Iron and oxidizing species in oxidative stress and Alzheimer's disease

Iron species can participate in the Fenton or Fenton‐like reaction to generate oxidizing species that can cause oxidative damages to biomolecules and induce oxidative stress in the body. Furthermore, iron accumulation and oxidative stress have been shown to associate with the pathological progression of neurodegenerative disorders, including Alzheimer's disease (AD) and Parkinson's disease (PD). In this review, the role of iron species in generating the most deleterious free radical species (ie, hydroxyl radical) and effects of this species in causing oxidative stress in vivo are described. The implications of oxidative stress and the recently recognized cell death pathway (ie, ferroptosis) to AD are addressed. Strategies to combat this neurodegenerative disease, such as iron chelation and antioxidant therapies, and future research directions on this aspect are also discussed.

Cognitive impairment of cerebral small vessel disease (CSVD) is one of the most common cognitive disorders. It has a high incidence and results in heavy social burden; thus, it is essential to provide reasonable diagnosis and treatment in clinical practice. Based on the results of clinical research and related reports, combined with the actual situation in China, we propose a diagnosis and treatment guideline for cognitive impairment of CSVD.

Advances in screening instruments for Alzheimer's disease

At its fundamental basis, Alzheimer's disease (AD) is a pathological process that affects neuroplasticity, leading to a specific disruption of episodic memory. This review will provide a rationale for calls to screen for the early detection of AD, appraise the currently available cognitive instruments for AD detection, and focus on the development of the MemTrax test, which provides a new approach to detect the early manifestations and progression of the dementia associated with AD. MemTrax assesses metrics that reflect the effects of neuroplastic processes on learning, memory, and cognition, which are affected by age and AD, particularly episodic memory functions, which cannot presently be measured with enough precision for meaningful use. Further development of MemTrax would be of great value to the early detection of AD and would provide support for the testing of early interventions.

Serum 24‐hydroxycholesterol in probable Alzheimer's dementia: Reexploring the significance of a tentative Alzheimer's disease biomarker

Objective This study measured and analyzed the serum levels of 24‐hydroxycholesterol in patients with probable Alzheimer's disease (AD) and age‐/sex‐matched controls. Methods A case‐control study involving 40 AD patients and 40 controls was performed at a tertiary neurological teaching hospital in eastern India. Blood and serum samples were collected for APOE genotyping and 24‐hydroxycholesterol levels, respectively. Results Serum 24‐hydroxycholesterol was significantly lower in AD patients compared to controls (median concentration: controls, 47.14 ng/mL (interquartile range, 16.34); AD patients, 32.93 ng/mL (interquartile range, 9.45); P < 0.001) but showed no significant correlation with Mini Mental State Examination (MMSE) score in AD cases (r = −0.169, P = 0.298) or in controls (r = 0.18, P = 0.26). No statistically significant difference was observed between serum 24‐hydroxycholesterol levels of the APOE4‐positive and ‐negative subgroups in AD patients (P = 0.79). Findings were consistent and unchanged even when the ratio of serum 24‐hydroxycholesterol to serum total cholesterol was considered. Conclusion The decreased 24‐hydroxycholesterol level in peripheral circulation in AD cases observed in the present study may suggest its role in AD pathogenesis. The lack of a clear correlation between serum levels of 24‐hydroxycholesterol and MMSE score—a surrogate marker of AD severity—raises the question as to whether 24‐hydroxycholesterol level declines with decreasing neuronal mass or whether the steroid continues to play a protective role.

Memory for Melodies and Lyrics in Alzheimer's Disease

this research addressed the question: is musical
memory preserved in dementia, specifically, dementia of the Alzheimer type (AD)? Six tests involving different aspects of melody and language processing were administered to each of five groups of participants: 50 younger adults, 100 older adults, and 50 AD older adults classified into three levels of AD severity—mild, moderate and severe. No test was immune to, but not all tests were equally sensitive to, the presence of dementia. Long-term familiarity for melody was preserved across levels of AD, even at the severe stage for a few individuals. Detecting pitch distortions in melodies was possible for mild and some of the moderate AD participants. The ability to sing a melody when prompted by its lyrics was retained at the mild stage and was retained by a few individuals through the severe stages of AD. Long-term familiarity with the lyrics of familiar melodies was also found across levels of AD. In contrast, detection of grammatical distortions in the lyrics of familiar melodies and the ability to complete familiar proverbs were affected even at the mild stage of AD. We conclude that musical semantic memory may be spared through the mild and moderate stages of AD and may be preserved even in some individuals at the severe stage.

