Molecular analyses of the Kalotermes dispar-complex

The Canary Islands is a Macaronesian volcanic archipelago with a depauperate community of three species of Kalotermitidae, including Kalotermes dispar. A total of 54 Kalotermes colonies were collected from Gran Canaria, Tenerife, La Gomera, La Palma, and El Hierro islands. Soldiers and imagos were morphologically examined and sequenced for four mitochondrial markers. Although morphological differences could not be detected, phylogenetic analysis of both cox1/tRNA/cox2 and rrnL markers revealed two distinct clades of K. dispar, suggesting cryptic diversity. The diversification within the Canary Kalotermes lineage most likely occurred around 75 Mya, while the divergence within the two clades was reconstructed at about 3.6 Mya and 1.9 Mya. Kalotermes approximatus from the southeastern Nearctic constitutes a sister to the Canary Kalotermes, while the Palearctic K. flavicollis, K. italicus, and K. phoenicae form a separate clade. It is hypothesized that a faunal exchange of Kalotermes from the Nearctic to the Canary Islands occurred via transoceanic rafting during the mid-Miocene.

(1) Background: Beetles in the superfamily Scarabaeoidea are one of the most important groups of Coleoptera. They are found in various ecosystems all over the world and belong to coprophagous, necrophagous, saproxylophagous, phyllophagous and rhizophagous types. The aim of the study is to describe the fauna and distribution of Scarabaeoidea in the Republic of Mordovia (central part of European Russia); (2) Methods: The study was conducted from 2003 to 2023. Collection material (specimens from 1972 and 1986) was also used. Specimens were collected using traditional Coleoptera collecting methods (manual collecting, light trap, collection of rotten remains and pitfall traps). For each observation, the coordinates, number of specimens and dates were recorded; (3) Results: The dataset contains 3198 occurrences. We examined 11,011 specimens of Scarabaeoidea. The dataset contains information on 88 species of Scarabaeoidea. Of these, five species (Aphodius pedellus, Nobius serotinus, Phaeaphodius rectus, Planolinus fasciatus and Onthophagus medius) are listed for the region for the first time. Another seven species are additionally known from others taken from the literature (but were not found during the present field survey); (4) Conclusions: Species diversity of Scarabaeoidea of Mordovia accounts for 95 species from 4 families (Geotrupidae, Trogidae, Lucanidae and Scarabaeidae). Ten species (Protaetia marmorata, Anoplotrupes stercorosus, Cetonia aurata, Protaetia cuprea volhyniensis, Oxythyrea funesta, Platycerus caraboides, Serica brunnea, Melolontha hippocastani, Trichius fasciatus and Protaetia fieberi) constitute the main population of Scarabaeoidea fauna given in the dataset. The species diversity of Scarabaeoidea of the Republic of Mordovia is roughly similar in number of species to that of neighboring or more northern regions.

Knockdown of Two Trehalase Genes by RNA Interference Is Lethal to the White-Backed Planthopper ISogatella furcifera/I

Trehalase (Tre) is a crucial enzyme involved in trehalose metabolism, and it plays pivotal roles in insect development and metamorphosis. However, the biological function of Tre genes in Sogatella furcifera remains unclear. In the present study, two Tre genes—SfTre1 and SfTre2—were cloned and identified based on the S. furcifera transcriptome data. Bioinformatic analysis revealed that the full-length complementary DNA of SfTre1 and SfTre2 genes were 3700 and 2757 bp long, with 1728- and 1902-bp open reading frame encoding 575 and 633 amino acid residues, respectively. Expression analysis indicated that SfTre1 and SfTre2 were expressed at all developmental stages, with the highest expression in day two adults. Furthermore, the highest expression levels of SfTre1 and SfTre2 were observed in the ovary; enriched expression was also noted in head tissues. The knockdown of SfTre1 and SfTre2 via injecting double-stranded RNAs decreased the transcription levels of the corresponding mRNAs and led to various malformed phenotypes and high lethality rates. The results of our present study indicate that SfTre1 and SfTre2 play crucial roles in S. furcifera growth and development, which can provide referable information for Tre genes as a potential target for planthopper control.
