Physalis pubescens L. branch and leaf extracts inhibit lymphoma proliferation by inducing apoptosis and cell cycle arrest
Physalis pubescens L. is an annual or perennial plant in the family Solanaceae It is used in traditional medicine for treating sore throats, coughs, urinary discomfort, and astringent pain, and externally for pemphigus and eczema in northern China. The proliferation inhibitory activity and mechanisms of the ethyl acetate extract (PHY-EA) from the leaves of Physalis pubescens were investigated. High performance liquid chromatography was used to identify the chemical composition of PHY-EA; sulforhodamine B was used to detect the proliferation inhibitory effect of PHY-EA on MCF-7, CA-46, Hela, HepG2, B16, and other tumor cells; flow cytometry was used to detect the effect of PHY-EA on the lymphoma cell cycle and apoptosis; Western blot was used to detect the expression of the cycle- and apoptosis-related proteins. The expression of Ki-67 and cleaved caspase 3 was detected by immunohistochemistry. The results showed that PHY-EA contained physalin B, physalin O, and physalin L. PHY-EA blocked the cell cycle of G2/M→G0/G1 in lymphoma cells and induced apoptosis in tumor cells. Mouse transplantation tumor experiments showed that PHY-EA had a significant inhibitory effect on mouse transplantation tumors, and the tumor volume and weight were significantly reduced. In conclusion, PHY-EA has a good antiproliferative effect on Burkkit lymphoma, indicating its potential medicinal value.
Yield, quality, antioxidants and mineral nutrients of Physalis angulata L. and Physalis pubescens L. fruits as affected by genotype under organic management
Introduction and selection of unconventional plants with high concentration of biologically active compounds is one of the worthy ways for producing functional food, which is beneficial to human health. Research was carried out in northern Europe (Russia) with the purpose to assess yield, quality and biologically active compounds concentration in Physalis angulata and Physalis pubescens fruits. P. angulata cultivars Konditer and Konditer 2 gave the highest yield (11.3 and 11.0 t·ha−1 respectively), due to the highest mean fruit weight (80 and 70 g respectively); P. pubescens variety Zolotaya rossip had the worst outcome due to the very small berries (3 g), in spite of their highest number per plant (165). ‘Zolotaya rossip’ fruits overall attained higher values of quality indicators compared to all P. angulata cultivars. Positive correlations were recorded between dry matter and polyphenols as well as between total sugars and polyphenols. Physalis fruits showed to be a good source of antioxidants, K, Mg and P for human beings. Taste index turned out highly reliable in evaluating fruit quality and it was dependant on dry matter, total sugars, polyphenols and Ca.
Production of a Yogurt Drink Enriched with Golden Berry (Physalispubescens L.) Juice and Its Therapeutic Effect on Hepatitis in Rats
Fermented dairy products have been associated with multiple health benefits. The present study aimed to produce a functional yogurt drink fortified with golden berry juice and assess its therapeutic effect on hepatitis rats. Thirty male albino rats were randomly divided into two major groups. The first group included the control (-) animals (six rats) and was fed a standard diet, whereas the second group included 24 rats that were fed a standard diet and injected with carbon tetrachloride (CCl4) for 2 weeks to trigger chronic damage of the liver (hepatitis); they were then divided into four groups (six rats/group): Group 2: hepatitis, fed on a standard diet as a positive control group; Group 3: received a basal diet with 5 mL of the yogurt drink; Group 4: received a basal diet with 5 mL of the yogurt drink fortified with 10% golden berry juice. Group 5: received a basal diet with 5 mL of the yogurt drink fortified with 20% golden berry juice. Various biological parameters were determined. Yogurt drink treatments were evaluated for their chemical, phytochemical, and sensory properties, as well as for their effects on hepatoprotective activity by determining various biochemical parameters. We found that the yogurt drinks containing golden berry juice exhibited no significant differences in fat, protein, and ash content compared with the control samples. Moreover, the yogurt drinks containing golden berry juice exhibited the highest content of total phenolic compounds, antioxidant activity, and organoleptic scores among all treatments. In addition, rats fed on a diet fortified with yogurt drinks containing golden berry juice for 8 weeks exhibited higher potential hepatoprotective effects compared with the liver injury control group. This improvement was partly observed in the group that received the yogurt drink containing golden berry juice. Therefore, we concluded that golden berry juice can be recommended as a natural additive in the manufacture of functional yogurt drinks, as it showed a potential hepatoprotective effect in rats with hepatitis.
