Genome-wide association study of alcohol dependence:significant findings in African- and European-Americans including novel risk loci
We report a GWAS of alcohol dependence (AD) in European-American (EA) and African-American (AA) populations, with replication in independent samples of EAs, AAs and Germans. Our sample for discovery and replication was 16 087 subjects, the largest sample for AD GWAS to date. Numerous genome-wide significant (GWS) associations were identified, many novel. Most associations were population specific, but in several cases were GWS in EAs and AAs for different SNPs at the same locus,showing biological convergence across populations. We confirmed well-known risk loci mapped to alcohol-metabolizing enzyme genes, notably ADH1B (EAs: Arg48His, P =1.17 × 10 −31 ; AAs: Arg369Cys, P =6.33 × 10 −17 ) and ADH1C in AAs (Thr151Thr, P =4.94 × 10 −10 ), and identified novel risk loci mapping to the ADH gene cluster on chromosome 4 and extending centromerically beyond it to include GWS associations at LOC100507053 in AAs ( P =2.63 × 10 −11 ), PDLIM5 in EAs ( P =2.01 × 10 −8 ), and METAP in AAs ( P =3.35 × 10 −8 ). We also identified a novel GWS association (1.17 × 10 −10 ) mapped to chromosome 2 at rs1437396, between MTIF2 and CCDC88A, across all of the EA and AA cohorts, with supportive gene expression evidence, and population-specific GWS for markers on chromosomes 5, 9 and 19. Several of the novel associations implicate direct involvement of, or interaction with, genes previously identified as schizophrenia risk loci. Confirmation of known AD risk loci supports the overall validity of the study; the novel loci are worthy of genetic and biological follow-up. The findings support a convergence of risk genes (but not necessarily risk alleles) between populations, and, to a lesser extent, between psychiatric traits.
Ontogeny of prosocial behavior across diverse societies
Humans are an exceptionally cooperative species, but there is substantial variation in the extent of cooperation across societies. Understanding the sources of this variability may provide insights about the forces that sustain cooperation. We examined the ontogeny of prosocial behavior by studying 326 children 3–14 y of age and 120 adults from six societies (age distributions varied across societies). These six societies span a wide range of extant human variation in culture, geography, and subsistence strategies, including foragers, herders, horticulturalists, and urban dwellers across the Americas, Oceania, and Africa. When delivering benefits to others was personally costly, rates of prosocial behavior dropped across all six societies as children approached middle childhood and then rates of prosociality diverged as children tracked toward the behavior of adults in their own societies. When prosocial acts did not require personal sacrifice, prosocial responses increased steadily as children matured with little variation in behavior across societies. Our results are consistent with theories emphasizing the importance of acquired cultural norms in shaping costly forms of cooperation and creating cross-cultural diversity.
Healthcare utilization is increased in children living in urban areas, with ethnicity-related disparities: A big data analysis study
This study aimed to investigate differences in pediatric healthcare utilization in Israel over 10 years by examining differences across populations defined by living environment and ethnicity. Data was obtained from the Clalit Health Care data warehouse, covering over 250,000 children residing in Haifa and Western Galilee districts. The population groups were categorized based on ethnicity (Jewish vs Arab) and residential settings (urban vs rural). Healthcare utilization was consistently higher among Jewish than Arab children, irrespective of the specific dimension analyzed. Additionally, urban-dwelling children exhibited higher usage rates than those residing in rural areas in all investigated dimensions. However, Jewish children showed significantly about 18% lower hospitalization rates than Arab children across all years ( P  < 0.001). No significant differences in hospitalizations were observed between urban and rural children (RR 0.999, CI (0.987–1.011)). Notably, the study revealed reduced antibiotic consumption and hospitalizations over the years for all populations. Additionally, we found that Arab children and those living in rural areas had reduced access to healthcare, as evidenced by 10–40% fewer physician visits, laboratory tests, and imaging ( P  < 0.001).    Conclusion : This study highlights the substantial population-based disparities in healthcare utilization among children in Israel despite the equalizing effect of the national health insurance law. Rural and low socioeconomic populations seem to have reduced healthcare access, showing decreased healthcare utilization. Consequently, it is imperative to address these disparities and implement targeted interventions to enhance healthcare access for Arab children and rural communities. The decline in antibiotic usage and hospitalizations suggests positive trends in pediatric health care, necessitating ongoing efforts to ensure equitable access and quality of care for all populations. What is Known: • Healthcare systems worldwide vary in coverage and accessibility, including Israel, which stands out for its diverse population. • Existing research primarily focuses on healthcare utilization among adults, leaving a need for comprehensive data on children’s healthcare patterns globally. What is New: • Investigating over 250,000 children, this study reveals higher healthcare utilization among Jewish and urban children across all dimensions. • Despite Israel’s national health insurance law, the study underscores the significant population-based disparities in healthcare utilization.
