Gamma-aminobutyric acid (GABA) improves salinity stress tolerance in soybean seedlings by modulating their mineral nutrition, osmolyte contents, and ascorbate-glutathione cycle

Background
In plants, GABA plays a critical role in regulating salinity stress tolerance. However, the response of soybean seedlings (
Glycine max
L.) to exogenous gamma-aminobutyric acid (GABA) under saline stress conditions has not been fully elucidated.
Results
This study investigated the effects of exogenous GABA (2 mM) on plant biomass and the physiological mechanism through which soybean plants are affected by saline stress conditions (0, 40, and 80 mM of NaCl and Na
2
SO
4
at a 1:1 molar ratio). We noticed that increased salinity stress negatively impacted the growth and metabolism of soybean seedlings, compared to control. The root-stem-leaf biomass (27- and 33%, 20- and 58%, and 25- and 59% under 40- and 80 mM stress, respectively]) and the concentration of chlorophyll a and chlorophyll b significantly decreased. Moreover, the carotenoid content increased significantly (by 35%) following treatment with 40 mM stress. The results exhibited significant increase in the concentration of hydrogen peroxide (H

), malondialdehyde (MDA), dehydroascorbic acid (DHA) oxidized glutathione (GSSG), Na

The positive impact of exogenously applied GABA on soybean plants could be attributed to its ability to improve their physiological stress response mechanisms and reduce harmful substances.
Conclusion
Applying GABA to soybean plants could be an effective strategy for mitigating salinity stress. In the future, molecular studies may contribute to a better understanding of the mechanisms by which GABA regulates salt tolerance in soybeans.

Background
Salinity is one of the most challenging abiotic stresses restricting the growth of plants.
In vitro
screening will increase the efficiency and speed of salinity tolerant genotypes identifications. The response of four tomato cultivars under salinity was analyzed
in vitro
to evaluate the seedlings growth, biochemical, and gene expression responses as well as the effect of nano zinc and iron on callus induction and plant regeneration.
Results
The results showed that an increase in salinity stress in the medium decreased the germination percentage, fresh and dry weight of shoot, root length, chlorophyll a, b and carotenoids content, K and Ca content, and on the other hand, Na content was increased. MDA content (‘Nora’, ‘PS-10’, ‘Peto’ and ‘Roma’: 1.71, 1.78, 1.66 and 2.16 folds, respectively), electrolyte leakage (‘PS-10’: 33.33%; ‘Roma’: 56.33%), were increased with salinity of 100 mM compared to control. Proline content was increased in 50 mM NaCl (10.8 fold). The most activity of antioxidant enzymes including CAT, SOD, APX, GPX, and GR was observed in the ‘PS-10’ cultivar, and the lowest activity of these enzymes was observed in ‘Roma’ under salinity stress. The AsA and GSH were decreased and DHA and GSSG were increased with the increased intensity of salinity. The relative expression of
SOD

APX
, and
GR
genes varied in different cultivars at different salinity concentrations. The most percentage of callus induction was observed with applying iron oxide nanoparticles, and the most regeneration rate was recorded using zinc oxide nanoparticles.
Conclusion
The results showed that salt-tolerant cultivars such as ‘PS-10’ with better osmotic adjustment, are suitable candidates for the future production and breeding programs. The use of nutrient nanoparticles under salinity stress for different tomato cultivars increased their performance.

Assessing the potential of exogenous caffeic acid application in boosting wheat (Triticum aestivum L.) crop productivity under salt stress

