The Influence of Physical Fields (Magnetic and Electric) and LASER Exposure on the Composition and Bioactivity of Cinnamon Bark, Patchouli, and Geranium Essential Oils
In recent years, essential oils (EOs) have received increased attention from the research community, and the EOs of cinnamon, patchouli, and geranium have become highly recognized for their antibacterial, antifungal, antiviral, and antioxidant effects. Due to these properties, they have become valuable and promising candidates for addressing the worldwide threat of antimicrobial resistance and other diseases. Simultaneously, studies have revealed promising new results regarding the effects of physical fields (magnetic and electric) and LASER (MEL) exposure on seed germination, plant growth, biomass accumulation, and the yield and composition of EOs. In this frame, the present study aims to investigate the influence of MEL treatments on cinnamon, patchouli, and geranium EOs, by specifically examining their composition, antimicrobial properties, and antioxidant activities. Results showed that the magnetic influence has improved the potency of patchouli EO against L. monocytogenes, S. enteritidis, and P. aeruginosa, while the antimicrobial activity of cinnamon EO against L. monocytogenes was enhanced by the electric and laser treatments. All exposures have increased the antifungal effect of geranium EO against C. albicans. The antioxidant activity was not modified by any of the treatments. These findings could potentially pave the way for a deeper understanding of the efficiency, the mechanisms of action, and the utilization of EOs, offering new insights for further exploration and application.
Pharmacological activities and mechanisms of action of Pogostemon cablin Benth: a review
Patchouli (“Guanghuoxiang”) or scientifically known as Pogostemon cablin Benth, belonging to the family Lamiaceae, has been used in traditional Chinse medicine (TCM) since the time of the Eastern Han dynasty. In TCM theory, patchouli can treat colds, nausea, fever, headache, and diarrhea. Various bioactive compounds have been identified in patchouli, including terpenoids, phytosterols, flavonoids, organic acids, lignins, glycosides, alcohols, pyrone, and aldehydes. Among the numerous compounds, patchouli alcohol, β-patchoulene, patchoulene epoxide, pogostone, and pachypodol are of great importance. The pharmacological impacts of these compounds include anti-peptic ulcer effect, antimicrobial effect, anti-oxidative effect, anti-inflammatory effect, effect on ischemia/reperfusion injury, analgesic effect, antitumor effect, antidiabetic effect, anti-hypertensive effect, immunoregulatory effect, and others.For this review, we examined publications from the previous five years collected from PubMed, Web of Science, Springer, and the Chinese National Knowledge Infrastructure databases. This review summarizes the recent progress in phytochemistry, pharmacology, and mechanisms of action and provides a reference for future studies focused on clinical applications of this important plant extract.
Integrated Analysis of Physiological, mRNA Sequencing, and miRNA Sequencing Data Reveals a Specific Mechanism for the Response to Continuous Cropping Obstacles in Pogostemon cablin Roots
Pogostemon cablin (patchouli) is a commercially important medicinal and industrial crop grown worldwide for its medicinal and aromatic properties. Patchoulol and pogostone, derived from the essential oil of patchouli, are considered valuable components in the cosmetic and pharmaceutical industries. Due to its high application value in the clinic and industry, the demand for patchouli is constantly growing. Unfortunately, patchouli cultivation has suffered due to severe continuous cropping obstacles, resulting in a significant decline in yield and quality. Moreover, the physiological and transcriptional changes in patchouli in response to continuous cropping obstacles remain unclear. This has greatly restricted the development of the patchouli industry. To explore the mechanism underlying the rapid response of patchouli roots to continuous cropping stress, integrated analysis of the transcriptome and miRNA profiles of patchouli roots under continuous and noncontinuous cropping conditions in different growth periods was conducted using RNA sequencing (RNA-seq) and miRNA-seq and complemented with physiological data. The physiological and biochemical results showed that continuous cropping significantly inhibited root growth, decreased root activity, and increased the activity of antioxidant enzymes (superoxide dismutase, peroxidase, and catalase) and the levels of osmoregulators (malondialdehyde, soluble protein, soluble sugar, and proline). Subsequently, we found 4,238, 3,494, and 7,290 upregulated and 4,176, 3,202, and 8,599 downregulated differentially expressed genes (DEGs) in the three growth periods of continuously cropped patchouli, many of which were associated with primary carbon and nitrogen metabolism, defense responses, secondary metabolite biosynthesis, and transcription factors. Based on miRNA-seq, 927 known miRNAs and 130 novel miRNAs were identified, among which 67 differentially expressed miRNAs (DEMIs) belonging to 24 miRNA families were induced or repressed by continuous cropping. By combining transcriptome and miRNA profiling, we obtained 47 miRNA-target gene pairs, consisting of 18 DEMIs and 43 DEGs, that likely play important roles in the continuous cropping response of patchouli. The information provided in this study will contribute to clarifying the intricate mechanism underlying the patchouli response to continuous cropping obstacles. In addition, the candidate miRNAs and genes can provide a new strategy for breeding continuous cropping-tolerant patchouli.