Vaccine hesitancy in the vaccination of children in Brazil
Since 2016, there has been a decrease in vaccination coverage for the childhood schedule of the National Immunization Program (PNI) in Brazil. To identify the reasons for vaccine hesitancy, we conducted a household survey of 31,001 live-born children living in the 26 Brazilian state capitals and the Federal District in 2017 and 2018. Census tracts were stratified according to socioeconomic status, we interviewed parents and/or guardians to collect information on the mother, child, reasons for not vaccinating, and other data. To identify the determinants of vaccine hesitancy, were used the “5Cs” (confidence, complacency and convenience, communication and context). Confidence: 94.5 % of parents/guardians of children trust the vaccines; 98.4 % believe it is important to vaccinate their children, and 20,0 % believe that vaccines are associated with serious adverse events. Complacency: 96.7 % decided to vaccinate their children with all the vaccines recommended; 7.2 % experienced difficulties with vaccination (distance and opening hours of the vaccination service, lack of time, problems with the child's health) despite going to the vaccination appointment; 22.9 % did not vaccinate their children (vaccine unavailable, clinic closed, health care provider absent, fear of adverse events). Communication: 24.5 % decided not to take their child for vaccination because of the pandemic; 24.4 % feared a reaction to the vaccine, and 9.2 % did not have the vaccine recommended by their doctor or health care provider. Socioeconomic status: vaccine confidence was lower among mothers with low education levels and lower-income families. Conclusions: Operational difficulties in the health care network were the main reasons for vaccine hesitancy in Brazilian state capitals during the study period. Although the proportion of parents/guardians who do not vaccinate their children voluntarily is low, this is a problem that needs to be consider so that it does not increase over time due to the infodemic.
Probabilistic coverage of the frontal aslant tract in young adults: Insights into individual variability, lateralization, and language functions
The frontal aslant tract (FAT) is a crucial neural pathway of language and speech, but little is known about its connectivity and segmentation differences across populations. In this study, we investigate the probabilistic coverage of the FAT in a large sample of 1065 young adults. Our primary goal was to reveal individual variability and lateralization of FAT and its structure–function correlations in language processing. The study utilized diffusion MRI data from 1065 subjects obtained from the Human Connectome Project. Automated tractography using DSI Studio software was employed to map white matter bundles, and the results were examined to study the population variation of the FAT. Additionally, anatomical dissections were performed to validate the fiber tracking results. The tract‐to‐region connectome, based on Human Connectome Project‐MMP parcellations, was utilized to provide population probability of the tract‐to‐region connections. Our results showed that the left anterior FAT exhibited the most substantial individual differences, particularly in the superior and middle frontal gyrus, with greater variability in the superior than the inferior region. Furthermore, we found left lateralization in FAT, with a greater difference in coverage in the inferior and posterior portions. Additionally, our analysis revealed a significant positive correlation between the left FAT inferior coverage area and the performance on the oral reading recognition (p = .016) and picture vocabulary (p = .0026) tests. In comparison, fractional anisotropy of the right FAT exhibited marginal significance in its correlation (p = .056) with Picture Vocabulary Test. Our findings, combined with the connectivity patterns of the FAT, allowed us to segment its structure into anterior and posterior segments. We found significant variability in FAT coverage among individuals, with left lateralization observed in both macroscopic shape measures and microscopic diffusion metrics. Our findings also suggested a potential link between the size of the left FAT's inferior coverage area and language function tests. These results enhance our understanding of the FAT's role in brain connectivity and its potential implications for language and executive functions. The study revealed individual differences in the left anterior frontal aslant tract (FAT), especially in the superior region, with significant left lateralization. Functional analysis reveals a correlation between the left inferior coverage area and language function. Overall, it provides valuable insights into FAT connectivity and its role in language processing.
