How female directors, producers, and writers navigate the challenges and barriers facing female-driven projects at each stage of filmmaking in contemporary Hollywood. The Impact of COVID-19 Crisis upon the Consumer Buying Behavior of Fresh Vegetables Directly from Local Producers. Case Study: The Quarantined Area of Suceava County, Romania
The present paper intends to address the impact of COVID-19 crisis upon the consumer buying behavior of fresh vegetables directly from local producers as observed 30 days later, after enforcing the state of emergency in Romania within a well-defined area, namely, the quarantined area of Suceava. The study relies on the interpretation of answers received from the quarantined area (N = 257) to a questionnaire applied online nationwide. The starting point of this paper is the analysis of the sociodemographic factors on the purchasing decision of fresh vegetables directly from local producers before declaring the state of emergency in Romania (16 March 2020). Further research has been conducted by interpreting the changes triggered by the COVID-19 crisis on the purchasing intention of such products before and after the end of the respective crisis. The aim of this scientific investigation relies on identifying the methods by which these behavioral changes can influence the digital transformation of short food supply chains.
Designer exosomes produced by implanted cells intracerebrally deliver therapeutic cargo for Parkinson’s disease treatment
Exosomes are cell-derived nanovesicles (50–150 nm), which mediate intercellular communication, and are candidate therapeutic agents. However, inefficiency of exosomal message transfer, such as mRNA, and lack of methods to create designer exosomes have hampered their development into therapeutic interventions. Here, we report a set of EXOsomal transfer into cells (EXOtic) devices that enable efficient, customizable production of designer exosomes in engineered mammalian cells. These genetically encoded devices in exosome producer cells enhance exosome production, specific mRNA packaging, and delivery of the mRNA into the cytosol of target cells, enabling efficient cell-to-cell communication without the need to concentrate exosomes. Further, engineered producer cells implanted in living mice could consistently deliver cargo mRNA to the brain. Therapeutic catalase mRNA delivery by designer exosomes attenuated neurotoxicity and neuroinflammation in in vitro and in vivo models of Parkinson’s disease, indicating the potential usefulness of the EXOtic devices for RNA delivery-based therapeutic applications. Exosomes function as intercellular information transmitters and are candidates for delivery of therapeutic agents. Here the authors present EXOtic, a synthetic biology device for in-situ production of designer exosomes and demonstrate in vivo application in models of Parkinson's disease.
Active Starvation Responses Mediate Antibiotic Tolerance in Biofilms and Nutrient-Limited Bacteria
Bacteria become highly tolerant to antibiotics when nutrients are limited. The inactivity of antibiotic targets caused by starvation-induced growth arrest is thought to be a key mechanism producing tolerance. Here we show that the antibiotic tolerance of nutrient-limited and biofilm Pseudomonas aeruginosa is mediated by active responses to starvation, rather than by the passive effects of growth arrest. The protective mechanism is controlled by the starvation-signaling stringent response (SR), and our experiments link SR-mediated tolerance to reduced levels of oxidant stress in bacterial cells. Furthermore, inactivating this protective mechanism sensitized biofilms by several orders of magnitude to four different classes of antibiotics and markedly enhanced the efficacy of antibiotic treatment in experimental infections.
