Common bacterial blight of bean: a model of seed transmission and pathological convergence
Background Xanthomonas citri pv. fuscans (Xcf) and Xanthomonas phaseoli pv. phaseoli (Xpp) are the causal agents of common bacterial blight of bean (CBB), an important disease worldwide that remains difficult to control. These pathogens belong to distinct species within the Xanthomonas genus and have undergone a dynamic evolutionary history including the horizontal transfer of genes encoding factors probably involved in adaptation to and pathogenicity on common bean. Seed transmission is a key point of the CBB disease cycle, favouring both vertical transmission of the pathogen and worldwide distribution of the disease through global seed trade. Taxonomy Kingdom: Bacteria; phylum: Proteobacteria; class: Gammaproteobacteria; order: Lysobacterales (also known as Xanthomonadales); family: Lysobacteraceae (also known as Xanthomonadaceae); genus: Xanthomonas; species: X. citri pv. fuscans and X. phaseoli pv. phaseoli (Xcf‐Xpp). Host range The main host of Xcf‐Xpp is the common bean (Phaseolus vulgaris). Lima bean (Phaseolus lunatus) and members of the Vigna genus (Vigna aconitifolia, Vigna angularis, Vigna mungo, Vigna radiata, and Vigna umbellata) are also natural hosts of Xcf‐Xpp. Natural occurrence of Xcf‐Xpp has been reported for a handful of other legumes such as Calopogonium sp., Pueraria sp., pea (Pisum sativum), Lablab purpureus, Macroptilium lathyroides, and Strophostyles helvola. There are conflicting reports concerning the natural occurrence of CBB agents on tepary bean (Phaseolus acutifolius) and cowpea (Vigna unguiculata subsp. unguiculata). Symptoms CBB symptoms occur on all aerial parts of beans, that is, seedlings, leaves, stems, pods, and seeds. Symptoms initially appear as water‐soaked spots evolving into necrosis on leaves, pustules on pods, and cankers on twigs. In severe infections, defoliation and wilting may occur. Distribution CBB is distributed worldwide, meaning that it is frequently encountered in most places where bean is cultivated in the Americas, Asia, Africa, and Oceania, except for arid tropical areas. Xcf‐Xpp are regulated nonquarantine pathogens in Europe and are listed in the A2 list by the European and Mediterranean Plant Protection Organization (EPPO). Genome The genome consists of a single circular chromosome plus one to four extrachromosomal plasmids of various sizes, for a total mean size of 5.27 Mb with 64.7% GC content and an average predicted number of 4,181 coding sequences. Disease control Management of CBB is based on integrated approaches that comprise measures aimed at avoiding Xcf‐Xpp introduction through infected seeds, cultural practices to limit Xcf‐Xpp survival between host crops, whenever possible the use of tolerant or resistant bean genotypes, and chemical treatments, mainly restricted to copper compounds. The use of pathogen‐free seeds is essential in an effective management strategy and requires appropriate sampling, detection, and identification methods. Novel Genetic Resources in the Genus Vigna Unveiled from Gene Bank Accessions
The genus Vigna (Fabaceae) consists of five subgenera, and includes more than 100 wild species. In Vigna, 10 crops have been domesticated from three subgenera, Vigna, Plectrotropis, and Ceratotropis. The habitats of wild Vigna species are so diverse that their genomes could harbor various genes responsible for environmental stress adaptation, which could lead to innovations in agriculture. Since some of the gene bank Vigna accessions were unidentified and they seemed to be novel genetic resources, these accessions were identified based on morphological traits. The phylogenetic positions were estimated based on the DNA sequences of nuclear rDNA-ITS and chloroplast atpB-rbcL spacer regions. Based on the results, the potential usefulness of the recently described species V. indica and V. sahyadriana, and some wild Vigna species, i.e., V. aconitifolia, V. dalzelliana, V. khandalensis, V. marina var. oblonga, and V. vexillata, was discussed.
