The Water Balance Representation in Urban‐PLUMBER Land Surface Models

Urban Land Surface Models (ULSMs) simulate energy and water exchanges between the urban surface and atmosphere. However, earlier systematic ULSM comparison projects assessed the energy balance but ignored the water balance, which is coupled to the energy balance. Here, we analyze the water balance representation in 19 ULSMs participating in the Urban‐PLUMBER project using results for 20 sites spread across a range of climates and urban form characteristics. As observations for most water fluxes are unavailable, we examine the water balance closure, flux timing, and magnitude with a score derived from seven indicators expecting better scoring models to capture the latent heat flux more accurately. We find that the water budget is only closed in 57% of the model‐site combinations assuming closure when annual total incoming fluxes (precipitation and irrigation) fluxes are within 3% of the outgoing (all other) fluxes. Results show the timing is better captured than magnitude. No ULSM has passed all water balance indicators for any site. Models passing more indicators do not capture the latent heat flux more accurately refuting our hypothesis. While output reporting inconsistencies may have negatively affected model performance, our results indicate models could be improved by explicitly verifying water balance closure and revising runoff parameterizations. By expanding ULSM evaluation to the water balance and related to latent heat flux performance, we demonstrate the benefits of evaluating processes with direct feedback mechanisms to the processes of interest. Plain Language Summary Urban environments have their own local climates including typically higher nocturnal temperatures compared with rural areas. Ideally, modeling cities should capture their influences on the atmosphere above them. As the energy and water balances are linked by evaporation, a good water balance representation will support a good energy balance simulation. Focusing on the water balance, we find the water balance in models could be improved by paying attention to closure and runoff. Intermediate filaments (IF) are a major component of the metazoan cytoskeleton and are essential for normal cell morphology, motility, and signal transduction. Dysregulation of IFs causes a wide range of human diseases, including skin disorders, cardiomyopathies, lipodystrophy, and neuropathy. Despite this pathophysiological significance, how cells regulate IF structure, dynamics, and function remains poorly understood. Here, we show that site-specific modification of the prototypical IF protein vimentin with O-linked β-N-acetylglucosamine (O-GlcNAc) mediates its homotypic protein-protein interactions and is required in human cells for IF morphology and cell migration. In addition, we show that the intracellular pathogen Chlamydia trachomatis, which remodels the host IF cytoskeleton during infection, requires specific vimentin glycosylation sites and O-GlcNAc transferase activity to maintain its replicative niche. Our results provide new insight into the biochemical and cell biological functions of vimentin O-GlcNAcylation, and may have broad implications for our understanding of the regulation of IF proteins in general. Like the body's skeleton, the cytoskeleton gives shape and structure to the inside of a cell. Yet, unlike a skeleton, the cytoskeleton is ever changing. The cytoskeleton consists of many fibers each made from chains of protein molecules. One of these proteins is called vimentin and it forms intermediate filaments in the cytoskeleton. Many different types of cells contain vimentin and a lot of it is found in cancer cells that have spread beyond their original location to other sites in the body. Cells use chemical modifications to regulate cytoskeleton proteins. For example, through a process called glycosylation, cells can reversibly attach a sugar modification called O-GlcNAc to vimentin. O-GlcNAc can be attached to several different parts of vimentin and each location may have a different effect. It is not currently clear how cells control their vimentin filaments or what role O-GlcNAc plays in this process. Using genetic engineering, Tarbet et al. produced human cells in the laboratory with modified vimentin proteins. These altered proteins lacked some of the sites for O-GlcNAc attachment. The goal was to see whether the loss of O-GlcNAc at a specific location would affect fiber formation and cell behavior. The results showed one site where vimentin needs O-GlcNAc to form fibers. Without O-GlcNAc at this site, cells could not migrate towards chemical signals. In addition, in normal human cells, Chlamydia bacteria hijack vimentin and rearrange the filaments to form a cage around themselves for protection. However, the cells lacking O-GlcNAc on vimentin were resistant to infection by Chlamydia bacteria. These findings highlight the importance of O-GlcNAc on vimentin in healthy cells and during infection. Vimentin’s contribution to cell migration may also help to explain its role in the spread of cancer. The importance of O-GlcNAc suggests it could be a new target for therapies. Yet, it also highlights the need for caution due to the delicate balance between the activity of vimentin in healthy and diseased cells. In addition, human cells produce about 70 other vimentin-like proteins and further work will examine if they are also affected by O-GlcNAc.

Driving Profiles of Light Commercial Vehicles of Craftsmen and the Potential of Battery Electric Vehicles When Charging on Company Premises

This paper examines the extent to which it is possible to replace conventional light commercial vehicles in the heating, ventilation and air conditioning and plumbing trade with battery electric vehicles with an unchanged usage profile. GPS trackers are used to record the position data of 22 craft vehicles with combustion engines from eleven companies over the duration of one working week. Within this paper, various assumptions (battery capacity and average consumption) are made for battery electric vehicles and the charging power on the company premises. The potential of battery electric vehicles is evaluated based on the assumption that they are charged only on company premises. Using the collected data and the assumptions made, theoretical state of charge curves are calculated for the vehicles. The driving profiles of the individual vehicles differ greatly, and the suitability of battery electric vehicles should be considered individually. Battery capacity, vehicle energy consumption and charging power at the company have a substantial influence on the suitability of battery electric vehicles. Furthermore, there are differences between vehicles that can charge on the company premises at night and those that cannot or can only do so on some days.
