Ypsiloturia bi-tentaculata ha sido considerada una especie del Océano Pacífico (Oshima 1915, Pawson 1965, Gage et al. 1985), sin embargo, Martins y Tavares (2018) la reportó para el Atlántico sudoccidental y el océano Indico. Antibody design using LSTM based deep generative model from phage display library for affinity maturation

Molecular evolution is an important step in the development of therapeutic antibodies. However, the current method of affinity maturation is overly costly and labor-intensive because of the repetitive mutation experiments needed to adequately explore sequence space. Here, we employed a long short term memory network (LSTM)—a widely used deep generative model—based sequence generation and prioritization procedure to efficiently discover antibody sequences with higher affinity. We applied our method to the affinity maturation of antibodies against kynurenine, which is a metabolite related to the niacin synthesis pathway. Kynurenine binding sequences were enriched through phage display panning using a kynurenine-binding oriented human synthetic Fab library. We defined binding antibodies using a sequence repertoire from the NGS data to train the LSTM model. We confirmed that likelihood of generated sequences from a trained LSTM correlated well with binding affinity. The affinity of generated sequences are over 1800-fold higher than that of the parental clone. Moreover, compared to frequency based screening using the same dataset, our machine learning approach generated sequences with greater affinity.

What is Traditional Leadership of Tatuha Luang?: The Ethnographic Studies at Diamond Mining in Kalimantan, Indonesia

Purpose: This study aims to analyze the leadership of Tatuha Luang in diamond panning, to analyze the leadership of Tatuha Luang as the application of leadership based on Banjar cultural values, and to analyze the contribution of diamond panning to the community in Cempaka Village, Banjarbaru City, South Borneo. Design/Methodology/Approach: The research design used is qualitative research with an ethnographic approach as a knife of analysis. The key informants consisted of 3 key informants and 7 assistant informants. The data analysis of this ethnographic research refers to the Spradley model. Findings: Tatuha Luang's leadership has role as a director, mentor, and strategy for the success of diamond panning. A leader has the knowledge and skills to lead and behave as a role model for the community. Tatuha Luang's leadership plays important role, as a highly valued and respected figure by the community. Tatuha Luang has strong principles and convictions in living the optimism of success in running a diamond panning business. Tatuha Luang's leadership has a strong charisma to the community for the comprehension and behavior is consistent with those held by the Cempaka Banjarbaru people of South Kalimantan. It facilitates Tatuha Luang's leadership give advice that will be easily followed by the community. Tatuha Luang's leadership was given the position of coordinator regarding customary law, customs and habits of the community. The selection of informal leaders is determined past success and the feelings of sympathy and antipathy of a person or group towards him. Research limitations/Implications: The information sources are Limited on leadership bibliography based on local wisdom Tatuha Luang. This leadership contains the values of collaboration and synergy of the balance of the real world and the supernatural which is unique and special Practical implications: The results of this research model can be implemented for the diamond mining community in Banjarbaru in particular and in South Kalimantan in general, and even other community organizations can adapt it for community organizations. Social Implications: The findings of the major proposition with a leadership model based on local wisdom which has universal values and can be applied not only in the mining community but can be used in all lines of life in the wider social organization. Originality/Value: This study contribute to leadership theory based on local wisdom, especially in the findings of a conceptual model, namely the leadership of Tatuha Luang which prioritizes the local wisdom of the Banjarbaru community in particular and in South Kalimantan in general.

Semi-Automated Cell Panning for Efficient Isolation of FGFR3-Targeting Antibody

Phage display technology is a widely used practical tool for isolating binding molecules against the desired targets in phage libraries. In the case of targeting the membrane protein with its natural conformation, conventional bio-panning has limitations on the efficient screening of the functionally relevant antibodies. To enrich the single-chain variable fragment (scFv) pools for recognizing the natural conformation of the membrane targets, the conventional bio-panning and screening process was modified to include the semi-automated cell panning protocol. Using FGFR3-overexpressing patient-derived cancer cells, biotin-X-DHPE was introduced and coupled to Streptavidin-coated magnetic beads for use in the solution-phage bio-panning procedure. The resulting clones of scFv were compared to the diversity of the binding region, especially on CDR-H3. The clones enriched further by cell-based panning procedure possessed a similar binding site and the CDR-H3 loop structure. The resulting antibodies inhibited cell growth and induced target degradation. This process may be a useful tool for screening biologically related antibodies that recognize natural conformational structure on cell membrane protein. Furthermore, cell-based panning has the potential to further expand to a high-throughput screening (HTS) system and automation process.

