Synergistic Inhibiting Effect of Phytochemicals in IRheum palmatum/I on Tyrosinase Based on Metabolomics and Isobologram Analyses
Tyrosinase (TYR) plays a key role in the enzymatic reaction that is responsible for a range of unwanted discoloration effects, such as food browning and skin hyperpigmentation. TYR inhibitors could, therefore, be candidates for skin care products that aim to repair pigmentation problems. In this study, we used a metabolomics approach combined with the isobologram analysis to identify anti-TYR compounds within natural resources, and evaluate their possible synergism with each other. Rheum palmatum was determined to be a model plant for observing the effect, of which seven extracts with diverse phytochemicals were prepared by way of pressurized solvent extraction. Each Rheum palmatum extract (RPE) was profiled using nuclear magnetic resonance spectroscopy and its activity of tyrosinase inhibition was evaluated. According to the orthogonal partial least square analysis used to correlate phytochemicals in RPE with the corresponding activity, the goodness of fit of the model (R[sub.2] = 0.838) and its predictive ability (Q[sub.2] = 0.711) were high. Gallic acid and catechin were identified as the active compounds most relevant to the anti-TYR effect of RPE. Subsequently, the activity of gallic acid and catechin were evaluated individually, and when combined in various ratios by using isobologram analysis. The results showed that gallic acid and catechin in the molar ratios of 9:5 and 9:1 exhibited a synergistic inhibition on TYR, with a combination index lower than 0.77, suggesting that certain combinations of these compounds may prove effective for use in cosmetic, pharmaceutical, and food industries.

A Review on Rhubarb-Derived Substances as Modulators of Cardiovascular Risk Factors—A Special Emphasis on Anti-Obesity Action
The currently available anti-obesity therapies encounter many associated risks and side effects often causing the ineffectiveness of treatment. Therefore, various plant-derived substances have been extensively studied as a promising support or even an alternative for existing anti-obesity therapies. This review is dealing with the anti-obesity potential of edible and ethnomedicinal rhubarb species and emerging possible role of the rhubarb-derived extracts or individual compounds in the prevention of obesity and perspectives for their use in an anti-obesity treatment. A special emphasis is put on the most popular edible specimens, i.e., Rheum rhabarbarum L. (garden rhubarb) and Rheum rhaponticum L. (rhapontic rhubarb, Siberian rhubarb); however, the anti-obesity potential of other rhubarb species (e.g., R. officinale, R. palmatum, and R. emodi) is presented as well. The significance of rhubarb-derived extracts and low-molecular specialized rhubarb metabolites of diversified chemical background, e.g., anthraquinones and stilbenes, as potential modulators of human metabolism is highlighted, including the context of cardiovascular disease prevention. The available reports present multiple encouraging rhubarb properties starting from the anti-lipidemic action of rhubarb fibre or its use as purgative medicines, through various actions of rhubarb-derived extracts and their individual compounds: inhibition of enzymes of cholesterol and lipid metabolism, targeting of key molecular regulators of adipogenesis, regulators of cell energy metabolism, the ability to inhibit pro-inflammatory signalling pathways and to regulate glucose and lipid homeostasis contributing to overall in vivo and clinical anti-obesity effects.
Impacts of climate change on the distribution pattern of Himalayan Rhubarb
While there has been substantial consensus that climate change poses a severe threat to the Himalayan biota; we still lack comprehensive data on the potential impact of climate change on the important Himalayan medicinal plants connected with the livelihood of local people and the national economy. In this study, using MaxEnt, we modelled the distribution pattern of Rheum australe, a medicinal plant prioritized by the Government of Nepal for the nation's economic prosperity, for the current as well as all four future projections (SSP126, SSP245, SSP370, and SSP585) to all the data available periods (2021-2040, 2041-2060, 2061-2080, and 2081-2100). In addition, we performed spatial overlay analysis to identify the optimum zones that could be developed as potential planting/conservation sites for R. australe in any of the projected future climate scenarios. Our results revealed that the suitability area of R. australe is expected to contract in all the scenarios and periods with a significant loss expected to occur in SSP585 for 2081-2100. On the individual district level, northwestern districts are expected to have a huge loss of suitable habitats in the future, while four districts: Jumla, Kalikot, Dolpa, and Jajarkot of the Karnali Province are expected to gain suitable habitats remarkably. Therefore, we suggest that the forests and rangelands of the four districts at an elevation range of 3300 m - 4,400 m could be developed as potential planting areas for commercial cultivation/for establishing genetic resource conservation centres. This finding thus has wider policy implications for both government and conservation organizations.
Influence of Resveratrol on the Immune Response
Resveratrol is the most well-known polyphenolic stilbenoid, present in grapes, mulberries, peanuts, rhubarb, and in several other plants. Resveratrol can play a beneficial role in the prevention and in the progression of chronic diseases related to inflammation such as diabetes, obesity, cardiovascular diseases, neurodegeneration, and cancers among other conditions. Moreover, resveratrol regulates immunity by interfering with immune cell regulation, proinflammatory cytokines’ synthesis, and gene expression. At the molecular level, it targets sirtuin, adenosine monophosphate kinase, nuclear factor-κB, inflammatory cytokines, anti-oxidant enzymes along with cellular processes such as gluconeogenesis, lipid metabolism, mitochondrial biogenesis, angiogenesis, and apoptosis. Resveratrol can suppress the toll-like receptor (TLR) and pro-inflammatory genes’ expression. The antioxidant activity of resveratrol and the ability to inhibit enzymes involved in the production of eicosanoids contribute to its anti-inflammation properties. The effects of this biologically active compound on the immune system are associated with widespread health benefits for different autoimmune and chronic inflammatory diseases. This review offers a systematic understanding of how resveratrol targets multiple inflammatory components and exerts immune-regulatory effects on immune cells.