QTL analysis of domestication syndrome in zombi pea (Vigna vexillata), an underutilized legume crop
Zombi pea (Vigna vexillata (L.) A. Rich) is an underutilized crop belonging to the genus Vigna. Two domesticated forms of zombi pea are cultivated as crop plants; seed and tuber forms. The cultivated seed form is present in Africa, while the cultivated tuber form is present in a very limited part of Asia. Genetics of domestication have been investigated in most of cultivated Vigna crops by means of quantitative trait locus (QTL) mapping. In this study, we investigated genetics of domestication in zombi pea by QTL analysis using an F2 population of 139 plants derived from a cross between cultivated tuber form of V. vexillata (JP235863) and wild V. vexillata (AusTRCF66514). A linkage map with 11 linkage groups (LGs) was constructed from this F2 population using 145 SSR, 117 RAD-seq and 2 morphological markers. Many highly segregation distorted markers were found on LGs 5, 6, 7, 8, 10 and 11. Most of the distorted markers were clustered together and all the markers on LG8 were highly distorted markers. Comparing this V. vexillata linkage map with linkage maps of other four Vigna species demonstrated several genome rearrangements in V. vexillata. QTL analysis for 22 domestication-related traits was investigated by inclusive composite interval mapping in which 37 QTLs were identified for 18 traits; no QTL was detected for 4 traits. Number of QTLs detected in each trait ranged from 1 to 5 with an average of only 2.3. Five QTLs for tuber width and three QTLs for tuber weight. Interestingly, 2 QTLs each for tuber width and tuber weight detected on LG2 and LG4 were located at similar position and wild allele increased tuber width and weight. This indicated wild germplasm having small tuber have potential to increase yield of large tuber cultivated type. Large-effect QTLs (PVE > 20%) were on LG4 (pod length), LG5 (leaf size and seed thickness), and LG7 (for seed-related traits). Comparison of domestication-related QTLs of the zombi pea with those of cowpea (Vigna unguiculata), azuki bean (Vigna angularis), mungbean (Vigna radiata) and rice bean (Vigna umbellata) revealed that there was conservation of some QTLs for seed size, pod size and leaf size between zombi pea and cowpea and that QTLs associated with seed size (weight, length, width and thickness) in each species were clustered on same linkage.
AMMI and GGE biplot analysis for yield performance and stability assessment of selected Bambara groundnut (Vigna subterranea L. Verdc.) genotypes under the multi-environmental trails (METs)
The stability and high yielding of Vigna subterranea L. Verdc. genotype is an important factor for long-term development and food security. The effects of G × E interaction on yield stability in 30 Bambara groundnut genotypes in four different Malaysian environments were investigated in this research. The experiment used a randomized complete block design with three replications in each environment. Over multiple harvests, yield component traits such as the total number of pods per plant, fresh pods weight (g), hundred seeds weight (g), and yield per hectare were evaluated in the main and off-season in 2020 and 2021. Stability tests for multivariate stability parameters were performed based on analyses of variance. For all the traits, the pooled analysis of variance revealed highly significant (p < 0.01) variations between genotypes, locations, seasons, and genotypes by environment (G × E interaction). A two-dimensional GGE biplot was generated using the first two principal components (axis 1 and axis 2), which accounted for 94.97% and 3.11% difference in GEI for yield per hectare, respectively. Season and location were found to be the most significant causes of yield heterogeneity, accounting for 31.13% and 14.02% of overall G + E + G × E variation, respectively, according to the combined study of variance. The GGE biplot revealed that the three winning genotypes G1, G3, and G5 appear across environments whereas AMMI model exposed genotypes viz G18, G14, G7, G3, G1, and G5 as best performer. Based on ideal genotype ranking genotype G1 was the best performer, with a high mean yield and high stability in the tested environment. According to the AEC line, genotypes G1 and G3 were extremely stable, while genotypes G2 and G4 were low stable, with a high average yielding per hectare. A GGE and AMMI biplot graphically showed the interrelationships between the tested environment and genotypes, classified genotypes into three categories as well as simplifying visual evaluations, according to this investigation. According to our results, breeding could improve yield production, and the genotypes discovered could be recommended for commercial cultivation.