Combined physiological and transcriptome analysis revealed the response mechanism of Pogostemon cablin roots to p-hydroxybenzoic acid
Pogostemon cablin (patchouli) cultivation is challenged by serious soil sickness, of which autotoxins accumulation is a major cause. p-hydroxybenzoic acid (p-HBA) is one of the main autotoxins of patchouli. However, the molecular mechanism underlying the response of patchouli to p-HBA remains unclear. In this study, RNA-sequencing combined with physiological analysis was used to monitor the dynamic transcriptomic and physiological changes in patchouli seedlings 0, 6, 12, 24, 48, and 96 h after p-HBA treatment. p-HBA stress inhibited root biomass accumulation, induced excessive hydrogen peroxide accumulation and lipid peroxidation, and activated most antioxidant enzymes. Compared with that of the control, the osmotic adjustment substance content was elevated with treatment. Subsequently, 15,532, 8,217, 8,946, 2,489, and 5,843 differentially expressed genes (DEGs) at 6, 12, 24, 48, and 96 h after p-HBA treatment, respectively, were identified in patchouli roots. GO functional enrichment analysis showed that the DEGs were enriched mainly in plasma membrane, defense response, response to chitin, DNA-binding transcription factor activity and abscisic acid-activated signaling pathway. The upregulated genes were involved in glycolysis/gluconeogenesis, cysteine and methionine metabolism, starch and sucrose metabolism, biosynthesis of unsaturated fatty acids, and linoleic acid metabolism. Genes associated with MAPK signaling pathway-plant, plant-pathogen interaction, plant hormone signal transduction were downregulated with p-HBA treatment. These pathways are related to root browning and rotting, leading to plant death.
Simultaneous determination of multiple components in rat plasma by UPLC-MS/MS for pharmacokinetic studies after oral administration of Pogostemon cablin extract
Introduction: Pogostemon cablin (PC) is used in traditional Chinese medicine and food, as it exerts pharmacological effects, such as immune-modulatory, antibacterial, antioxidant, antitumor, and antiviral. Currently, the pharmacokinetics (PK) studies of PC mainly focus on individual components. However, research on these individual components cannot reflect the actual PK characteristics of PC after administration. Therefore, the simultaneous determination of multiple components in rat plasma using UPLC-MS/MS was used for the pharmacokinetic study after oral administration of PC extract in this study, providing reference value for the clinical application of PC. Methods: In the present study, a reliable and sensitive ultra-high performance liquid chromatography/tandem mass spectrometry (UPLC-MS/MS) method was developed and validated for the simultaneous determination of 15 prototype components (vanillic acid, vitexin, verbascoside, isoacteoside, hyperoside, cosmosiin, apigenin, β-rhamnocitrin, acacetin, ombuin, pogostone, pachypodol, vicenin-2, retusin, and diosmetin-7-O-β-D-glucopyranoside) in rat plasma after oral administration of the PC extract. Plasma samples were prepared via protein precipitation using acetonitrile, and icariin was used as the internal standard (IS). Results: The intra-day and inter-day accuracies ranged from −12.0 to 14.3%, and the precision of the analytes was less than 11.3%. The extraction recovery rate of the analytes ranged from 70.6−104.5%, and the matrix effects ranged from 67.4−104.8%. Stability studies proved that the analytes were stable under the tested conditions, with a relative standard deviation lower than 14.1%. Conclusion: The developed method can be applied to evaluate the PK of 15 prototype components in PC extracts of rats after oral administration using UPLC-MS/MS, providing valuable information for the development and clinical safe, effective, and rational use of PC.