Cross-HLA targeting of intracellular oncoproteins with peptide-centric CARs

The majority of oncogenic drivers are intracellular proteins, thus constraining their immunotherapeutic targeting to mutated peptides (neoantigens) presented by individual human leukocyte antigen (HLA) allotypes
. However, most cancers have a modest mutational burden that is insufficient to generate responses using neoantigen-based therapies
. Neuroblastoma is a paediatric cancer that harbours few mutations and is instead driven by epigenetically deregulated transcriptional networks
. Here we show that the neuroblastoma immunopeptidome is enriched with peptides derived from proteins that are essential for tumourigenesis and focus on targeting the unmutated peptide QYNPIRTTF, discovered on HLA-A*24:02, which is derived from the neuroblastoma dependency gene and master transcriptional regulator PHOX2B. To target QYNPIRTTF, we developed peptide-centric chimeric antigen receptors (CARs) using a counter-panning strategy with predicted potentially cross-reactive peptides. We further hypothesized that peptide-centric CARs could recognize peptides on additional HLA allotypes when presented in a similar manner. Informed by computational modelling, we showed that PHOX2B peptide-centric CARs also recognize QYNPIRTTF presented by HLA-A*23:01 and the highly divergent HLA-B*14:02. Finally, we demonstrated potent and specific killing of neuroblastoma cells expressing these HLAs in vitro and complete tumour regression in mice. These data suggest that peptide-centric CARs have the potential to vastly expand the pool of immunotherapeutic targets to include non-immunogenic intracellular oncoproteins and widen the population of patients who would benefit from such therapy by breaking conventional HLA restriction.

Translating ribosome affinity purification (TRAP) combines cell type–specific transgene expression with affinity purification of translating ribosomes and can be used to study the cell type–specific mRNA profiles of any genetically defined cell type.
Cellular diversity and architectural complexity create barriers to understanding the function of the mammalian CNS at a molecular level. To address this problem, we have recently developed a methodology that provides the ability to profile the entire translated mRNA complement of any genetically defined cell population. This methodology, which we termed translating ribosome affinity purification, or TRAP, combines cell type–specific transgene expression with affinity purification of translating ribosomes. TRAP can be used to study the cell type–specific mRNA profiles of any genetically defined cell type, and it has been used in organisms ranging from
Drosophila melanogaster
to mice and human cultured cells. Unlike other methodologies that rely on microdissection, cell panning or cell sorting, the TRAP methodology bypasses the need for tissue fixation or single-cell suspensions (and the potential artifacts that these treatments introduce) and reports on mRNAs in the entire cell body. This protocol provides a step-by-step guide to implement the TRAP methodology, which takes 2 d to complete once all materials are in hand.

Recent Advances of Proteins, Polysaccharides and Lipids-Based Edible Films/Coatings for Food Packaging Applications: a Review

Most fruits and vegetables are susceptible to spoilage after harvest, the transportation and preservation conditions can further decrease the product quality. As one of the main food preservation technologies, edible films/coatings are made of eatable materials to form thin layers that can maintain food freshness during transport and storage. This review summarizes different films/coating materials, including proteins (soy, whey, wheat gluten, gelatin), polysaccharides (chitosan, starch, cellulose) and lipids. However, the films/coatings prepared by single material have many deficiencies which can be made up by the combination of composite films/coatings. Moreover, several prepared methods (electrospinning, casting, extrusion, dipping, fluidized-bed, spraying, panning) used for films/coatings are introduced. Finally, the application and future directions of films/coatings in the preservation of fruits, vegetables and other food products are also presented.