Beyond the spotlight: Unveiling the gender bias curtain in movie reviews
Historically, the film industry has been male-dominated both in front of and behind the camera, resulting in a longstanding gender imbalance in storytelling and representation. This legacy of male-centric narratives may unconsciously influence critics’ expectations and judgments. Existing literature suggests that negative critiques in movie reviews can significantly impact actors’ earnings by diminishing a film’s commercial prospects. This influence extends to potential reductions in back-end compensation and marketability for future projects, and it can directly affect actors’ well-being, leading to increased stress levels and elevated cardiovascular reactivity. Previously, gender biases in movie reviews were computed using disparities in male-led versus female-led movie ratings or box-office earnings; however, no work has been done to quantify the linguistic biases within movie review transcripts. This work aims to leverage our language-model-powered gender bias detection system to measure benevolent sexism, hostile sexism, explicit marking of sex, dehumanization, and generic pronouns in reviews published by professional critics. Therefore, we analyzed 17,165 professionally written reviews, comprising a total of 735,000 sentences. Our analysis uncovers pronounced representation bias in key movie roles, with 72% of first actors, 91% of first directors, and 86% of first writers being male. More importantly, the findings indicate that movies with female-dominated casts evoke, on average, 149% higher magnitude of hostile sexism and 44% higher magnitude of benevolent sexism in their reviews compared to movies with male-dominated casts. We also find that benevolent sexism is most common in Family and Music genres, reflecting the romanticization of gender roles, whereas hostile sexism peaks in Romance. A non-parametric statistical analysis revealed significant gender differences in benevolent and hostile sexism scores for movie reviews, with female first actors, directors, and writers receiving higher levels of both benevolent and hostile sexist criticism compared to their male counterparts.
Individual EEG measures of attention, memory, and motivation predict population level TV viewership and Twitter engagement
Television (TV) programming attracts ever-growing audiences and dominates the cultural zeitgeist. Viewership and social media engagement have become standard indices of programming success. However, accurately predicting individual episode success or future show performance using traditional metrics remains a challenge. Here we examine whether TV viewership and Twitter activity can be predicted using electroencephalography (EEG) measures, which are less affected by reporting biases and which are commonly associated with different cognitive processes. 331 participants watched an hour-long episode from one of nine prime-time shows (~36 participants per episode). Three frequency-based measures were extracted: fronto-central alpha/beta asymmetry (indexing approach motivation), fronto-central alpha/theta power (indexing attention), and fronto-central theta/gamma power (indexing memory processing). All three EEG measures and the composite EEG score significantly correlated across episode segments with the two behavioral measures of TV viewership and Twitter volume. EEG measures explained more variance than either of the behavioral metrics and mediated the relationship between the two. Attentional focus was integral for both audience retention and Twitter activity, while emotional motivation was specifically linked with social engagement and program segments with high TV viewership. These findings highlight the viability of using EEG measures to predict success of TV programming and identify cognitive processes that contribute to audience engagement with television shows.
Biofilm formation and antibiotic resistance of Klebsiella pneumoniae isolated from clinical samples in a tertiary care hospital, Klaten, Indonesia
Background Klebsiella pneumoniae (K. pneumoniae) is a common cause of health-care associated infections (HAIs) and has high levels of antibiotic resistance. These bacteria are well-known for their ability to produce biofilm. The purpose of this study was to identify the antibiotic resistance pattern and biofilm-producing capacity of K. pneumoniae isolated from clinical samples in a tertiary care hospital in Klaten, Indonesia. Methods K. pneumoniae was isolated from inpatients in Soeradji Tirtonegoro Hospital Klaten from June 2017 to May 2018. Identification of K. pneumoniae isolate was done by analyzing colony morphology, microscopic examination, and by performing biochemical testing. Testing of antibiotics susceptibility and biofilm-producing capacity used the Kirby-Bauer disk diffusion method and adherence quantitative assays, respectively. Results A total of 167 (17.36%) K. pneumoniae isolates were isolated from 962 total clinical bacterial isolates during the study. Most of them were collected from patients aged more than 60 years old and were mainly obtained from respiratory specimens (51.50%). Most of K. pneumoniae isolates were extensively resistant to antibiotics. A more favorable profile was found only towards meropenem, amikacin, and piperacillin-tazobactam, showing 1.20%; 4.79% and 10.53% of resistance, respectively. The overall proportion of multidrug-resistant K. pneumoniae isolates was 54.49%. In addition, 148 (85.63%) isolates were biofilm producers, with 45 (26.95%) isolates as strong, 48 (28.74%) isolates as moderate, and 50 (29.94%) isolates as weak biofilm producers. Conclusion Most of the K. pneumoniae isolates demonstrated resistance to a wide range of antibiotics and are biofilm producers.