Polysaccharides From Pogostemon cablin (Blanco) Benth.: Characterization and Antioxidant Activities
Two polysaccharide fractions from Pogostemon cablin (Blanco) Benth. ( P. cablin ) (designated as PCB-1 and PCB2-1) were isolated by water extraction and purified by Sepharose chromatography. The chemical properties of the polysaccharides were characterised, and their antioxidant activities were evaluated. The sugar content of the crude polysaccharide (PCB), PCB-1, and PCB2-1 was 58.74, 90.23 and 88.61%, respectively. The molecular weights of PCB-1 and PCB2-1 were determined to be 97.8 and 12.8 kDa, respectively. Monosaccharide composition analysis showed that all the three polysaccharides consisted of mannose, rhamnose, galacturonic acid, galactose, glucose, and arabinose, but with varying molar ratios. The polysaccharides exhibited significantly high antioxidant activities in vitro based on the scavenging activity against hydroxyl radicals, metal ion-chelating and ferric-reducing abilities. In vivo experiments in an oxidatively damaged mice model showed that PCB-1 increased the levels of antioxidant enzymes, including superoxide dismutase, catalase, and glutathione peroxidase, and inhibited malondialdehyde formation in the serum and liver. These findings suggest that PCB-1 has significant potential as an antioxidant in functional foods.
In vitro induction and characterization of tetraploid Patchouli (Pogostemon cablin Benth.) plant
Patchouli (Pogostemon cablin Benth.) is one kind of plant from Indonesia which is producing oil. In vitro induction of tetraploid in Patchouli plant and cytological and morphological characterization were conducted. Induced polyploidy was done by culturing leaf explant on MS medium added with a varying concentration of colchicine. It was observed that colchicine on MS medium mostly affect to growth of explant and shoot regeneration. Tetraploid plants were determined by observation of chromosome number. The tetraploid plants have chromosome number (2n = 4x = 128), twice higher than those of diploid plants (2n = 2x = 64). The tetraploid plants exhibited much larger stomata size but less than diploid plants. The tetraploid plants also showed more glandular hairs than diploid plants. Significant different in morphological characters between the diploid and tetraploid plants were noted. Leaf size and stem diameter of the tetraploids plant are larger than the diploid plants. In this study, the tetraploids Patchouli have a promising potential as a superior variety.
Network Pharmacology-Based Analysis of Pogostemon cablin (Blanco) Benth Beneficial Effects to Alleviate Nonalcoholic Fatty Liver Disease in Mice
Nonalcoholic fatty liver disease (NAFLD) is the most common cause of chronic liver disease and is associated with high morbidity and mortality. Pogostemon cablin (Blanco) Benth/Huo Xiang (HX) is a perennial herb with unique anti-oxidant and anti-inflammatory properties, and thus, can positively affect liver function. In this study, we used network pharmacology to predict the potential mechanism of HX on NAFLD. Pharmacological experiments were used to verify the effect of HX on the functions of NAFLD. Network pharmacology identified nine components that interacted with 82 NAFLD-related targets, revealing four target genes: TNF , IL6 , TP53 , and AKT1 . HX prevents the development and progression of NAFLD through different pathways and targets with quercetin-regulated lipid metabolism, anti-inflammatory, and anti-oxidant pathways playing an essential role in the treatment of NAFLD. Compared with feeding HFD, HX significantly attenuated lipid accumulation in vivo with mice and also in vitro with mouse liver cells. A high dose of HX decreased hepatocyte lipid accumulation and the abundance of SREBF1 and FASN. Validation experiments revealed that HX inhibited the activation of NF-κB/IκB signaling and decreased the release and levels of pro-inflammatory factors (TNF-α and IL-6). These data suggest that HX can attenuate abnormal lipid metabolic responses and enhance antioxidant mechanisms. Thus, the pharmacological effects from plants used in traditional Chinese medicine are achievde through a multi-level response.
