Phytochemical screening and in vitro antibacterial, antifungal, antioxidant and antitumor activities of the red propolis Alagoas
Abstract The study aimed to evaluate the antimicrobial activity, antioxidant, toxicity and phytochemical screening of the Red Propolis Alagoas. Antimicrobial activity was evaluated by disk diffusion method. Determination of antioxidant activity was performed using the DPPH assay (1.1-diphenyl-2-picrylhydrazyl), FTC (ferric thiocyanate) and determination of phenolic compounds by Follin method. Toxicity was performed by the method of Artemia salina and cytotoxicity by MTT method. The phytochemical screening for the detection of allelochemicals was performed. The ethanol extract of propolis of Alagoas showed significant results for antimicrobial activity, and inhibitory activity for Staphylococcus aureus and Candida krusei. The antioxidant activity of the FTC method was 80% to 108.3% hydrogen peroxide kidnapping, the DPPH method showed an EC50 3.97 mg/mL, the content of total phenolic compounds was determined by calibration curve gallic acid, resulting from 0.0005 mg/100 g of gallic acid equivalent. The extract was non-toxic by A. salina method. The propolis extract showed high activity with a higher percentage than 75% inhibition of tumor cells OVCAR-8, SF-295 and HCT116. Chemical constituents were observed as flavonones, xanthones, flavonols, and Chalcones Auronas, Catechins and leucoanthocyanidins. It is concluded that the extract can be tested is considered a potential source of bioactive metabolites. Resumo O trabalho teve como objetivo avaliar a atividade antimicrobiana, antioxidante, a toxicidade e a prospecção fitoquímica da Própolis Vermelha de Alagoas. A atividade antimicrobiana foi avaliada pelo método de difusão em disco. A determinação do potencial antioxidante foi realizada utilizando o método de DPPH (1,1-difenil-2-picrilhidrazil), FTC (Tiocianato Férrico) e determinação de compostos fenólicos pelo método de Follin. A toxicidade foi realizada pelo método de Artemia salina e a citotoxicidade pelo método do MTT. Foi realizada a prospecção fitoquímica para a pesquisa de aleloquímicos. O extrato etanólico da própolis vermelha de Alagoas apresentou resultados significantes para atividade antimicrobiana, tendo a atividade inibitória para Staphylococcus aureus e Candida krusei. Quanto a atividade antioxidante o método de FTC teve 80% a 108,3% de sequestro de peróxido de hidrogênio, o método de DPPH apresentou um CE50 de 3,97 μg/mL, o teor de compostos fenólicos totais foi determinado mediante curva de calibração do ácido gálico, tendo resultado de 0,0005 mg/100 g equivalente de ácido gálico. O extrato foi atóxico pelo método de A. salina. O extrato da própolis mostrou elevada atividade com percentual de inibição maior que 75% sobre células tumorais OVCAR-8, SF-295 e HCT116. Foram observados constituintes químicos como flavononas, xantonas, flavonóis, Chalconas e Auronas, Catequinas e Leucoantocianidinas. Conclui-se que o extrato testado pode ser considerado é uma fonte potencial de metabólitos bioativos.
