The social life of Norway rats (Rattus norvegicus)
The Norway rat has important impacts on our life. They are amongst the most used research subjects, resulting in ground-breaking advances. At the same time, wild rats live in close association with us, leading to various adverse interactions. In face of this relevance, it is surprising how little is known about their natural behaviour. While recent laboratory studies revealed their complex social skills, little is known about their social behaviour in the wild. An integration of these different scientific approaches is crucial to understand their social life, which will enable us to design more valid research paradigms, develop more effective management strategies, and to provide better welfare standards. Hence, I first summarise the literature on their natural social behaviour. Second, I provide an overview of recent developments concerning their social cognition. Third, I illustrate why an integration of these areas would be beneficial to optimise our interactions with them.
Neurobiology of rodent self-grooming and its value for translational neuroscience
Key Points Self-grooming is an evolutionarily conserved complex innate behaviour that has a role in hygiene maintenance and other physiological functions. Self-grooming is the most frequently occurring awake behaviour in laboratory rodents. Self-grooming is an important phenotype to study in translational neuroscience, as it may allow the modelling of human diseases that have symptoms similar to, and/or share pathogenetic mechanisms with, aberrant grooming in rodents. Analysing animal self-grooming also has a broader value in the study of neurobiology underlying complex repetitive behaviours, which may be disrupted in certain neurological diseases. In this Review, we discuss the neurobiology of grooming, including its underlying circuitry, genetic mechanisms and pharmacological modulation. We also highlight studies of rodent self-grooming behaviour in models of neuropsychiatric disorders that suggest that it is valuable asset for clinical and translational neuroscience research, including the identification of neural circuits that control complex patterned behaviours. These findings suggest that the study of rodent self-grooming has multiple implications for translational neuroscience, which may extend beyond understanding the self-grooming behaviour itself. Rodents spend a large proportion of their waking time engaged in self-grooming behaviour. In this Review, Kalueff and colleagues describe the characteristics and underlying neural circuitry of rodent self-grooming, and discuss its use as a measure of repetitive behaviour in models of psychiatric disease. Self-grooming is a complex innate behaviour with an evolutionarily conserved sequencing pattern and is one of the most frequently performed behavioural activities in rodents. In this Review, we discuss the neurobiology of rodent self-grooming, and we highlight studies of rodent models of neuropsychiatric disorders — including models of autism spectrum disorder and obsessive compulsive disorder — that have assessed self-grooming phenotypes. We suggest that rodent self-grooming may be a useful measure of repetitive behaviour in such models, and therefore of value to translational psychiatry. Assessment of rodent self-grooming may also be useful for understanding the neural circuits that are involved in complex sequential patterns of action.
Maternal glyphosate exposure causes autism-like behaviors in offspring through increased expression of soluble epoxide hydrolase
Epidemiological studies suggest that exposure to herbicides during pregnancy might increase risk for autism spectrum disorder (ASD) in offspring. However, the precise mechanisms underlying the risk of ASD by herbicides such as glyphosate remain unclear. Soluble epoxide hydrolase (sEH) in the metabolism of polyunsaturated fatty acids is shown to play a key role in the development of ASD in offspring after maternal immune activation. Here, we found ASD-like behavioral abnormalities in juvenile offspring after maternal exposure to high levels of formulated glyphosate. Furthermore, we found higher levels of sEH in the prefrontal cortex (PFC), hippocampus, and striatum of juvenile offspring, and oxylipin analysis showed decreased levels of epoxy-fatty acids such as 8 (9)-EpETrE in the blood, PFC, hippocampus, and striatum of juvenile offspring after maternal glyphosate exposure, supporting increased activity of sEH in the offspring. Moreover, we found abnormal composition of gut microbiota and short-chain fatty acids in fecal samples of juvenile offspring after maternal glyphosate exposure. Interestingly, oral administration of TPPU (an sEH inhibitor) to pregnant mothers from E5 to P21 prevented ASD-like behaviors such as social interaction deficits and increased grooming time in the juvenile offspring after maternal glyphosate exposure. These findings suggest that maternal exposure to high levels of glyphosate causes ASD-like behavioral abnormalities and abnormal composition of gut microbiota in juvenile offspring, and that increased activity of sEH might play a role in ASD-like behaviors in offspring after maternal glyphosate exposure. Therefore, sEH may represent a target for ASD in offspring after maternal stress from occupational exposure to contaminants.