Biological Activity of Pogostemon cablin Essential Oil and Its Potential Use for Food Preservation
This study aimed to analyze the biological activity of the essential oil Pogostemon cablin (PCEO) to determine the antioxidant, antimicrobial, antibiofilm, insecticidal activity, and chemical composition of the essential oil. We analyzed the structure of biofilms on various surfaces using the MALDI-TOF MS Biotyper and evaluated the antimicrobial effect of the vapor phase of the essential oil in a food model. We determined the main volatile components of PCEO as patchouli alcohol 31.0%, α-bulnesene 21.3%, and α-guaiene 14.3%. The free radical scavenging activity was high and reached 71.4 ± 0.9%, corresponding to 732 ± 8.1 TEAC. The antimicrobial activity against bacteria was weak to moderate. We recorded strong activity against yeast. The antifungal activity was very weak in the contact application. Biofilm-producing bacteria were moderately inhibited by PCEO. The change in biofilm structure due to essential oil was demonstrated by MALDI-TOF MS Biotyper analysis. Vapor phase application in a food model showed relatively strong effects against bacteria and significantly higher antifungal efficacy. The insecticidal activity was observed only at higher concentrations of essential oil. Based on the findings, PCEO can be used in the food industry as an antifungal substance in extending the shelf life of bakery products and as protection in the storage of root vegetables.
SMRT sequencing of full-length transcriptome and gene expression analysis in two chemical types of Pogostemon cablin (Blanco) Benth
(Blanco) Benth. also called patchouli, is a traditional medicinal and aromatic plant that grows mainly in Southeast Asia and China. In China, is divided into two chemical types: the patchouliol-type and the pogostone-type. Patchouliol-type patchouli usually grow taller, with thicker stems and bigger leaves, and produce more aromatic oil. To better understand the genetic differences between the two chemical types that contribute to their differences in morphology and biosynthetic capabilities, we constructed transcriptomes from both chemical types using the Pacific Biosciences (PacBio) Sequel platform and performed differential expression analysis of multiple tissues using Illumina short reads. In this study, using single-molecule real-time (SMRT) long-read sequencing, we obtained 22.07 GB of clean data and 134,647 nonredundant transcripts from two chemical types. Additionally, we identified 126,576 open reading frames (ORFs), 100,638 coding sequences (CDSs), 4,106 long noncoding RNAs (lncRNAs) and 6,829 transcription factors (TFs) from two chemical types of . We adopted PacBio and Illumina sequencing to identify differentially expressed transcripts (DEGs) in three tissues of the two chemical types. More DEGs were observed in comparisons of different tissues collected from the same chemical type relative to comparisons of the same tissue collected from different chemical types. Furthormore, using KEGG enrichment analysis of DEGs, we found that the most enriched biosynthetic pathways of secondary metabolites of the two chemical types were \"terpenoid backbone biosynthesis\", \"phenylpropanoid biosynthesis\", \"plant hormone signal transduction\", \"sesquiterpenoid and triterpenoid biosynthesis\", \"ubiquinone and other terpenoid-quinone biosynthesis\", \"flavonoid biosynthesis\", and \"flavone and flavonol biosynthesis\". However, the main pathways of the patchouliol-type also included \"diterpene biosynthesis\" and \"monoterpene biosynthesis\". Additionally, by comparing the expression levels of the three tissues verified by qRT-PCR, more DEGs in the roots were upregulated in the mevalonate (MVA) pathway in the cytoplasm, but more DEGs in the leaves were upregulated in the methylerythritol phosphate (MEP) pathway in the plastid, both of which are important pathways for terpenoids biosynthesis.