Phytochemistry and antibacterial property of Arisaema liemiana
Introduction: Arisaema liemiana was newly identified as a new species for the flora of Vietnam in 2020. It was discovered in Takou Nature Reserve, Binh Thuan province, Vietnam, characterized by its height of 20-50 centimeters, deciduous nature, light yellow spathe limb, white spathe tube, and spadix appendix covered with filiform neuter structures. Aim: This study aimed to investigate the chemical components and antibacterial activity of the acetone extracts derived from the aerial parts and tubers of A. liemiana. Methodology: A. liemiana specimens were collected from Takou Mountain within Takou Nature Reserve, Binh Thuan Province, and subjected to acetone extraction. The chemical components of these extracts were analyzed using GC-MS, while the antibacterial activity was assessed through the agar disc diffusion method. Results: A total of 57 compounds were identified in the aerial part and tuber extracts of which 32 components were found in the first extract while the latter one contained 34 constituents. The antibacterial effectiveness of the tuber acetone extracts was assessed against 10 bacterial strains, revealing diverse inhibition zones. Notably, S. aureus ATCC 25923 exhibited the largest inhibition zone diameter, followed by B. cereus, V. parahaemolyticus, S. flexneri, P. aeruginosa, E. coli, L. monocytogenes, S. saprophyticus, S. typhimurium, and S. enteritidis. Conclusion: The chemical composition and antibacterial properties of acetone extracts derived from Arisaema liemiana was investigated for the first time. The observed notable antibacterial efficacy suggests potential pharmacological implications of A. liemiana. Additionally, this research unveils broader applications of A. liemiana in herbal medicine. Introducción: Arisaema liemiana fue identificada recientemente como una nueva especie para la flora de Vietnam en 2020. Fue descubierta en la Reserva Natural de Takou, provincia de Binh Thuan, Vietnam, caracterizada por su altura de 20-50 centímetros, naturaleza caducifolia, extremidad de espata de color amarillo claro, tubo de espata blanco y apéndice del espádice cubierto de estructuras neutras filiformes. Objetivo: Este estudio tuvo como objetivo investigar los componentes químicos y la actividad antibacteriana de los extractos de acetona derivados de las partes aéreas y tubérculos de A. liemiana. Metodología: Los especímenes de A. liemiana se recolectaron de la montaña Takou dentro de la Reserva Natural de Takou, provincia de Binh Thuan, y se sometieron a extracción con acetona. Los componentes químicos de estos extractos se analizaron mediante GC-MS, mientras que la actividad antibacteriana se evaluó mediante el método de difusión en disco de agar. Resultados: Se identificaron 57 compuestos en los extractos de la parte aérea y del tubérculo, de los cuales 32 componentes se encontraron en el primer extracto, mientras que el último contenía 34 constituyentes. Se evaluó la eficacia antibacteriana de los extractos de acetona del tubérculo frente a 10 cepas bacterianas, lo que reveló diversas zonas de inhibición. Cabe destacar que S. aureus ATCC 25923 exhibió el mayor diámetro de zona de inhibición, seguido de B. cereus, V. parahaemolyticus, S. flexneri, P. aeruginosa, E. coli, L. monocytogenes, S. saprophyticus, S. typhimurium y S. enteritidis. Conclusión: Se investigó por primera vez la composición química y las propiedades antibacterianas de los extractos de acetona derivados de Arisaema liemiana. La notable eficacia antibacteriana observada sugiere posibles implicaciones farmacológicas de A. liemiana. Además, esta investigación revela aplicaciones más amplias de A. liemiana en la medicina herbal. Chemical composition and in vitro antibacterial activity of three lichen species (Usnea sp., Thamnolia vermicularis subsp. solida and Ramalina asperula) collected from the Jauja and Huaral provinces-Peru
