Retroviruses can leave a \"fossil record\" in their hosts' genomes in the form of endogenous retroviruses. Foamy viruses, complex retroviruses that infect mammals, have been notably absent from this record. We have found an endogenous foamy virus within the genomes of sloths and show that foamy viruses were infecting mammals more than 100 million years ago and codiverged with their hosts across an entire geological era. Our analysis highlights the role of evolutionary constraint in maintaining viral genome structure and indicates that accessory genes and mammalian mechanisms of innate immunity are the products of macroevolutionary conflict played out over a geological time scale.

Seroprevalence of Arboviruses and Genetic Characterization of Orbiviruses in Sloths from Western Panama

Arthropod-borne viruses (arboviruses) are an increasingly significant threat to public health in tropical regions. In this study, we investigated the seroprevalence of various arboviruses in two species of sloth (Choloepus hoffmanni and Bradypus variegatus) in rural and peri-urban areas of Western Panama province. Between 2013 and 2018, blood samples from 60 sloths were tested for neutralizing antibodies against ten arboviruses. Significant variation in seroprevalence of different arboviruses was observed: 6.7% of sloths were seropositive for Madariaga virus, 6.7% for Venezuelan equine encephalitis virus, and 4.8% for Oropouche virus, while none were seropositive for dengue type 2, Zika, chikungunya, Una, Mayaro, or Punta Toro viruses. Notably, two Changuinola virus (CGLV) strains, which were previously isolated from Panamanian sloths in the 1970s, showed high seroprevalence: Pansloth 149 (23.3%) and D50 (53.3%). Given the high seroprevalence detected in our study and the lack of genomic characterization of the historical Pansloth 149 isolate, we performed next-generation sequencing of its complete genome using Illumina technology to understand its genetic diversity and evolutionary relationship with other CGLV strains.

SARS-CoV-2 Serological Surveillance of Both Vaccinated and Unvaccinated Zoo Animals with the Identification of a Sloth Bear and a Tapir with Previous Infection

Since its discovery in 2019, SARS-CoV-2 has continued to be detected in both humans and animals worldwide. Currently there is limited research focusing on serological surveillance of wildlife under human care. Here we tested 230 serum samples of 134 animals from two zoological institutions collected between 2015 and 2024. To assess prior exposure and antibody responses from natural infection or vaccination, we used three serological assays: a nucleocapsid protein-based ELISA (N-ELISA), a surrogate virus neutralization test (sVNT) for spike (S) protein and a neutralization assay with S-pseudotyped viral particles. Among the 114 samples collected from 58 animals at Fort Wayne Zoo in Indiana, 37 samples from 20 vaccinated animals were sVNT-positive, and 2 of the positive animals had 2 samples prior to vaccination that tested positive by N-ELISA. Of the 116 samples from 76 animals at Brookfield Zoo in Illinois, 20 samples of 20 animals were sVNT-positive, and 19 of the positive animals had been vaccinated. Among these 20 sVNT-positive samples, only one sample from a South American Tapir was positive from prior to vaccination and 1 sample from a sloth bear was also positive by N-ELISA, marking the first documented cases of SARS-CoV-2 exposure in both species. Neutralization assays with S-pseudotyped virus revealed that some of the sVNT-positive samples have strong activity against the WH1-S pseudovirus but showed significantly reduced neutralization against the Omicron LP.8.1-S pseudovirus. These results underscore the need for updated vaccines tailored to emerging variants. Overall, our findings highlight the importance of continued serological surveillance across multiple species to detect new SARS-CoV-2 exposures and monitor vaccine-induced immunity in captive animal populations.

Molecular detection of piroplasmids in mammals from the Superorder Xenarthra in Brazil

Xenarthra mammals can be found from southern North America to southern South America, including all Brazilian biomes. Although it has been shown that Xenarthra mammals can play a role as reservoirs for several zoonotic agents, few studies investigate the diversity of piroplasmids (Apicomplexa: Piroplasmida) in this group of mammals. Taking into account that piroplasmids can cause disease in animals and humans, understanding the prevalence and diversity of piroplasmids in Xenarthra mammals would contribute to conservation efforts for this group of animals as well as to infer risk areas for transmission of emergent zoonosis. The present study aimed to investigate the occurrence and molecular identity of piroplasmids in free-living mammals of the Superorder Xenarthra from four Brazilian states (Mato Grosso do Sul, São Paulo, Rondônia, and Pará). For this, DNA was extracted from blood or spleen samples from 455 animals. A nested PCR based on the 18S rRNA gene was used as screening for piroplasmids. ) and
Amblyomma dubitatum
collected from opossums and coatis in Brazil. On the other hand, the 18S rRNA sequence obtained from
Dasypus novemcinctus
was closely related to a
Theileria
sp. sequence previously detected in armadillos from Mato Grosso State, grouping in a subclade within the
Theileria sensu stricto
clade. In the phylogenetic analysis based on the ITS-1 region, the sequences obtained from
Myrmecophaga tridactyla
and
Tamandua tetradactyla
were placed into a single clade, apart from the other piroplasmid clades. The present study demonstrated the molecular occurrence of Piroplasmida in anteaters and
Babesia
sp. and
Theileria
sp. in armadillos from Brazil.

For years, mammals of the order Pilosa have been considered Leishmania reservoirs. But while most studies have focused on sloth species, anteaters have been overlooked, and in many Leishmania endemic countries like Mexico, no studies have been carried out. The aims of this work were to identify the presence of Leishmania spp. in tissue samples from road-killed northern tamanduas (Tamandua mexicana), using PCR amplification and sequencing of ITS1 DNA, and to discuss the role of Pilosa mammals as reservoirs of Leishmania based on available scientific records. This is the first study that identifies Leishmania in T. mexicana, from 1 of 16 individuals analyzed, so the estimated prevalence (CI 95%) of infection was 6.3% (0.3–27.2). Amplified sequence exhibited a 98.9% (727/735) similarity with L. mexicana, and phylogenetic analysis grouped the species in the L. mexicana-amazonensis cluster. The literature review revealed 241 cases of Leishmania spp. infection among 1219 Pilosa mammals evaluated, with prevalence between studies ranging from 3.5% in the brown-throated three-toed sloth (Bradypus variegatus) to 78% in the Hoffman’s two-toed sloth (Choloepus hoffmanni). Current scientific information indicates that C. hoffmanni sloths are reservoirs of Leishmania, and further studies are needed in order to clarify if other Pilosa species play a role in Leishmania transmission.

The aim of this paper is to report the first description of gross pathological aspects of otoacariasis due to Edentalges bradypus Fonseca 1954 (Acari; Psoroptidae) infestation in the brown-throated three-toed sloth Bradypus variegatus Schinz, 1825 (Xenarthra; Bradypodidae) in Brazil. Mites were collected from massive skin crusts seen in both external ear canals and around both eyes of an extremely debilitated advanced-aged female sloth brought to the Wildlife Care Section of Universidade Estácio de Sá, Rio de Janeiro, Brazil.