Genotypic effect on fruit production and quality, antioxidant content and elemental composition of organically grown Physalis angulata L. and Physalis pubescens L
Research was carried out in northern Europe (Russia) with the purpose of assessing the yield, quality, biologically active compounds and elemental composition of and fruits. cultivars ‘Konditer’ and ‘Konditer 2’ gave the highest yield and mean fruit weight (11.2 t ha and 75 g, on average), whereas variety Zolotaya Rossip had the worst outcome as it produced the smallest berries (3 g). ‘Zolotaya Rossip’ fruits attained the highest values of dry matter and soluble solids, while ‘Violet’ and ‘Korolek’ the highest polyphenol and ascorbic acid concentrations, respectively. Organic acid composition of fruits revealed high varietal differences and predominant accumulation of tartaric and malic acids. Positive correlations were recorded between dry matter and polyphenols, as well as between Fe and Mn, but an adverse correlation between Si and Cd content. fruits showed to be a good source of beneficial compounds for human beings, such as polyphenols and ascorbic acid, as well as mineral nutrients, i.e. K, Mg, P, B, Co, Cu, Fe, Li, Mn, Si, V and Zn.
Application of a chitosan coating as a carrier for natamycin to maintain the storage quality of ground cherry (Physalis pubescens L.)
Ground cherry ( Physalis pubescens L.) is a kind of berry fruit favored by consumers in China; however, this fruit is particularly vulnerable to physiological senescence and pathogen attack during the traditional cold storage period. In order to maintain storage quality, a 1.5% (w/w) chitosan (CS) water solution containing 50 mg/L of natamycin (NA) was introduced. After all treatments were completed, the fruit was stored at 0 °C and sampled every 10 d. At each sampling date, the following tests were performed: mold and yeast analyses; enzyme activity and content analyses which included superoxide dismutase (SOD), ascorbate peroxidase (APX), and malondialdehyde (MDA); and color analysis. In addition, a sensory evaluation was carried out for quality assessment at the end of the storage period. The results showed that the application of a chitosan coating combined with natamycin (CSNA) enhanced the activity of superoxide dismutase (SOD) and ascorbate peroxidase (APX), reduced the physiological rate of senescence, and inhibited pathogen attack. Thus, CSNA treatment can maintain ground cherries at an acceptable level of storage quality for 50 d.
A centenary for bacterial spot of tomato and pepper
Disease symptoms Symptoms include water‐soaked areas surrounded by chlorosis turning into necrotic spots on all aerial parts of plants. On tomato fruits, small, water‐soaked, or slightly raised pale‐green spots with greenish‐white halos are formed, ultimately becoming dark brown and slightly sunken with a scabby or wart‐like surface. Deciphering the evolutionary development of the “Chinese lantern” within Solanaceae
Main conclusionThe key genetic variation underlying the evo-devo of ICS in Solanaceae may be further pinpointed using an integrated strategy of forward and reverse genetics studies under the framework of phylogeny.The calyx of Physalis remains persistent throughout fruit development. Post-flowering, the fruiting calyx is inflated rapidly to encapsulate the berry, giving rise to a “Chinese lantern” structure called inflated calyx syndrome (ICS). It is unclear how this novelty arises. Over the past 2 decades, the role of MADS-box genes in the evolutionary development (evo-devo) of ICS has mainly been investigated within Solanaceae. In this review, we analyze the main achievements, challenges, and new progress. ICS acts as a source for fruit development, provides a microenvironment to protect fruit development, and assists in long-distance fruit dispersal. ICS is a typical post-floral trait, and the onset of its development is triggered by specific developmental signals that coincide with fertilization. These signals can be replaced by exogenous gibberellin and cytokinin application. MPF2-like heterotopic expression and MBP21-like loss have been proposed to be two essential evolutionary events for ICS origin, and manipulating the related MADS-box genes has been shown to affect the ICS size, sepal organ identity, and/or male fertility, but not completely disrupt ICS. Therefore, the core genes or key links in the ICS biosynthesis pathways may have undergone secondary mutations during evolution, or they have not yet been pinpointed. Recently, we have made some encouraging progress in acquiring lantern mutants in Physalis floridana. In addition to technological innovation, we propose an integrated strategy to further analyze the evo-devo mechanisms of ICS in Solanaceae using forward and reverse genetics studies under the framework of phylogeny.