Caffeic acid (CA) is known as an antioxidant to scavenge reactive oxygen species (ROS), but the underlying mechanism of mediation of plant salt tolerance against various abiotic stresses by caffeic acid is only partially understood. A field experiment (120 days duration) was conducted to investigate the protective role of caffeic acid under a high saline medium (EC 8.7 dS m -1 and textural class: sandy loam) in two wheat genotypes (FSD -08 and Zincol-16). Two levels of caffeic acid (50 μM and 100 μM) were applied exogenously in combination with the salinity stress and results revealed that salt alleviation is more prominent when caffeic acid was applied at the rate of 100 μM. Under saline conditions, wheat genotypes show poor fresh and dry matter accumulation, chlorophyll contents, relative water contents (RWC), membrane stability index (MSI) and activities of antioxidant enzymes and increased uptake of Na + ions. However, wheat genotype FSD-08 eminently responded to caffeic acid application as compared to wheat genotype Zincol-16 as demonstrated by higher growth indicators, RWC, MSI, activities of antioxidant enzymes, accumulation of mineral ions in grain along with yield attributes. In addition, caffeic acid also mitigated salt-induced oxidative stress malondialdehyde (MDA) and hydrogen peroxide (H 2 O 2 ) contents as well as significantly reduced Na + uptake. It can be concluded that caffeic acid-induced salinity tolerance in wheat is attributed to improved plant water relations, K + uptake, yield contents and activities of antioxidant stress enzymes.

MeJA-mediated enhancement of salt-tolerance of Populus wutunensis by 5-aminolevulinic acid

Results
Physiological results showed that 50 mg·L

ALA-treatment significantly reduced the malondialdehyde (MDA) content and the relative electrical conductivity (REC) and enhanced antioxidant activities of enzymes such as SOD, POD and CAT in salt-stressed
P. wutunensis
seedlings. Transcriptome analysis identified ALA-induced differentially expressed genes (DEGs) associating with increased salt-tolerance in
P. wutunensis
. GO and KEGG enrichment analyses showed that ALA activated the jasmonic acid signaling and significantly enhanced the protein processing in endoplasmic reticulum and the flavonoid biosynthesis pathways. Results of the hormone-quantification by LC-MS/MS-based assays showed that ALA could increase the accumulation of methyl jasmonate (MeJA) in salt-stressed
P. wutunensis
. Induced contents of soluble proteins and flavonoids by exogenous ALA in salt-treated seedlings were also correlated with the MeJA content.
Conclusion
5-aminolevulinic acid improved the protein-folding efficiency in the endoplasmic reticulum and the flavonoid-accumulation through the MeJA-activated jasmonic acid signaling, thereby increased salt-tolerance in

Although long-term high dietary sodium consumption often aggravates hypertension and bone loss, sodium in the intestinal lumen has been known to promote absorption of nutrients and other ions, e.g., glucose and calcium. However, whether high-salt diet (HSD) altered mucosal morphology, villous cell turnover and calcium transporter expression remained elusive. Herein, rats were treated with HSD containing 8% wt/wt NaCl for up to 5 months. HSD rats exhibited a marked increase in sodium intake with high fecal and urinary sodium excretion, as compared to the control group treated with normal diet. Intestinal histomorphometry revealed increasing of crypt depth and villous height in 3- and 4-month HSD groups, respectively, consistent with larger mucosal-to-serosal amplification ratio that reflected an increased surface area for nutrient absorption. The signals of Ki-67-positive cells was enhanced in the crypts as visualized by multiphoton fluorescence microscopy, whereas the TUNEL-positive cells were decreased in the villi of HSD, suggesting greater crypt cell proliferation and a reduction of villous cell apoptosis. Confocal microscopy showed higher expression of TRPV6 protein in the villous tip of HSD, while PMCA.sub.1 expression was increased in villous tip and crypt areas. The percentage of cells with highly expressed uroguanylin-an endogenous intestinal natriuretic peptide-was significantly higher in HSD group. In conclusion, HSD profoundly changed the intestinal morphology and turnover of epithelial cell, increased the expression of calcium transporters and uroguanylin. Our findings reflect pathophysiological adaptations in the intestine, which might be another target organ for drug discovery against HSD-induced osteopathy in the future.