Increased alertness and moderate ingroup cohesion in bonobos’ response to outgroup cues
In a number of species, including humans, perceived outgroup threat can promote ingroup cohesion. However, the distribution and selection history of this association across species with varied intergroup relations remains unclear. Using a sample of 8 captive groups (N = 43 individuals), we here tested whether bonobos, like chimpanzees, show more affiliative ingroup behaviour following perception of outgroup cues (unfamiliar male long-distance vocalisations). We used comparable methods to our previous study of captive chimpanzees, and found that, although weaker, there was an association for more frequent social grooming in response to the outgroup condition than the control condition, alongside more alert posture and increased self-directed behaviour. This provides preliminary evidence for an ancestral origin to the proximate association between outgroup cues and ingroup cohesion, at least prior to the Pan-Homo split, and suggests the presence of intergroup competition in our last common ancestor.
Repeated Cortico-Striatal Stimulation Generates Persistent OCD-Like Behavior
Although cortico-striato-thalamo-cortical (CSTC) circuit dysregulation is correlated with obsessive compulsive disorder (OCD), causation cannot be tested in humans. We used optogenetics in mice to simulate CSTC hyperactivation observed in OCD patients. Whereas acute orbitofrontal cortex (OFC)-ventromedial striatum (VMS) stimulation did not produce repetitive behaviors, repeated hyperactivation over multiple days generated a progressive increase in grooming, a mouse behavior related to OCD. Increased grooming persisted for 2 weeks after stimulation cessation. The grooming increase was temporally coupled with a progressive increase in light-evoked firing of postsynaptic VMS cells. Both increased grooming and evoked firing were reversed by chronic fluoxetine, a first-line OCD treatment. Brief but repeated episodes of abnormal circuit activity may thus set the stage for the development of persistent psychopathology.
Acute Behavioral Effects of Regrouping Dairy Cows
Regrouping is a common management practice on commercial dairy farms. The objectives of this study were to examine the effects of regrouping on feeding behavior, social behavior, and milk production. Eleven mid-lactation Holstein cows were individually introduced into previously established social groups. Behavior and milk production were monitored from 3 d before until 3 d after regrouping. Cows were fed a total mixed ration ad libitum twice daily. Time-lapse video was used to quantify feeding and social behavior. Cows spent approximately 15min less time eating in the first hour following regrouping compared with the 3 d before regrouping. Cows were displaced from the feeding area on average 10 times/d before regrouping, but this value increased to more than 25 times (±2.6) on the day of mixing and gradually declined on the days following. The number of lying bouts declined from 12.2±0.9 to 10.5±0.9 on the day of regrouping; lying time showed a similar tendency. Cows initiated, on average, 7.5±1.3 allogrooming events/d over the 3 d before regrouping but this declined to 1.3±1.3 events on the day of mixing and remained lower than baseline for the following 3 d. Milk production declined from 43.4±1.5 kg/d to 39.7±1.5 kg/d on the day of regrouping, but did not differ from premixing levels on subsequent days. This study shows that regrouping can disrupt behavior and production in the hours and days following regrouping and suggests the need for future research to identify management changes that reduce these effects.
Familiarity modulates social approach toward stressed conspecifics in female rats
Familiarity between conspecifics may influence how social affective cues shape social behaviors. In a social affective preference test, experimental rats, when given the choice to explore an unfamiliar stressed or a naive adult, will avoid interaction with a stressed conspecific. To determine if familiarity would influence social interactions with stressed conspecifics, male and female test rats underwent 2 social affective preference tests in isosexual triads where an experimental rat was presented with a naïve and a stressed target conspecific who were either familiar (cagemate) or unfamiliar. Male and female experimental rats avoided stressed unfamiliar conspecifics. However, experimental female rats demonstrated a preference to interact with their stressed, familiar cagemates. Male and female rats exhibited more self-grooming and immobility behavior in the presence of stressed conspecifics, which may indicate emotion contagion. These findings suggest a sex-specific role of familiarity in social approach and avoidance, and warrant further mechanistic exploration.