Magnetic resonance imaging (MRI) of the brain done at a hospital in her locality at the onset of her psychiatric symptoms had been reported as normal, and a cerebrospinal fluid analysis was unremarkable. There is no significant voltage asymmetry between right and left hemisphere DISCUSSION Sporadic CJD results from either a somatic mutation in the prion protein (PrP) gene or a random structural change in the PrP causing formation of PrPSc (abnormal or scrapie type prion protein). the goal of this study is to assess whether new lyrics are
better learned and memorized when presented in a spoken or sung form. In normal young adults, mixed results have been reported, with studies showing a positive, a negative, or a null effect of singing on verbal recall. Several factors can account for this limited aid of music. First, the familiarity of the melody might play a role. Second, successive learning sessions and long-term retention intervals may be necessary. These two factors are considered here in a case study of a participant who suffers from mild Alzheimer's disease. As expected, initial learning of new lyrics showed better performance for the spoken condition over the sung version unless the lyrics are learned on a familiar melody. After repeated learning episodes, learning sung lyrics – even on an unfamiliar melody – led to better retention of words. Thus, music may provide a more robust aid for consolidation in memory than spoken lyrics alone. The therapeutic implications of these results are discussed.

in this study, we examined short and longer term effects
of musical and cooking interventions on emotional well-being of severe Alzheimer's disease (AD) patients. These two pleasurable activities (i.e., listening to music, tasting sweets) that were collectively performed (i.e., playing music together, collaborative preparation of a cake) were compared in two groups of matched patients with AD (N= 14). Each intervention lasted four weeks (two sessions per week) and their effects were regularly assessed up to four weeks after the end of the intervention. We repeatedly evaluated the emotional state of both groups before, during, and after the intervention periods by analyzing discourse content and facial expressions from short filmed interviews as well as caregivers' judgments of mood. The results reveal short-term benefits of both music and cooking interventions on emotional state on all these measures, but long-term benefits were only evident after the music intervention. The present finding suggests that non-pharmacological approaches offer promising methods to improve the quality of life of patients with dementia and that music stimulation is particularly effective to produce long lasting effects on patients' emotional well-being.

Semantic Memory for Music in Dementia

there is currently limited information about the
effects of dementia diseases on semantic memory for music: memory for musical objects and concepts. Here we review available evidence and emerging research directions in semantic memory for music in the degenerative dementias. Neurodegenerative pathologies affect distributed brain networks and can therefore provide a perspective on musical semantic memory that complements the traditional neuropsychological paradigm of the focal brain lesion. Recent work suggests that semantic memory for music may be fractionated and may share certain cognitive organizational principles with semantic memory for other kinds of material. Profiles of impairment on different dimensions of musical semantic memory may show some specificity for particular dementia diseases (for example, semantic dementia versus Alzheimer's disease).

Episodic and Semantic Memory for Melodies in Alzheimer's Disease

the present study addressed episodic and semantic
memory for melodies in three groups of participants: 35 younger adults, 40 older adults, and 10 individuals with mild to moderate Alzheimer's disease (AD). To assess episodic memory, a study list of eight novel target melodies was presented three times, followed by a test trial in which target melodies were mixed with foil (previously unheard) melodies. Both explicit and implicit measures were obtained. Explicit memory was assessed by the accuracy of discrimination of the target melodies from the foils. Younger adults were significantly more accurate than older adults, who in turn had significantly higher scores than AD adults. Implicit memory was assessed by examining the difference in pleasantness ratings between target and foil melodies. Younger adults showed significantly greater differences in pleasantness ratings than older adults and AD adults; scores for the two latter groups did not differ. To assess semantic memory, participants were asked to identify traditional melodies within a series of traditional and novel melodies. In contrast to the episodic memory results, all three groups showed very high scores on the semantic memory task with no significant differences among groups. The results support the notion that, though other forms of musical memory may be compromised, semantic memory for melody may be preserved in normal aging and in AD.
