The unprecedented 2015/16 Tasman Sea marine heatwave

The Tasman Sea off southeast Australia exhibited its longest and most intense marine heatwave ever recorded in 2015/16. Here we report on several inter-related aspects of this event: observed characteristics, physical drivers, ecological impacts and the role of climate change. This marine heatwave lasted for 251 days reaching a maximum intensity of 2.9 °C above climatology. The anomalous warming is dominated by anomalous convergence of heat linked to the southward flowing East Australian Current. Ecosystem impacts range from new disease outbreaks in farmed shellfish, mortality of wild molluscs and out-of-range species observations. Global climate models indicate it is very likely to be that the occurrence of an extreme warming event of this duration or intensity in this region is respectively ≥330 times and ≥6.8 times as likely to be due to the influence of anthropogenic climate change. Climate projections indicate that event likelihoods will increase in the future, due to increasing anthropogenic influences.

. link this event to warm, southwards flowing waters from East Australia and find that climate change has made these events almost seven times more likely.

Aquaculture is remarkably one of the most promising industries among the food-producing industries in the world. Aquaculture production as well as fish consumption per capita have been dramatically increasing over the past two decades. Shifting of culture method from semi-intensive to intensive technique and applying of antibiotics to control the disease outbreak are the major factors for the increasing trend of aquaculture production. Antibiotics are usually present at subtherapeutic levels in the aquaculture environment, which increases the selective pressure to the resistant bacteria and stimulates resistant gene transfer in the aquatic environment. It is now widely documented that antibiotic resistance genes and resistant bacteria are transported from the aquatic environment to the terrestrial environment and may pose adverse effects on human and animal health. However, data related to antibiotic usage and bacterial resistance in aquaculture is very limited or even absent in major aquaculture-producing countries. In particular, residual levels of antibiotics in fish and shellfish are not well documented. Recently, some of the countries have already decided the maximum residue levels (MRLs) of antibiotics in fish muscle or skin; however, many antibiotics are yet not to be decided. Therefore, an urgent universal effort needs to be taken to monitor antibiotic concentration and resistant bacteria particularly multiple antibiotic-resistant bacteria and to assess the associated risks in aquaculture. Finally, we suggest to take an initiative to make a uniform antibiotic registration process, to establish the MRLs for fish/shrimp and to ensure the use of only aquaculture antibiotics in fish and shellfish farming globally.

Characterization of a new member of Iridoviridae, Shrimp hemocyte iridescent virus (SHIV), found in white leg shrimp (Litopenaeus vannamei)

A newly discovered iridescent virus that causes severe disease and high mortality in farmed
Litopenaeus vannamei
in Zhejiang, China, has been verified and temporarily specified as shrimp hemocyte iridescent virus (SHIV). Histopathological examination revealed basophilic inclusions and pyknosis in hematopoietic tissue and hemocytes in gills, hepatopancreas, periopods and muscle. Using viral metagenomics sequencing, we obtained partial sequences annotated as potential iridoviridae. Phylogenetic analyses using amino acid sequences of major capsid protein (MCP) and ATPase revealed that it is a new iridescent virus but does not belong to the five known genera of
Iridoviridae
. Transmission electron microscopy showed that the virus exhibited a typical icosahedral structure with a mean diameter of 158.6 ± 12.5 nm (n = 30)(v-v) and 143.6 ± 10.8 nm (n = 30)(f-f), and an 85.8 ± 6.0 nm (n = 30) nucleoid. Challenge tests of
L. vannamei
via intermuscular injection, per os and reverse gavage all exhibited 100% cumulative mortality rates. The
in situ
hybridization showed that hemopoietic tissue, gills, and hepatopancreatic sinus were the positively reacting tissues. Additionally, a specific nested PCR assay was developed. PCR results revealed that
L. vannamei
,
Fenneropenaeus chinensis
, and
Macrobrachium rosenbergii
were SHIV-positive, indicating a new threat existing in the shrimp farming industry in China.

