occurred later and for a shorter time than in other argali subspecies experiencing milder climates. This phenomenon was related to the severe environmental conditions of the Pamir Plateau that provided a very short period of favorable weather for birthing and rearing young; correspondingly, aggregation into mixed-sex groups during the rut also occurred later and for a shorter period of time than in other argali populations at lower elevations. Moreover, the degree of segregation between females and males increased with the males’ age. Subadult males switched between male and female groups, staying more with males during lambing and with females during the rut. The timing and duration of sexual segregation and aggregation of Marco Polo sheep were a result of adaption to their local environment.
Habitat shift and time budget of the Tibetan argali: the influence of livestock grazing
Livestock production is the primary source of livelihood and income in most of the high steppe and alpine regions of the Indian Trans‐Himalaya. In some areas, especially those established or proposed for biodiversity conservation, recent increases in populations of domestic livestock, primarily sheep and goats, have raised concern about domestic animals competitively excluding wild herbivores from the rangelands. We evaluated the influence of domestic sheep and goat grazing on the habitat use and time budget of the endangered Tibetan argali Ovis ammon hodgsoni in the proposed Gya‐Miru Wildlife Sanctuary, Ladakh, India. We asked if the domestic sheep and goat grazing and collateral human activities relegate the argali to sub‐optimal habitats, and alter their foraging time budgets. Data were collected on habitat use and time budget of a population of c. 50 argalis before and after c. 2,000 sheep and goats moved onto their winter pasture in the Tsabra catchment of the aforementioned reserve. Following the introduction of domestic sheep and goats, argalis continued to use the same catchment but shifted to steeper habitats, closer to cliffs, with lower vegetation cover, thus abandoning previously used plant communities with denser cover. Argalis’ active time spent foraging also decreased by 10% in response to the presence of livestock. These results suggest a clear disturbance effect of livestock on argalis, and indicate a potential for competition, conceivably a significant disadvantage for argalis in winter when forage availability is minimal.
The complete mitochondrial genome of Ovis ammon darwini (Artiodactyla: Bovidae)
Ovis ammon darwini : has been listed as a near-threatened species by IUCN-World Conservation Union due to over-hunting and poaching, competition with domestic livestock, and habitat destruction. Study on molecular biology research provides the scientific basis for the protection and sustainable utilization of key endangered species. Mitochondrial genome is very useful for researches in ecology, systematics and conservation biology. In this study, the complete mitochondrial genome sequence of O. a. darwini was determined by next-generation sequencing data, which is 16,618 bp in length and contains 13 protein-coding genes(PCGs), 2 rRNAs genes, 22 tRNAs genes and a non-coding control region. Base composition of genome is A(33.7 %), C(25.8 %), G(13.1 %), T(27.4 %) with an A + T content of 61.1 %. Phylogenetic analysis suggested that the systematic status of O. a. darwini was more closed related to O. a. hodgsoni clustered with O. ammon . The mitogenome of O. a. darwini offered significant information for molecular genetic research of O. ammon .
Status of Marco Polo sheep Ovis ammon polii in China and adjacent countries: conservation of a Vulnerable subspecies
To determine the status and distribution of the Vulnerable Marco Polo sheep or argali Ovis ammon polii surveys were made in the Pamir Mountains, where the Hindu Kush, Karakoram, and Kunlun ranges meet. In China a total of 851 Marco Polo sheep were observed within the Taxkorgan Nature Reserve and 1,448 north of the Reserve. No reliable population estimate is available for Tajikistan but in five census blocks totalling 1,977 km2 densities ranged from negligible to 1.4 km−2. A total of 624 sheep were counted in Afghanistan, and total numbers may approach 1,000. Fewer than 150 individuals are said to visit Pakistan seasonally. Hunting for meat has decimated the Marco Polo sheep populations in all four countries but with better protection they appear to be on the increase in China. Transboundary cooperation is essential if the species is to be adequately protected and managed as argali readily cross international borders, and a four-country Pamir Peace Park has been proposed and is under discussion.
Snow Leopard and Himalayan Wolf: Food Habits and Prey Selection in the Central Himalayas, Nepal
Top carnivores play an important role in maintaining energy flow and functioning of the ecosystem, and a clear understanding of their diets and foraging strategies is essential for developing effective conservation strategies. In this paper, we compared diets and prey selection of snow leopards and wolves based on analyses of genotyped scats (snow leopards n = 182, wolves n = 57), collected within 26 sampling grid cells (5×5 km) that were distributed across a vast landscape of ca 5000 km2 in the Central Himalayas, Nepal. Within the grid cells, we sampled prey abundances using the double observer method. We found that interspecific differences in diet composition and prey selection reflected their respective habitat preferences, i.e. snow leopards significantly preferred cliff-dwelling wild ungulates (mainly bharal, 57% of identified material in scat samples), whereas wolves preferred typically plain-dwellers (Tibetan gazelle, kiang and argali, 31%). Livestock was consumed less frequently than their proportional availability by both predators (snow leopard = 27%; wolf = 24%), but significant avoidance was only detected among snow leopards. Among livestock species, snow leopards significantly preferred horses and goats, avoided yaks, and used sheep as available. We identified factors influencing diet composition using Generalized Linear Mixed Models. Wolves showed seasonal differences in the occurrence of small mammals/birds, probably due to the winter hibernation of an important prey, marmots. For snow leopard, occurrence of both wild ungulates and livestock in scats depended on sex and latitude. Wild ungulates occurrence increased while livestock decreased from south to north, probably due to a latitudinal gradient in prey availability. Livestock occurred more frequently in scats from male snow leopards (males: 47%, females: 21%), and wild ungulates more frequently in scats from females (males: 48%, females: 70%). The sexual difference agrees with previous telemetry studies on snow leopards and other large carnivores, and may reflect a high-risk high-gain strategy among males.
Prey Preferences of the Snow Leopard (Panthera uncia): Regional Diet Specificity Holds Global Significance for Conservation
The endangered snow leopard is a large felid that is distributed over 1.83 million km(2) globally. Throughout its range it relies on a limited number of prey species in some of the most inhospitable landscapes on the planet where high rates of human persecution exist for both predator and prey. We reviewed 14 published and 11 unpublished studies pertaining to snow leopard diet throughout its range. We calculated prey consumption in terms of frequency of occurrence and biomass consumed based on 1696 analysed scats from throughout the snow leopard's range. Prey biomass consumed was calculated based on the Ackerman's linear correction factor. We identified four distinct physiographic and snow leopard prey type zones, using cluster analysis that had unique prey assemblages and had key prey characteristics which supported snow leopard occurrence there. Levin's index showed the snow leopard had a specialized dietary niche breadth. The main prey of the snow leopard were Siberian ibex (Capra sibrica), blue sheep (Pseudois nayaur), Himalayan tahr (Hemitragus jemlahicus), argali (Ovis ammon) and marmots (Marmota spp). The significantly preferred prey species of snow leopard weighed 55±5 kg, while the preferred prey weight range of snow leopard was 36-76 kg with a significant preference for Siberian ibex and blue sheep. Our meta-analysis identified critical dietary resources for snow leopards throughout their distribution and illustrates the importance of understanding regional variation in species ecology; particularly prey species that have global implications for conservation.