Nut-cracking success and efficiency in two wild capuchin monkey populations
Capuchins can employ several strategies to deal with environmental challenges, such as using stone tools to access encapsulated resources. Nut-cracking is customary in several capuchin populations and can be affected by ecological and cultural factors; however, data on success and efficiency are only known for two wild populations. In this work, using camera traps, we assessed palm nut-cracking success and efficiency in two newly studied wild bearded capuchin populations ( Sapajus libidinosus ) and compared them with other sites. We tested the hypothesis that the overall success and efficiency of nut-cracking would be similar between sites when processing similar resources, finding partial support for it. Although using hammerstones of different sizes, capuchins had a similar success frequency. However, efficiency (number of strikes to crack a nut) was different, with one population being more efficient. We also tested whether success and efficiency varied between sexes in adults. We predict adult males would be more successful and efficient when cracking hard nuts. We found no differences between the sexes in one site but found sex differences in the other, although also for the low-resistant nut, which was unexpected. Our data add to the knowledge of capuchin nut-cracking behaviour flexibility, variance and potential cultural traits.
Mutation analysis of 28 autosomal short tandem repeats in the Chinese Han population
Short tandem repeats (STRs) have been extensively used in forensic genetics. However, according to previous studies, the mutation rates of STRs are relatively high and are affected by many factors. Therefore, it is important to analyze STR mutations and determine the influence of underlying factors on STR mutation rates. Mutation rates of 28 autosomal STRs were determined from 8708 paternity testing cases in the Chinese Han population, and the relationships between STR mutation rates and population, sex, age, allele length and heterozygosity were investigated. A total of 279 mutations were observed at 27 loci in a total of 233,530 meiosis cases, including 273 (97.8%) one-step, 5 (1.8%) two-step and 1 (0.4%) three-step mutations. The overall average mutation rate was 1.19 × 10–3 (95% CI 1.06 × 10–3 − 1.34 × 10–3) ranging from 0 (TPOX) to 2.79 × 10–3 (D13S325). Mutation rate comparisons revealed statistically significant differences at several STRs among populations. Paternal mutations occurred more frequently than maternal mutations, at a ratio of 6.04:1, and the mutation rate tended to increase with paternal age. Moreover, our study revealed a bias towards contraction mutations for long alleles and expansion mutations for short alleles. No obvious bias was observed in the overall mutation direction. In addition, STR loci with higher expected heterozygosity (Hexp) tended to have higher mutation rates. This work revealed the relationships between STR mutation rates and several influencing factors, providing useful data and information for further research on STR mutations in forensic genetics.
Physiological responses of invasive round goby (Neogobius melanostomus) to environmental stressors across a latitudinal span
Ecosystems worldwide are struggling with increasingly high levels of human-induced stressors, impacting their ecological health at local, regional, and global scales. Among the factors affecting freshwater organisms, temperature stands out, especially given the recent escalation of global warming, with possible adverse effects. In this study, we examined the basal levels of oxidative stress parameters in the round goby Neogobius melanostomus , spanning its native and non-native ranges of occurrence. We aimed to assess five populations across transcontinental scale (Turkey, Croatia, Slovakia, Poland, Finland) to determine whether individuals already experiencing conditions other than their thermal optimum (such as those in Turkey and Finland) would display elevated non-enzymatic indicators (level of lipid peroxidation and reduced glutathione levels) and enzymatic indicators (activity of glutathione peroxidase and catalase activity) of oxidative stress compared to those inhabiting milder ecosystems. The results obtained did not align with our initial predictions. The native Turkish population exhibited the lowest values for both antioxidants and oxidative damage, indicating an efficient redox system in this species. All tested parameters showed variation among populations, as well as between tissues and sexes. Temperature did not emerge as significant factor in any of the parameters tested, while for glutathione peroxidase, our analysis indicated a positive relationship with enzyme activity and salinity. The lack of a continuous latitudinal pattern in the non-native populations suggests the species’ potential for adaptive shifts, indicating superior adaptive abilities, especially in human affected ecosystems, rather than strictly adhering to environmental gradients.