Carotenogenesis and chromoplast development during ripening of yellow, orange and red colored Physalis fruit
Main conclusion Formation of specific ultrastructural chromoplastidal elements during ripening of fruits of three different colored Physalis spp. is closely related to their distinct carotenoid profiles. The accumulation of color-determining carotenoids within the chromoplasts of ripening yellow, orange, and red fruit of Physalis pubescens  L., Physalis peruviana  L., and Physalis alkekengi  L., respectively, was monitored by high-performance liquid chromatography/diode array detector/tandem mass spectrometry (HPLC–DAD-MS/MS) as well as light and transmission electron microscopy. Both yellow and orange fruit gradually accumulated mainly β-carotene and lutein esters at variable levels, explaining their different colors at full ripeness. Upon commencing β-carotene biosynthesis, large crystals appeared in their chromoplasts, while large filaments protruding from plastoglobules were characteristic elements of chromoplasts of orange fruit. In contrast to yellow and orange fruit, fully ripe red fruit contained almost no β-carotene, but esters of both β-cryptoxanthin and zeaxanthin at very high levels. Tubule bundles and unusual disc-like crystallites were predominant carotenoid-bearing elements in red fruit. Our study supports the earlier hypothesis that the predominant carotenoid type might shape the ultrastructural carotenoid deposition form, which is considered important for color, stability and bioavailability of the contained carotenoids.
Multiple and integrated functions of floral C-class MADS-box genes in flower and fruit development of Physalis floridana
Key messageThis work reveals potentially multiple and integrated roles in flower and fruit development of floral C-class MADS-box genes in Physalis.The Physalis fruit features a morphological novelty, the Chinese lantern. Floral C-class MADS-domain AGAMOUS-like (AG-like) proteins can interact with the identified regulators of this novel structure. However, the developmental role of the floral C-class genes is unknown in Physalis. Here, we characterized two AG-like genes from Physalis floridana, designated PFAG1 and PFAG2. The two paralogous genes shared around 61.0% of sequence identity and had similar expression domains, with different expression levels in the floral and berry development. However, the genes had distinct expression patterns in leaf and calyx development. Protein–protein interaction analyses revealed that PFAG1 and PFAG2 could commonly or specifically dimerize with certain floral MADS-domain proteins as well as non-MADS-domain proteins involved in various floral developmental processes. Gene downregulation analyses demonstrated that PFAG1 may repress PFAG2, but PFAG2 did not affect PFAG1. Downregulating PFAG1 led to incomplete floral homeotic variation in the stamens and carpels, and alteration of petal coloration pattern, while downregulating PFAG2 did not result in any floral homeotic variation. PFAG1 affected pollen maturation, while PFAG2 affected female fertility. However, simultaneously downregulating PFAG1 and PFAG2 caused loss of the complete C-function, indicating that the two PFAG genes interact to determine the identity and functionality of androecia and gynoecia organs. Their potential roles in regulating fruit size and the Chinese lantern are also discussed. Our results reveal functional divergence of floral C-class MADS-box genes in Physalis, demonstrating that they may play multiple and integrated roles in flower and fruit development.
Transcriptomic variation of the flower–fruit transition in Physalis and Solanum
Main conclusion Gene expression variations in response to fertilization between Physalis and Solanum might play essential roles in species divergence and fruit evolution. Fertilization triggers variation in fruit development and morphology. The Chinese lantern, a morphological novelty derived from the calyx, is formed upon fertilization in Physalis but is not observed in Solanum . The underlying genetic variations are largely unknown. Here, we documented the developmental and morphological differences in the flower and fruit between Physalis floridana and Solanum pimpinellifolium and then evaluated both the transcript sequence variation and gene expression at the transcriptomic level at fertilization between the two species. In Physalis transcriptomic analysis, 468 unigenes were identified as differentially expressed genes (DEGs) that were strongly regulated by fertilization across 3 years. In comparison with tomato, 14,536 strict single-copy orthologous gene pairs were identified between P. floridana and S. pimpinellifolium in the flower–fruit transcriptome. Nine types of gene variations with specific GO-enriched patterns were identified, covering 58.82% orthologous gene pairs that were DEGs in either trend or dosage at the flower–fruit transition between the two species, which could adequately distinguish Solanum and Physalis , implying that differential gene expression at fertilization might play essential roles during the divergence and fruit evolution of Solanum – Physalis . Virus-induced gene silencing analyses revealed the developmental roles of some transcription factor genes in fertility, Chinese lantern development, and fruit weight control in Physalis . This study presents the first floral transcriptomic resource of Physalis , and reveals some candidate genetic variations accounting for the early fruit developmental evolution in Physalis in comparison to Solanum .