Introduction: Lichens are complex organisms conformed by a symbiotic association between algae and fungi; they are haven considerable interest owing to their pharmacological properties. Objectives: To determine chemical composition of the fatty acid (FA) and the percentage of usnic acid (UA), and to evaluate the in vitro antibacterial activity of the ethanolic extract obtained from the lichens Usnea sp., Thamnolia vermicularis subsp. solida and Ramalina asperula. Materials and Methods: Organic extracts from the three lichens were obtained using a rotary evaporator, soxhlet extractor and ultrasound equipment. The chemical composition of FA was determined by gas chromatography-flame ionisation detector, and the percentage of UA was quantified by UV-Vis spectroscopy. The in vitro antibacterial activity of the lichen ethanolic extracts was evaluated against E. coli ATCC 25922,Bacillus subtilis ATCC 11774, Staphylococcus aureus ATCC 43300 and S. aureus ATCC 29213 using the diffusion and microdilution methods. The minimum inhibitory concentration (MIC) was determined. Results: In the three tested lichens, the identified FAs were palmitic, stearic, oleic, linoleic, and linolenic acids; and the obtained percentage of UA was between 0.2 and 1.07 %. All the ethanolic extracts obtained from three lichens showed a high activity against B. subtilis (MIC = 0.5 mg/mL), while the ethanolic extracts obtained from T. vermicularis subsp. solida and R. asperula showed moderate activity against S. aureus ATCC 43300 (MIC = 3 and 1 mg/mL) and S. aureus ATCC29213 (MIC = 4 and 1 mg/mL). Conclusions: Our findings found for the three studied lichens confirmed the presence of some FAs and AU. These species provide an acceptable nutritional content and besides they can be considered as potential antibacterial agents.   Introducción: Los líquenes son organismos complejos conformados por una asociación simbiótica entre algas y hongos; tienen un interés considerable debido a sus propiedades farmacológicas. Objetivos: Determinar la composición química de los ácidos grasos (AG) y el porcentaje de ácido úsnico (AU), y evaluar la actividad antibacteriana in vitro del extracto etanólico obtenido a partir de los líquenes Usnea sp., Thamnolia vermicularis subsp. solida y Ramalina asperula. Materiales y Métodos: Los extractos orgánicos de los tres líquenes fueron obtenidos usando un rotavapor, extractor soxhlet y un equipo de ultrasonido. La composición química de los AG fue determinada mediante cromatografía de gases con detector de ionización de llama y el porcentaje de AU fue cuantificado mediante espectroscopia UV-Vis. La actividad antibacteriana in vitro de los extractos etanólicos de líquenes fue evaluada frente a las especies E. coli ATCC 25922, Bacillus subtilis ATCC 11774, Staphylococcus aureus ATCC 43300 y S. aureus ATCC 29213 mediante los métodos de difusión y microdilución. Se determinó la concentración inhibitoria mínima (MIC). Resultados: En los tres líquenes ensayados, los ácidos grasos identificados fueron los ácidos palmítico, esteárico, oleico, linoleico y linolénico; y el porcentaje de AU obtenido estuvo entre 0,2 y 1,07 %. Todos los extractos etanólicos obtenidos mostraron una alta actividad frente a la especie B. subtilis (MIC = 0,5 mg/mL), mientras que los extractos etanólicos obtenidos a partir de T. vermicularis subsp. solida y R. asperula mostraron actividad moderada frente a la especie S. aureus ATCC 43300 (MIC = 3 y 1 mg/mL) y S. aureus ATCC29213 (MIC = 4 y 1 mg/ mL). Conclusiones: Nuestros hallazgos encontrados para los tres líquenes estudiados confirmaron la presencia de algunos AG y AU. Estas especies aportan un aceptable contenido nutricional y además, pueden ser consideradas como potenciales agentes antibacterianos.   Introdução: Os líquenes são organismos complexos constituídos por uma associação simbiótica entre algas e fungos; eles são de considerável interesse devido às suas propriedades farmacológicas. Resultados: Nos três líquenes testados, os ácidos graxos identificados foram os ácidos palmítico, esteárico, oleico, linoléico e linolênico; e o percentual de UA obtido ficou entre 0,2 e 1,07%. Todos os extratos etanólicos obtidos apresentaram alta atividade contra a espécie B. subtilis (CIM = 0,5 mg/mL), enquanto os extratos etanólicos obtidos de T. vermicularis subsp. solida e R. asperula apresentaram atividade moderada contra as espécies S. aureus ATCC 43300 (CIM = 3 e 1 mg/mL) e S. aureus ATCC29213 (CIM = 4 e 1 mg/mL). Conclusões: Nossos achados para os três líquenes estudados confirmaram a presença de alguns AG e AU. Estas espécies fornecem um conteúdo nutricional aceitável e também podem ser consideradas como potenciais agentes antibacterianos.  