Refining animal care through technology: Addressing alopecia in Jaculus jaculus with validated computer vision analysis
We validated the use of an open-source computer vision toolkit to analyze high-quality behavioral data and evaluate welfare in the Lesser Egyptian Jerboa ( Jaculus jaculus ). Movements of these small, nocturnal rodents are rapid and difficult to observe, potentially obscuring behavioral assessment. However, assessment became warranted when alopecia and jumping were noted. We compared trained human observers to machine learning trained computer vision algorithms, evaluating accuracy and precision in behavioral classification. Human observers categorized behaviors with an overall accuracy of 0.71 ± 0.11 and an intraclass correlation coefficient (ICC) of 0.92 ± 0.07, with greater odds of misidentifying behaviors lasting less than one second. Computer vision classifiers successfully met human-grade accuracy and ICC, with significantly less sensitivity to behavioral duration. As 34% of manually classified behaviors lasted less than 0.5 seconds, we used computer vision to annotate activity budgets of captive jerboas before and after adding novel enrichment. Alopecia was significantly associated with grooming, and while grooming was negatively associated with terrarium height and with opaque dividers between terraria, conventional rodent enrichment had no significant effect on behavior. Inflammatory causes of alopecia were not found with cytologic, molecular, or histopathologic analysis. These results suggest captive jerboa may demonstrate psychogenic alopecia. Furthermore, computer vision automation allows for fast, accurate analysis of large volumes of behavioral data that can be used to tailor species-specific husbandry practices and improve animal welfare.
Low fertility, fecundity and numbers of mated female offspring explain the lower reproductive success of the parasitic mite Varroa destructor in African honeybees
Although Varroa destructor is the most serious ecto-parasite to the honeybee, Apis mellifera L., some honeybee populations such as Apis mellifera scutellata in Kenya can survive mite infestations without treatment. Previously, we reported that grooming behaviour could be a potential tolerant mechanism expressed by this honeybee subspecies towards mite infestation. However, both hygienic and grooming behaviours could not explain the lower mite-infestation levels recorded in these colonies. Here, we investigated the involvement of other potential resistant mechanisms including suppression of mite reproduction in worker brood cells of A. m. scutellata to explain the low mite numbers in their colonies. High infertility rates (26–27%) and percentages of unmated female offspring (39–58%) as well as low fecundity (1.7–2.2, average offspring produced) were identified as key parameters that seem to interact with one another during different seasons to suppress mite reproduction in A. m. scutellata colonies. We also identified offspring mortality in both sexes and absence of male offspring as key factors accounting for the low numbers of mated daughter mites produced in A. m. scutellata colonies. These results suggest that reduced mite reproductive success could explain the slow mite population growth in A. m. scutellata colonies.
Dog Behavior Co-Varies with Height, Bodyweight and Skull Shape
Dogs offer unique opportunities to study correlations between morphology and behavior because skull shapes and body shape are so diverse among breeds. Several studies have shown relationships between canine cephalic index (CI: the ratio of skull width to skull length) and neural architecture. Data on the CI of adult, show-quality dogs (six males and six females) were sourced in Australia along with existing data on the breeds' height, bodyweight and related to data on 36 behavioral traits of companion dogs (n = 8,301) of various common breeds (n = 49) collected internationally using the Canine Behavioral Assessment and Research Questionnaire (C-BARQ). Stepwise backward elimination regressions revealed that, across the breeds, 33 behavioral traits all but one of which are undesirable in companion animals correlated with either height alone (n = 14), bodyweight alone (n = 5), CI alone (n = 3), bodyweight-and-skull shape combined (n = 2), height-and-skull shape combined (n = 3) or height-and-bodyweight combined (n = 6). For example, breed average height showed strongly significant inverse relationships (p<0.001) with mounting persons or objects, touch sensitivity, urination when left alone, dog-directed fear, separation-related problems, non-social fear, defecation when left alone, owner-directed aggression, begging for food, urine marking and attachment/attention-seeking, while bodyweight showed strongly significant inverse relationships (p<0.001) with excitability and being reported as hyperactive. Apart from trainability, all regression coefficients with height were negative indicating that, across the breeds, behavior becomes more problematic as height decreases. Allogrooming increased strongly (p<0.001) with CI and inversely with height. CI alone showed a strong significant positive relationship with self-grooming (p<0.001) but a negative relationship with chasing (p = 0.020). The current study demonstrates how aspects of CI (and therefore brain shape), bodyweight and height co-vary with behavior. The biological basis for, and significance of, these associations remain to be determined.