Comparative 16SrDNA Gene-Based Microbiota Profiles of the Pacific Oyster (Crassostrea gigas) and the Mediterranean Mussel (Mytilus galloprovincialis) from a Shellfish Farm (Ligurian Sea, Italy)

The pacific oyster Crassostrea gigas and the Mediterranean mussel Mytilus galloprovincialis are two widely farmed bivalve species which show contrasting behaviour in relation to microbial diseases, with C. gigas being more susceptible and M. galloprovincialis being generally resistant. In a recent study, we showed that different susceptibility to infection exhibited by these two bivalve species may depend on their different capability to kill invading pathogens (e.g., Vibrio spp.) through the action of haemolymph components. Specific microbial-host interactions may also impact bivalve microbiome structure and further influence susceptibility/resistance to microbial diseases. To further investigate this concept, a comparative study of haemolymph and digestive gland 16SrDNA gene-based bacterial microbiota profiles in C. gigas and M. galloprovincialis co-cultivated at the same aquaculture site was carried out using pyrosequencing. Bacterial communities associated with bivalve tissues (hemolymph and digestive gland) were significantly different from those of seawater, and were dominated by relatively few genera such as Vibrio and Pseudoalteromonas. In general, Vibrio accounted for a larger fraction of the microbiota in C. gigas (on average 1.7-fold in the haemolymph) compared to M. galloprovincialis, suggesting that C. gigas may provide better conditions for survival for these bacteria, including potential pathogenic species such as V. aestuarianus. Vibrios appeared to be important members of C. gigas and M. galloprovincialis microbiota and might play a contrasting role in health and disease of bivalve species. Accordingly, microbiome analyses performed on bivalve specimens subjected to commercial depuration highlighted the ineffectiveness of such practice in removing Vibrio species from bivalve tissues.

Microplastic Ingestion by Wild and Cultured Manila Clams (Venerupis philippinarum) from Baynes Sound, British Columbia

Microplastics, plastic particles <5 mm, are an emerging concern in aquatic ecosystems. Because microplastics are small, they are available to many filter-feeding organisms, which can then be consumed by higher trophic level organisms, including humans. This study documents the quantity of microplastics present in wild and cultured Manila clams (
Venerupis philippinarum
). Three active shellfish farms and three reference beaches (i.e., non-shellfish farm sites) in Baynes Sound, British Columbia were chosen to examine the microplastic concentrations in wild and cultured Manila clams. Microplastics were isolated using a nitric acid digestion technique and enumerated from 54 clams (27 farmed and 27 non-farmed). Qualitative attributes, such as colour and microplastic type (fiber, fragment, or film) also were recorded. There was no significant difference (

= 0.289) between microplastic concentrations in cultured and wild clams. Microplastic concentrations ranged from 0.07 to 5.47 particles/g (from reference beach and shellfish farm clams, respectively). Fibers were the dominant microplastic (90 %); colourless and dark gray fibers were the most common colours observed (36 and 26 %, respectively). Although this indicates that microplastics are definitely present in seafood consumed by humans, shellfish aquaculture operations do not appear to be increasing microplastic concentrations in farmed clams in this region.

A Nested PCR Assay to Avoid False Positive Detection of the Microsporidian Enterocytozoon hepatopenaei (EHP) in Environmental Samples in Shrimp Farms