Zinc biofortification potential of diverse mungbean Vigna radiata (L.) Wilczek genotypes under field conditions
Zinc (Zn) is an important micronutrient for crop plants and essential for human health. The Zn-deficiency is an important malnutrition problem known globally. Biofortified foods could overcome Zn deficiency in humans. Mungbean [ Vigna radiata (L.) Wilczek] is an important, pulse crop frequently grown in arid and semi-arid regions of the world. Mungbean could provide essential micronutrients, including Zn to humans. Therefore, it is very important to investigate the impact of Zn fertilization on the yield and grain biofortification of mungbean. Twelve mungbean genotypes (i.e., NM-28, NM-2011, NM-13-1, NM-2006, NM-51, NM-54, NM-19-19, NM-92, NM-121-25, NM-20-21, 7006, 7008) were assessed for their genetic diversity followed by Zn-biofortification, growth and yield under control (0 kg ha -1 ) and Zn-fertilized (10 kg ha -1 ) conditions. Data relating to allometric traits, yield components, grain yield and grain Zn contents were recorded. Zinc fertilization improved entire allometric and yield-related traits. Grain yield of different genotypes ranged from 439 to 904 kg ha -1 under control and 536 to 1462 kg ha -1 under Zn-fertilization. Zinc concentration in the grains varied from 15.50 to 45.60 mg kg -1 under control and 18.53 to 64.23 mg kg -1 under Zn-fertilized conditions. The tested genotypes differed in their Zn-biofortification potential. The highest and the lowest grain Zn contents were noted for genotypes NM-28 and NM-121-25, respectively. Significant variation in yield and Zn-biofortification indicated the potential for improvement in mungbean yield and grain Zn-biofortification. The genotypes NM-28 and NM-2006 could be used in breeding programs for improvement in grain Zn concentration due to their high Zn uptake potential. Nonetheless, all available genotypes in the country should be screened for their Zn-biofortification potential.
Agar with embedded channels to study root growth
Agar have long been used as a growth media for plants. Here, we made agar media with embedded fluidic channels to study the effect of exposure to nutrient solution on root growth and pull-out force. Black Eye bean ( Vigna Unguiculata ) and Mung bean ( Vigna Radiata ) were used in this study due to their rapid root development. Agar media were fabricated using casting process with removable cores to form channels which were subsequently filled with nutrient solution. Upon germination, beans were transplanted onto the agar media and allowed to grow. Pull-out force was determined at 96, 120 and 144 h after germination by applying a force on the hypocotyl above the gel surface. The effect of nutrients was investigated by comparing corresponding data obtained from control plants which have not been exposed to nutrient solution. Pull-out force of Black Eye bean plantlets grown in agar with nutrient solution in channels was greater than those grown in gel without nutrients and was 110% greater after 144 h of germination. Pull-out force of Mung bean plantlets grown in agar with and without nutrient solution was similar. Tap root lengths of Black Eye bean and Mung Bean plantlets grown in agar with nutrient solution are shorter than those grown without nutrient. 
Bambara groundnut
World agriculture is currently based on very limited numbers of crops, representing a significant risk to food supplies, particularly in the face of climate change which is expected to increase the frequency of extreme events. Minor and underutilised crops can help to develop a more resilient and nutritionally dense future agriculture. Bambara groundnut [Vigna subterranea (L.) Verdc.[, as a drought resistant, nitrogen-fixing, legume has a role to play. However, as with most underutilised crops, there are significant gaps in knowledge and also negative traits such as ‘hard-to-cook’ and ‘photoperiod sensitivity to pod filling’ associated with the crop which future breeding programmes and processing methods need to tackle, to allow it to make a significant contribution to the well-being of future generations. The current review assesses these factors and also considers what are the next steps towards realising the potential of this crop.