In vitro antibacterial activity of the crude methanol extract of Woodfordia fruticosa Kurz. flower (Lythraceae)
The antibacterial activity of the crude methanol extract of Woodfordia fruticosa Kurz. flower was evaluated at two different concentrations by the agar well diffusion method. The methanol extract of the flower exhibited antibacterial activity at varied levels except against Bacillus subtilis and Micrococcus flavus. The methanol extract was most active against Pseudomonas pseudoalcaligenes. The extract was more active against Gram-negative bacteria as compared to Gram-positive. The inhibitory effect of the extract was compared with standard antibiotics, amoxicillin and ciprofloxacin. A atividade antibacteriana do extrato metanólico bruto da flor Woodfordia fruticosa Kurz. (Lythraceae) foi avaliada em duas concentrações diferentes através do método de difusão em poços. A atividade antibacteriana ocorreu em diferentes níveis, exceto contra Bacillus subtilis e Micrococcus flavus. O extrato metanólico foi mais ativo contra Pseudomonas pseudoalcaligens. O extrato foi mais ativo contra bactérias Gram negativas do que bactérias Gram positivas. O efeito inibitório do extrato foi comparado ao dos antibióticos padrão amoxicilina e ciprofloxacina.
Coumarin (2H-1-Benzopyran-2-one) act against planktonic and biofilm forms of Staphylococcus aureus, Klebsiella pneumoniae and Pseudomonas aeruginosa
Introduction: biofilm-related infections caused by Staphylococcus aureus, Klebsi-ella pneumoniae and Pseudomonas aeruginosa are difficult to treat and few effective pharmacological options are currently available for this purpose. In this context, coumarin (2H-1-Benzopyran-2-one) has been reported to have antibacterial and antibiofilm activity, but this potential remains poorly understood. Aim: to investi-gate the action of coumarin on planktonic and biofilm forms of S. aureus, K. pneu-moniae and P. aeruginosa. Results: a minimum inhibitory concentration (MIC) of coumarin ranging from 256 to 1024 μg/mL was observed, with a remarkable ability to inhibit the formation of biofilms and to act on mature biofilms in concentrations close to MIC. Conclusion: coumarin has strong activity against planktonic and biofilm forms on the three species of great relevance in the clinical scenario. These results are interesting to enable a pharmacological alternative for the treatment of these infections.
Plant essential oils against bacteria isolated from fish: an in vitro screening and in vivo efficacy of Lippia origanoides
The use of natural products, such as essential oils (EOs), is a potential novel approach to treat fish bacterial infections with a lower risk of developing resistance. There has been a number of studies reporting the activity of EOs as those obtained from the species Achyrocline satureioides, Aniba parviflora, Aniba rosaeodora, Anthemis nobilis, Conobea scoparioides, Cupressus sempervirens, Illicium verum, Lippia origanoides, and Melaleuca alternifolia against bacteria. However, there are few studies investigating the effect of these EOs against fish bacteria. Therefore, the aim of this study was to evaluate the in vitro antibacterial activity of EOs against the following fish bacteria, Aeromonas hydrophila, Citrobacter freundii, and Raoultella ornithinolytica. Additionally, the in vivo antibacterial activity of the EO L. origanoides was evaluated against experimentally induced A. hydrophila infection of silver catfish (Rhamdia quelen). The EO of L. origanoides was chosen as it showed the highest in vitro antibacterial activity, with minimum inhibitory concentrations ranging from 0.2 to 0.8 mg mL-1. This EO also presented a therapeutic success of 58.33%, on a 30 day A. hydrophila infection. Therefore, we suggested that the EO of L. origanoides may be a viable alternative as a treatment for A. hydrophila infection. RESUMO: O uso de produtos naturais, como óleos essenciais (OEs), é uma nova abordagem potencial para o tratamento de infecções bacterianas em peixes com um baixo risco de desenvolvimento de resistência. Existem estudos reportando a atividade de OEs como aqueles obtidos das espécies Achyrocline satureioides, Aniba parviflora, Aniba rosaeodora, Anthemis nobilis, Conobea scoparioides, Cupressus sempervirens, Illicium verum, Lippia origanoides e Melaleuca alternifolia contra bactérias. Porém, existem poucos estudos investigando o efeito desses OEs supracitados contra bactérias isoladas de peixes. Portanto, o objetivo desse estudo foi avaliar a atividade antibacteriana in vitro desses OEs contra bactérias isoladas de peixes (Aeromonas hydrophila, Citrobacter freundii e Raoultella ornithinolytica). Assim como avaliar a atividade antibacteriana in vivo do OE de L. origanoides contra uma infecção por A. hydrophila experimentalmente induzida em jundiá (Rhamdia quelen). O OE de L. origanoides foi escolhido porque apresentou a melhor atividade antibacteriana in vitro, com concentrações inibitórias mínimas variando de 0,2 a 0,8 mg mL-1. Esse OE apresentou sucesso terapêutico de 58,33% no dia 30 pós-infecção. Portanto, sugerimos que o OE de L. origanoides possa ser uma alternativa viável no tratamento da infecção por A. hydrophila em peixes.