Rest-activity profiles among U.S. adults in a nationally representative sample: a functional principal component analysis
Background The 24-h rest and activity behaviors (i.e., physical activity, sedentary behaviors and sleep) are fundamental human behaviors essential to health and well-being. Functional principal component analysis (fPCA) is a flexible approach for characterizing rest-activity rhythms and does not rely on a priori assumptions about the activity shape. The objective of our study is to apply fPCA to a nationally representative sample of American adults to characterize variations in the 24-h rest-activity pattern, determine how the pattern differs according to demographic, socioeconomic and work characteristics, and examine its associations with general health status. Methods The current analysis used data from adults 25 or older in the National Health and Nutrition Examination Survey (NHANES, 2011–2014). Using 7-day 24-h actigraphy recordings, we applied fPCA to derive profiles for overall, weekday and weekend rest-activity patterns. We examined the association between each rest-activity profile in relation to age, gender, race/ethnicity, education, income and working status using multiple linear regression. We also used multiple logistic regression to determine the relationship between each rest-activity profile and the likelihood of reporting poor or fair health. Results We identified four distinct profiles (i.e., high amplitude, early rise, prolonged activity window, biphasic pattern) that together accounted for 86.8% of total variation in the study sample. We identified numerous associations between each rest-activity profile and multiple sociodemographic characteristics. We also found evidence suggesting the associations differed between weekdays and weekends. Finally, we reported that the rest-activity profiles were associated with self-rated health. Conclusions Our study provided evidence suggesting that rest-activity patterns in human populations are shaped by multiple demographic, socioeconomic and work factors, and are correlated with health status.
The Influence of Environmental Factors on Germination of Burcucumber (Sicyos angulatus) Seeds: Implications for Range Expansion and Management
Burcucumber (Sicyos angulatus L.) is a highly invasive vine that has become naturalized in the humid Black Sea region of Turkey, but previous to this study there was no information on the germination biology of this weed. The germination biology of three naturalized populations of S. angulatus from the Black Sea region was studied in laboratory and greenhouse experiments. The germination of nondormant seeds was observed under different photoperiods, constant temperatures, and varying levels of pH, salinity, and osmotic potential. Furthermore, seedling emergence from various soil depths was investigated. The seeds of all populations proved nonphotoblastic (82% to 90%, 55% to 66%, and 48% to 56% germination under 12-, 24-, and 0-h photoperiod, respectively) and germinated over a wide range of temperatures (5 to 40 C). Seedlings emerged from all seed burial depths (0 to 15 cm); however, germination was drastically reduced (>90% reduction) beyond 6 to 8 cm. Seed germination was 30% to 38% for seeds placed on the surface and increased to 78% to 88% for seeds buried 2 cm deep, while germination was severely reduced for seeds buried 10 and 15 cm deep (8 to 12% and 0 to 6%, respectively). Seeds of all populations were found to be sensitive to osmotic and salinity stress and to highly acidic and alkaline pH levels. All populations had similar requirements for germination that are directly related to their area of naturalization. These results indicate that the species has limited potential for range expansion to arid, semiarid (nonirrigated), and relatively saline areas. However, humid, salinity-free, and frequently irrigated areas of the country are at high risk of invasion. In arable lands, deep tillage followed by shallow tillage and effective management of emerging seedlings could deplete the soil seedbank in the long run. However, there is an urgent need for effective strategies to manage the species in other nonarable areas.