Mapping patterns of abiotic and biotic stress resilience uncovers conservation gaps and breeding potential of Vigna wild relatives
This study provides insights in patterns of distribution of abiotic and biotic stress resilience across Vigna gene pools to enhance the use and conservation of these genetic resources for legume breeding. Vigna is a pantropical genus with more than 88 taxa including important crops such as V. radiata (mung bean) and V. unguiculata (cowpea). Our results show that sources of pest and disease resistance occur in at least 75 percent of the Vigna taxa, which were part of screening assessments, while sources of abiotic stress resilience occur in less than 30 percent of screened taxa. This difference in levels of resilience suggests that Vigna taxa co-evolve with pests and diseases while taxa are more conservative to adapt to climatic changes and salinization. Twenty-two Vigna taxa are poorly conserved in genebanks or not at all. This germplasm is not available for legume breeding and requires urgent germplasm collecting before these taxa extirpate on farm and in the wild. Vigna taxa, which tolerate heat and drought stress are rare compared with taxa, which escape these stresses because of short growing seasons or with taxa, which tolerate salinity. We recommend prioritizing these rare Vigna taxa for conservation and screening for combined abiotic and biotic stress resilience resulting from stacked or multifunctional traits. The high presence of salinity tolerance compared with drought stress tolerance, suggests that Vigna taxa are good at developing salt-tolerant traits. Vigna taxa are therefore of high value for legume production in areas that will suffer from salinization under global climate change.
Transgenic cowpea plants expressing Bacillus thuringiensis Cry2Aa insecticidal protein imparts resistance to Maruca vitrata legume pod borer
Key messageFertile independent transgenic cowpea lines expressing the BtCry2Aa toxin with increased resistance to the most devastating lepidopteran insect pest, Maruca pod borer has been developed for the first time.Cowpea is a staple legume important for food and nutritional security in sub-Saharan Africa and Asia, where its production is limited by the key pest, legume pod borer (Maruca vitrata). Cowpea varieties resistant to M. vitrata are not known, hence, development of Maruca pod borer resistance cowpea through genetic engineering is a promising approach to improve its production. In the present study, transgenic cowpea plants expressing Bacillus thuringiensis Cry2Aa insecticidal protein were developed for the first time using Agrobacterium tumefaciens-mediated transformation of cotyledonary explants. T0 plants recovered from Agrobacterium cocultured explants on medium containing 120 mgl−1 of kanamycin were identified on the basis of the presence of transgenes by PCR, their integration into genome by Southern hybridization and expression of their transcripts by semi quantitative PCR (sqRT-PCR) and quantitative Real–time-PCR (qRT-PCR) and protein by Western blot analysis. The transformation efficiency obtained was 3.47% with 11 independent T0 transgenic lines. The bioefficacy of Cry2Aa protein expressed in randomly selected four T0 plant’s leaves and pods was evaluated by feeding Maruca pod borer demonstrated a significant lower damage and a high level of Maruca mortality (more than 90%) for all these Bt lines. The inheritance of transgenes from T0 to T1 progeny plants was demonstrated by PCR analysis. The transgenic plants generated in this study can be used in cowpea breeding program for durable and sustainable legume pod borer resistance.
Construction of a Genetic Linkage Map and Genetic Analysis of Domestication Related Traits in Mungbean (Vigna radiata)
The genetic differences between mungbean and its presumed wild ancestor were analyzed for domestication related traits by QTL mapping. A genetic linkage map of mungbean was constructed using 430 SSR and EST-SSR markers from mungbean and its related species, and all these markers were mapped onto 11 linkage groups spanning a total of 727.6 cM. The present mungbean map is the first map where the number of linkage groups coincided with the haploid chromosome number of mungbean. In total 105 QTLs and genes for 38 domestication related traits were identified. Compared with the situation in other Vigna crops, many linkage groups have played an important role in the domestication of mungbean. In particular the QTLs with high contribution were distributed on seven out of 11 linkage groups. In addition, a large number of QTLs with small contribution were found. The accumulation of many mutations with large and/or small contribution has contributed to the differentiation between wild and cultivated mungbean. The useful QTLs for seed size, pod dehiscence and pod maturity that have not been found in other Asian Vigna species were identified in mungbean, and these QTLs may play the important role as new gene resources for other Asian Vigna species. The results provide the foundation that will be useful for improvement of mungbean and related legumes.