Antimicrobial activity of leaf and root extracts of tough lovegrass
This study aimed to evaluate tough lovegrass leaf and root extracts antimicrobial activity. The extracts (plant material: solvent, 1:10) were prepared by maceration with methanol:water (1:1) during ten days followed by a concentration in a rotary evaporator under reduced pressure. The extracts were resuspended in water containing 1% of dimethylsulfoxide (DMSO) to obtain a final concentration of 100 mg/mL and then filtered through a sterilizing membrane with 0.22μm. The antibacterial activity of the leaf and root extracts were evaluated against pathogenic and phytopathogenic bacteria by agar well diffusion and microdilution broth methods for the minimum inhibitory concentrations (MIC) determination. The antifungal activity of tough lovegrass leaf and root extracts were evaluated by micelial growth inhibition and conidial germination inhibition. The extracts presented low antibacterial activity against Staphylococcus aureus, Staphylococcus epidermidis, and Xanthomonas translucens, but the leaf extracts presented significant antifungal activity against the phytopathogenic fungus Drechslera tritici-repentis. The results support the continuity of the study in improving the effectiveness of the active extract for a possible use in pharmacology and agronomy and in attempting to determine the probable active antimicrobial compound.
Antimicrobial activity of some of the south-Indian spices against serotypes of Escherichia coli, Salmonella, Listeria monocytogenes and Aeromonas hydrophila
Antibacterial activity of extracts of Allium sativum (garlic), Myristica fragrans (nutmeg), Zingiber officinale (ginger), Allium cepa (onion) and Piper nigrum (pepper) has been evaluated against 20 different serogroups of Escherichia coli, 8 serotypes of Salmonella, Listeria monocytogenes and Aeromonas hydrophila. Garlic extract showed excellent antibacterial activity against all the test organisms, except L. monocytogenes. Nutmeg showed good anti-listerial activity, although activity against E. coli and Salmonella were serotype dependent. Both garlic and nutmeg extracts were effective against A. hydrophila. Extracts of ginger showed inhibitory activity against two serogroups of E. coli: as O8 (enterotoxigenic E. coli) and O88 only. Extracts of onion and pepper did not show any antibacterial activity against the test organisms.