Hepatopancreatic microsporidiosis (HPM) caused by Enterocytozoon hepatopenaei (EHP) is an important disease of cultivated shrimp. Heavy infections may lead to retarded growth and unprofitable harvests. Existing PCR detection methods target the EHP small subunit ribosomal RNA (SSU rRNA) gene (SSU-PCR). However, we discovered that they can give false positive test results due to cross reactivity of the SSU-PCR primers with DNA from closely related microsporidia that infect other aquatic organisms. This is problematic for investigating and monitoring EHP infection pathways. To overcome this problem, a sensitive and specific nested PCR method was developed for detection of the spore wall protein (SWP) gene of EHP (SWP-PCR). The new SWP-PCR method did not produce false positive results from closely related microsporidia. The first PCR step of the SWP-PCR method was 100 times (104 plasmid copies per reaction vial) more sensitive than that of the existing SSU-PCR method (106 copies) but sensitivity was equal for both in the nested step (10 copies). Since the hepatopancreas of cultivated shrimp is not currently known to be infected with microsporidia other than EHP, the SSU-PCR methods are still valid for analyzing hepatopancreatic samples despite the lower sensitivity than the SWP-PCR method. However, due to its greater specificity and sensitivity, we recommend that the SWP-PCR method be used to screen for EHP in feces, feed and environmental samples for potential EHP carriers.

Acute Hepatopancreatic Necrosis Disease (AHPND): Virulence, Pathogenesis and Mitigation Strategies in Shrimp Aquaculture

Shrimp, as a high-protein animal food commodity, are one of the fastest growing food producing sectors in the world. It has emerged as a highly traded seafood product, currently exceeding 8 MT of high value. However, disease outbreaks, which are considered as the primary cause of production loss in shrimp farming, have moved to the forefront in recent years and brought socio-economic and environmental unsustainability to the shrimp aquaculture industry. Acute hepatopancreatic necrosis disease (AHPND), caused by Vibrio spp., is a relatively new farmed penaeid shrimp bacterial disease. The shrimp production in AHPND affected regions has dropped to ~60%, and the disease has caused a global loss of USD 43 billion to the shrimp farming industry. The conventional approaches, such as antibiotics and disinfectants, often applied for the mitigation or cure of AHPND, have had limited success. Additionally, their usage has been associated with alteration of host gut microbiota and immunity and development of antibiotic resistance in bacterial pathogens. For example, the Mexico AHPND-causing V. parahaemolyticus strain (13-306D/4 and 13-511/A1) were reported to carry tetB gene coding for tetracycline resistance gene, and V. campbellii from China was found to carry multiple antibiotic resistance genes. As a consequence, there is an urgent need to thoroughly understand the virulence mechanism of AHPND-causing Vibrio spp. and develop novel management strategies to control AHPND in shrimp aquaculture, that will be crucially important to ensure food security in the future and offer economic stability to farmers. In this review, the most important findings of AHPND are highlighted, discussed and put in perspective, and some directions for future research are presented.

Seaweed farming is expanding in Europe and may provide environmental benefits similar to those from natural kelp forests and shellfish farms, including habitat provisioning. Few studies have substantiated these claims however, and it remains uncertain whether seaweed farms will support similar biodiversity to kelp forests or provide valuable long-term habitat beyond the harvest season. We repeatedly surveyed an integrated sugar kelp (
Saccharina latissima
) and blue mussel (
Mytilus edulis
) farm in southwest UK to compare epibiont assemblages between cultivated kelps, to those from three nearby wild kelp populations, and to epibionts on farmed mussel lines and unseeded ‘bare’ lines. We found farmed kelps supported over 217 times the abundance of epibionts living on wild kelps at harvest time, however, taxonomic diversity per kelp was lower at the farm. Farmed kelp assemblages were dominated by amphipods, which were present on the wild kelps but in much lower numbers. Farmed kelp also supported distinct assemblages to cultivated mussels, which were similarly dominated by amphipods, but hosted higher relative abundances of crabs, echinoderms, worms and red algal biomass. The bare lines were heavily colonised by another pseudo-kelp,
Saccorhiza polyschides
, which supported similar epibiont assemblages to the seeded
S. latissima
lines. Our findings indicate that cultivating bivalves alongside seaweed can increase habitat provisioning at a seaweed farm and extend its permanence beyond typical seaweed cultivation periods as bivalves have longer, continuous farming periods. However, the presence of mussels will likely influence the epibiont assemblages on the farmed kelp, which are distinct from wild kelp populations.
