Molecular Characteristics and Pathogenicity of Staphylococcus aureus Exotoxins
Staphylococcus aureus stands as one of the most pervasive pathogens given its morbidity and mortality worldwide due to its roles as an infectious agent that causes a wide variety of diseases ranging from moderately severe skin infections to fatal pneumonia and sepsis. S. aureus produces a variety of exotoxins that serve as important virulence factors in S. aureus-related infectious diseases and food poisoning in both humans and animals. For example, staphylococcal enterotoxins (SEs) produced by S. aureus induce staphylococcal foodborne poisoning; toxic shock syndrome toxin-1 (TSST-1), as a typical superantigen, induces toxic shock syndrome; hemolysins induce cell damage in erythrocytes and leukocytes; and exfoliative toxin induces staphylococcal skin scalded syndrome. Recently, Panton–Valentine leucocidin, a cytotoxin produced by community-associated methicillin-resistant S. aureus (CA-MRSA), has been reported, and new types of SEs and staphylococcal enterotoxin-like toxins (SEls) were discovered and reported successively. This review addresses the progress of and novel insights into the molecular structure, biological activities, and pathogenicity of both the classic and the newly identified exotoxins produced by S. aureus.
Panton-Valentine Leukocidin–Positive Staphylococcus aureus in Family and Pet Cat
Continued detection of Panton-Valentine leukocidin-positive Staphylococcus aureus in samples from a family with severe repeated skin infections and their pet cat suggests transmission between the family and the cat. Decolonizing the pet led to successful elimination of the bacteria from the household. Clinicians should consider pet cats as possible reinfection sources.
Skin Infections Caused by Panton-Valentine Leukocidin and Methicillin-Susceptible Staphylococcus aureus in Child, Japan
We describe a pediatric case of recurrent skin infections caused by a Panton-Valentine leukocidin and exfoliative toxin E double-positive methicillin-susceptible Staphylococcus aureus clonal complex 188 clone. Most of the patient's family members were infected with the same strain, and intrafamilial transmission was strongly suspected. Decolonization procedures were not effective.
Panton-Valentine Leukocidin–Encoding Methicillin-Resistant Staphylococcus aureus, the Netherlands, 2023–2024
We describe a community outbreak of Panton-Valentine leukocidin–positive methicillin-resistant Staphylococcus aureus (MRSA) during November 2023–June 2024 in the Netherlands. We identified a massage center as the source. Case-patients experienced skin infections and abscesses. This study highlights the importance of genomic surveillance of MRSA in distinguishing Panton-Valentine leukocidin–positive MRSA.
Imagining Modernity
This book is a detailed study of the architecture of Valentine Gunasekara (1931-2017). It provides an innovative lens to understand the formation of a Ceylonese middle-class, which was inspired by the post-independence desire for modernity. Their experiments, values and dynamic social history are the framework for this research.Although neglected by his peers and marginalized by the prevalent discourse on vernacular regionalism, Gunasekara's work poses important questions regarding the utopian ideals of the modernist project and its successes and its failures in Asia. More significantly, his work reveals the European and American influences that shaped the first generation of Ceylonese architects and their efforts at adapting new materials and technologies to a very different climate and culture.This book documents a wide range of Gunasekara's projects including residential, religious and commercial buildings arguing that they represented a nascent cosmopolitanism from below that proved to be quite antithetical to regionalist trends in architecture.This e-book is a re-publication of an earlier edition published by Stamford Lake in 2007.
The genetic feature and virulence determinant of highly virulent community-associated MRSA ST338-SCCmec Vb in China
ST59 is the predominant pathotype of community-associated methicillin-resistant Staphylococcus aureus (CA-MRSA) in China. As a variant of ST59, there is relatively little known about the detailed information of ST338. To address this issue, here, we described thirteen ST338 CA-MRSA strains isolated from severe bloodstream infection cases, and focused on their epidemiology, genetic features and virulence potential. Phylogenetic analysis showed the earliest isolated strain of this study is likely a predecessor of recent ST338 lineage (after year of 2014). Furthermore, the phylogenetic reconstruction and time estimation suggested that ST338 evolved from ST59 in 1991. Notably, the carrying patten of virulence factors of all ST338 strains were similar, and the genomic islands νSaα, νSaγ and SaPI and the core virulence factors like hla and psm were detected in ST338 isolates. However, all ST338 isolates lacked some adhesion factors such as clfA, clfB, eap, cna and icaD. Additionally, among these ST338 strains, one PVL-negative ST338 isolate was detected. Experiment on mice nose and human alveolar epithelial cell showed that the nasal colonization ability of ST338 was weaker than that of CA-MRSA MW2. In a mouse bloodstream infection model and skin infection model, PVL+ and PVL− strains had the similar virulence, which was dependent on upregulation of toxin genes rather than the presence of mobile genetic elements such as ΦSa2 carrying PVL. Our findings provide important insight into the epidemiology and pathogenicity of the novel and highly virulent ST338-SCCmec Vb clone.
Prevalence and Genetic Characteristics of Staphylococcus aureus and Staphylococcus argenteus Isolates Harboring Panton-Valentine Leukocidin, Enterotoxins, and TSST-1 Genes from Food Handlers in Myanmar
Asymptomatic carriers of toxigenic Staphylococcus aureus are potential source of diseases, including food poisoning. Toxigenic potential and genetic traits of colonizing S. aureus were investigated for 563 healthy food handlers in Myanmar. Carriage of S. aureus was found in 110 individuals (19.5%), and a total of 144 S. aureus isolates were recovered from nasal cavities (110 isolates) and hands (34 isolates). Panton-Valentine leucocidin genes (pvl) were detected in 18 isolates (12.5%), among which 11 isolates were classified into coa-VIa, agr type III, and ST1930 (CC96) that had been also detected in pvl-positive clinical isolates in Myanmar. A pvl-positive, ST2250 nasal isolate was identified as S. argenteus, a novel coagulase-positive staphylococcus species. Toxic shock syndrome toxin-1 (TSST-1) gene was detected in five pvl-negative isolates. All of the 144 isolates harbored at least one of the 21 enterotoxin(-like) gene(s). The most prevalent enterotoxin(-like) gene was selw (98%), followed by selx (97%), sei (28%), sely (28%), sem (26%), sel (24%), and sea and sec (22% each). Considerable genetic diversity with five groups was detected for selw. The present study revealed the relatively high rate of pvl, as well as the wide distribution of enterotoxin(-like) genes among colonizing S. aureus in Myanmar.