Convergent synthesis of new N -substituted 2-5-(1H -indol-3-ylmethyl)-1,3,4-oxadiazol-2-ylsulfanylacetamides as suitable therapeutic agents
abstract A series of N-substituted 2-[5-(1H-indol-3-ylmethyl)-1,3,4-oxadiazol-2-yl]sulfanylacetamides (8a-w) was synthesized in three steps. The first step involved the sequential conversion of 2-(1H-indol-3-yl)acetic acid (1) to ester (2) followed by hydrazide (3) formation and finally cyclization in the presence of CS2 and alcoholic KOH yielded 5-(1H-indole-3-yl-methyl)-1,3,4-oxadiazole-2-thiol (4). In the second step, aryl/aralkyl amines (5a-w) were reacted with 2-bromoacetyl bromide (6) in basic medium to yield 2-bromo-N-substituted acetamides (7a-w). In the third step, these electrophiles (7a-w) were reacted with 4 to afford the target compounds (8a-w). Structural elucidation of all the synthesized derivatives was done by 1H-NMR, IR and EI-MS spectral techniques. Moreover, they were screened for antibacterial and hemolytic activity. Enzyme inhibition activity was well supported by molecular docking results, for example, compound 8q exhibited better inhibitory potential against α-glucosidase, while 8g and 8b exhibited comparatively better inhibition against butyrylcholinesterase and lipoxygenase, respectively. Similarly, compounds 8b and 8c showed very good antibacterial activity against Salmonella typhi, which was very close to that of ciprofloxacin, a standard antibiotic used in this study. 8c and 8l also showed very good antibacterial activity against Staphylococcus aureus as well. Almost all compounds showed very slight hemolytic activity, where 8p exhibited the least. Therefore, the molecules synthesized may have utility as suitable therapeutic agents. resumo Uma série de acetamidas 2-[5-(1H-indol-3-ilmetil)-1,3,4-oxadiazol-2-il]sulfanila N-substituídas (8a-w) foi sintetizada em três fases. A primeira etapa envolveu a conversão sequencial de ácido 2-(1H-indol-3-il)acético (1) a éster (2), seguido por hidrazida (3) e, finalmente, a e ciclização na presença de CS2 e KOH alcoólico produziu 5-(1H-indol-3-il- metil)-1,3,4-oxadiazole-2-tiol (4). Na segunda etapa, aminas arílicas/aralquílicas(5a-w) reagiram com brometo de 2-bromoacetila (6), em meio básico, para se obter acetamidas 2-bromo-N-substituídas (7a-w). Na terceira etapa, estes eletrófilos (7a- w) reagiram com 4, para se obter os compostos alvo (8a-w). A elucidação estrutural de todos os derivados sintetizados foi realizada por 1H-NMR, IR e técnicas de espectrometria de EI-MS. Além disso, eles foram submetidos a triagem de atividade antibacteriana e hemolítica. Análise da inibição enzimática foi bem apoiada pelos resultados de docking molecular. Por exemplo, o composto 8q exibiu melhor potencial inibitório contra α-glicosidase, e os compostos 8g e 8b exibiram, comparativamente, melhor inibição contra butirilcolinesterase (BChE) elipoxigenase (LOX), respectivamente. Do mesmo modo os compostos 8b e 8c mostraram excelente potencial antibacteriano contra SalmonellaTyphi, semelhante ao do ciprofloxacino, antibiótico padrão usado neste estudo. Os compostos 8c e 8l também mostraram excelente potencial antibacteriano contra Staphylococcus aureus . Quase todos os compostos mostraram pequena atividade hemolítica, sendo que o composto 8p apresentou menor atividade. Assim, as moléculas sintetizadas podem ter a sua utilidade como agentes terapêuticos adequados.
Antibacterial activity of extracts of six macroalgae from the northeastern brazilian coast
Hexane, chloroform and ethanol extracts of six marine macroalgae (Rhodophyta and Chlorophyta) from North Ceará coast (Northeast Brazil) were evaluated for antibacterial activity by the single disk method. Best results were shown by the hexane extracts of Amansia multifida against enteric Gram-negative strains such as Enterobacter aerogenes, Klebsiella pneumoniae, Pseudomonas aeruginosa, Salmonella typhi, S. cholerae-suis, Serratia marcescens, Vibrio cholerae and the Gram-positive bacteria Bacillus subtilis and Staphylococcus aureus. Extratos lipídicos de seis macroalgas (Rhodophyta e Chlorophyta) coletadas na costa do Estado do Ceará foram avaliadas para atividade antibacteriana pelo método de difusão em disco. Os extratos de Amansia multifida (obtidos a partir dos solventes orgânicos hexana, clorofórmio e etanol) demonstraram os melhores resultados contra as espécies entéricas Gram negativas de Enterobacter aerogenes, Klebsiella pneumoniae, Pseudomonas aeruginosa, Salmonella typhi, S. cholerae-suis, Serratia marcescens, Vibrio cholerae e bactérias Gram positivas Bacillus subtilis e Staphylococcus aureus.