Qualitative and quantitative analysis methods for quality control of rhubarb in Taiwan’s markets
Introduction: Rhubarb is a traditional Chinese medicine (TCM) used to release heat and has cathartic effects. Official rhubarb in Taiwan Herbal Pharmacopeias 4th edition (THP 4th) and China Pharmacopeia 2020 (CP 2020) are the roots and rhizomes of Rheum palmatum L., Rheum tanguticum Maxim. ex Balf., and Rheum officinale Baill. However, the Rheum genus is a large genus with many different species, and owing to the similarity in appearance and taste with official rhubarb, there needs to be more clarity in the distinction between the species of rhubarb and their applications. Given the time-consuming and complicated extraction and chromatography methods outlined in pharmacopeias, we improved the qualitative analysis and quantitative analysis methods for rhubarb in the market. Hence, we applied our method to identify the species and quality of official and unofficial rhubarb. Method: We analyzed 21 rhubarb samples from the Taiwanese market using a proposed HPLC-based extraction and qualitative analysis employing eight markers: aloe-emodin, rhein, emodin, chrysophanol, physcion, rhapontigenin, rhaponticin, and resveratrol. Additionally, we developed a TLC method for the analysis of rhubarb. KEGG pathway analysis was used to clarify the phytochemical and pharmacological knowledge of official and unofficial rhubarb. Results: Rhein and rhapontigenin emerged as key markers to differentiate official and unofficial rhubarb. Rhapontigenin is abundant in unofficial rhubarb; however, rhein content was low. In contrast, their contents in official rhubarb were opposite to their contents in unofficial rhubarb. The TLC analysis used rhein and rhapontigenin to identify rhubarb in Taiwan’s markets, whereas the KEGG pathway analysis revealed that anthraquinones and stilbenes affected different pathways. Discussion: Eight reference standards were used in this study to propose a quality control method for rhubarb in Taiwanese markets. We propose a rapid extraction method and quantitative analysis of rhubarb to differentiate between official and unofficial rhubarb.
Advances in bio-active constituents, pharmacology and clinical applications of rhubarb
Rhubarb is one of the most ancient, commonly used and important herbs in Chinese medicine. The modern researches of rhubarb clarified the efficacies, ingredients and mechanisms in a more scientific and rigorous way. The main chemical compositions of rhubarb include anthraquinones, anthrones, stilbenes, tannins, polysaccharides etc. These compositions show extensive pharmacological activities including regulating gastrointestinal, anticancer, antimicrobial, hepatoprotective, anti-inflammatory, protecting cardiovascular, cerebrovascular and so on. This paper reviews the recent studies on the active ingredients, pharmacological effects, clinical application and functional mechanism.
A Synthesis of the Best Evidence for Rhubarb Administration in the Treatment of Acute Pancreatitis in China
Acute pancreatitis has complex causes, and traditional Chinese medicine plays a significant role in its treatment. However, there is a lack of standardized protocols and a comprehensive best evidence summary for rhubarb administration in acute pancreatitis, leading to inconsistent clinical practice. This study aimed to retrieve and summarize the best evidence on the administration of rhubarb in the treatment of acute pancreatitis and to provide a theoretical reference for the development of clinical practice. A systematic search was performed from database inception to 31 October, 2025 across multiple electronic databases and guideline repositories, including the UK National Institute for Health and Care Excellence, the US Agency for Healthcare Research and Quality, Cochrane Library, PubMed, EMbase, China biomedical, CNKI, Wanfang, and VIP. Eligible evidence comprised clinical practice guidelines, expert consensus statements, and systematic reviews focusing on rhubarb administration in adult patients with acute pancreatitis. The methodological quality of included studies was independently evaluated using the AGREE II, JBI, and AMSTAR 2.0 tools. A total of 15 documents were included, including 3 clinical guidelines, 3 expert consensus, and 9 systematic evaluations. Twenty-seven pieces of evidence related to rhubarb administration in patients with acute pancreatitis were extracted and summarized into four themes: mechanism of action, feasibility, evaluation, and clinical implementation. Healthcare providers may apply the best evidence for rhubarb administration in acute pancreatitis to improve intestinal function recovery. However, recommendations should be interpreted cautiously, as all evidence comes from Chinese studies with limited generalizability and some low-level findings. This study is in accordance with the evidence summary reporting specifications of the Fudan University Center for the Evidence-based Nursing, and the registration number is \"ES20246947\".
COMPARISON OF QUALITY PARAMETERS OF EDIBLE FILMS WITH RHUBARB ADDITION PRODUCED BY TWO DIFFERENT METHODS
In edible films with added plant extracts, the effects of the polymer matrix used and the production method on film performance have not yet been sufficiently clarified. Therefore, in this study, the quality parameters of starch-gelatin and chitosan-based edible films with added rhubarb (Rheum ribes L.), produced by two different methods, were comparatively examined. The films were characterized in terms of thickness, water solubility, mechanical properties, water vapor permeability, optical properties, color parameters, antioxidant capacity, biodegradability, and structural properties. The results showed that the film properties changed significantly depending on both the polymer matrix used and the rhubarb extract concentration. Starch-gelatin-based films exhibited higher water solubility and flexibility, while chitosan-based films showed higher mechanical strength and better water vapor barrier properties. Rhubarb addition significantly increased the antioxidant potential of the film-forming mixtures in both methods. Color analyses revealed that the films darkened with increasing rhubarb concentration. SEM and FT-IR analyses showed that rhubarb addition caused physical changes in the film structure but did not alter the chemical structure. Chitosan-based films with rhubarb addition may be considered as a potential alternative for active and sustainable food packaging applications.