In this study, we characterized, identified, and determined the effect of salt-tolerant PGPR isolated from coastal saline areas on rice growth and yield. A total of 44 bacterial strains were isolated, and 5 were found to be tolerant at high salt concentration. These isolates were further characterized for salinity tolerance and beneficial traits through a series of quantitative tests. Biochemical characterization showed that bacterial survivability decreases gradually with the increase of salt concentration. One of the strains, UPMRB9, produced the highest amount of exopolysaccharides when exposed to 1.5M of NaCl. Moreover, UPMRB9 absorbed the highest amount of sodium from the 1.5M of NaCl-amended media. The highest floc yield and biofilm were produced by UPMRE6 and UPMRB9 respectively, at 1M of NaCl concentration. The SEM observation confirmed the EPS production of UPMRB9 and UPMRE6 at 1.5M of NaCl concentration. These two isolates were identified as Bacillus tequilensis and Bacillus aryabhattai based on the 16S rRNA gene sequence. The functional group characterization of EPS showed the presence of hydroxyl, carboxyl, and amino groups. This corresponded to the presence of carbohydrates and proteins in the EPS and glucose was identified as the major type of carbohydrate. The functional groups of EPS can help to bind and chelate Na.sup.+ in the soil and thereby reduces the plant's exposure to the ion under saline conditions. The plant inoculation study revealed significant beneficial effects of bacterial inoculation on photosynthesis, transpiration, and stomatal conductance of the plant which leads to a higher yield. The Bacillus tequilensis and Bacillus aryabhattai strains showed good potential as PGPR for salinity mitigation practice for coastal rice cultivation.

Comparative efficacy of bio-selenium nanoparticles and sodium selenite on morpho-physiochemical attributes under normal and salt stress conditions, besides selenium detoxification pathways in Brassica napus L

Selenium nanoparticles (SeNPs) have attracted considerable attention globally due to their significant potential for alleviating abiotic stresses in plants. Accordingly, further research has been conducted to develop nanoparticles using chemical ways. However, our knowledge about the potential benefit or phytotoxicity of bioSeNPs in rapeseed is still unclear. Herein, we investigated the effect of bioSeNPs on growth and physiochemical attributes, and selenium detoxification pathways compared to sodium selenite (Se (IV)) during the early seedling stage under normal and salt stress conditions. Our findings showed that the range between optimal and toxic levels of bioSeNPs was wider than Se (IV), which increased the plant’s ability to reduce salinity-induced oxidative stress. BioSeNPs improved the phenotypic characteristics of rapeseed seedlings without the sign of toxicity, markedly elevated germination, growth, photosynthetic efficiency and osmolyte accumulation versus Se (IV) under normal and salt stress conditions. In addition to modulation of Na
+
and K
+
uptake, bioSeNPs minimized the ROS level and MDA content by activating the antioxidant enzymes engaged in ROS detoxification by regulating these enzyme-related genes expression patterns. Importantly, the main effect of bioSeNPs and Se (IV) on plant growth appeared to be correlated with the change in the expression levels of Se-related genes. Our qRT-PCR results revealed that the genes involved in Se detoxification in root tissue were upregulated upon Se (IV) treated seedlings compared to NPs, indicating that bioSeNPs have a slightly toxic effect under higher concentrations. Furthermore, bioSeNPs might improve lateral root production by increasing the expression level of
LBD16
. Taken together, transamination and selenation were more functional methods of Se detoxification and proposed different degradation pathways that synthesized malformed or deformed selenoproteins, which provided essential mechanisms to increase Se tolerance at higher concentrations in rapeseed seedlings. Current findings could add more knowledge regarding the mechanisms underlying bioSeNPs induced plant growth.

Salinity is major abiotic stress affecting crop yield, productivity and reduces the land-usage area for agricultural practices. The purpose of this study is to analyze the effect of green-synthesized silver nanoparticle (AgNP) on physiological traits of wheat (Triticum aestivum) under salinity stress. Using augmented and high-throughput characterization of synthesized AgNPs, this study investigated the proximity of AgNPs-induced coping effects under stressful cues by measuring the germination efficiency, oxidative-biomarkers, enzymatic and non-enzymatic antioxidants, proline and nitrogen metabolism, stomatal dynamics, and ABA content. Taken together, the study shows a promising approach in salt tolerance and suggests that mechanisms of inducing the salt tolerance depend on proline metabolism, ions accumulation, and defense mechanisms. This study ascertains the queries regarding the correlation between nanoparticles use and traditional agriculture methodology; also significantly facilitates to reach the goal of sustainable developments for increasing crop productivity via much safer and greener approachability.
