EFFECT OF REPLACING SOYBEAN MEAL WITH LARVAE MEAL OF BLACK SOLDIER FLY HERMETIA ILLUCENS IN GROWTH PERFORMANCE AND SOME BIOCHEMICAL BLOOD PARAMETERS OF COMMON CARP CYPRINUS CARPIO L
This study was conducted at the College of Agriculture/ Diyala University/ Department of Animal production to determine the effect of replacing soybean meal with black soldier fly Hermetia illucens larvae meal on the growth performance and some biochemical blood parameters of common carp Cyprinus carpio L. One hundred and five fish with an average initial weight of 27.46±0.14 g was used in this experiment. They were randomly distributed into five treatments (3 replicate/treatment, 7 fish/ replicate). The first treatments (T1 control group) was fed with standard diet without black soldier fly larvae meal (0% BSFLM), black soldier fly larvae meal were substituted for soybean meal in other experimental groups at 25%, 50%, 75% and 100% (T2, T3, T4 and T5), respectively. After 12 weeks of feeding, no significant differences (P>0.05) were found in feed conversion ratio (FCR) and protein efficiency ratio (PER) between control and experimental groups. However, T4 (75% BSFLM) showed significantly improved in mean of final body weight (FBW) and weight gain (WG) compared to the control and other experimental groups. The results of serum biochemical indices showed no statistically significant differences (P>0.05) in the levels of ALT (Alanine aminotransferase), Glucose, Triglycerides, LDL (low-density lipoprotein) and total protein (TP) between control and experimental groups. While AST (Aspartate aminotransferase) level of T4 and T5 was significantly higher (P≤0.05) than T3. As a result, the study suggests that black soldier fly larval meal can be a sustainable protein alternative to soybean meal at 75% without affecting growth performance or blood biochemical characteristics.   
Effect of dietary threonine on growth performance and muscle growth, protein synthesis and antioxidant-related signalling pathways of hybrid catfish Pelteobagrus vachelli ♀ × Leiocassis longirostris
The experiment was conducted to investigate the effects of dietary threonine (Thr) on growth performance and muscle growth, protein synthesis and antioxidant-related signalling pathways of hybrid catfish Pelteobagrus vachelli ♀ × Leiocassis longirostris ♂. A total of 1200 fish (14·19 ( se 0·13) g) were randomly distributed into six groups with four replicates each, fed six diets with graded level of Thr (9·5, 11·5, 13·5, 15·4, 17·4 and 19·3 g/kg diets) for 56 d. Results showed ( P < 0·05) that dietary Thr (1) increased percentage weight gain, specific growth rate, feed efficiency and protein efficiency ratio; (2) up-regulated growth hormone (GH), insulin-like growth factor 1 (IGF-1), proliferating cell nuclear antigen, myogenic regulation factors (MyoD, Myf5, MyoG and Mrf4) and myosin heavy chain (MyHC) mRNA levels; (3) increased muscle protein content via regulating the protein kinase B/target of rapamycin signalling pathway and (4) decreased malondialdehyde and protein carbonyl contents, increased catalase, glutathione-S-transferase, glutathione reductase and GSH activities, up-regulated mRNA levels of antioxidant enzymes related to NFE2-related factor 2 and γ -glutamylcysteine ligase catalytic subunit. These results suggest that Thr has a potential role to improve muscle growth and protein synthesis, which might be due to the regulation of GH-IGF system, muscle growth-related gene, antioxidative capacity and protein synthesis-related signalling pathways. Based on the quadratic regression analysis of specific growth rate, the Thr requirement of hybrid catfish (14·19–25·77 g) was estimated to be 13·77 g/kg of the diet (33·40 g/kg of dietary protein).
High dietary lipid level alters the growth, hepatic metabolism enzyme, and anti-oxidative capacity in juvenile largemouth bass Micropterus salmoides
The present study was conducted to investigate the effects of high dietary lipid levels on growth, metabolism, antioxidant capacity, and immune responses of largemouth bass. Fish (initial body weight 13.38 ± 0.11 g) were fed three isonitrogenous semi-purified diets containing 5%, 10%, and 20% lipid, respectively. The results indicated that fish fed 10% lipid diet showed significantly better final body weight, specific growth rate (SGR), protein efficiency ratio (PER), and feed conversion ratio (FCR) compared with that fed 5% lipid diet. Meanwhile, fish fed 20% lipid diet had a significantly higher viscera ratio (VR), hepatosomatic index (HSI), intraperitoneal fat ratio (IPF), and liver lipid content than those fed the other diets. Higher alanine aminotransferase (ALT) and aspartate transaminase (AST) activities, total cholesterol (TC), triglyceride (TG), free fatty acids (FFA), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C) contents, and LDL-C/HDL-C value in plasma were recorded in fish fed 20% lipid diet, while higher insulin contents were obtained in fish fed 5% lipid diet. In addition, the highest carnitine palmitoyltransferase I (CPT1), AMP-activated protein kinase (AMPK), fructose-1,6-bisphosphatase (FBPase), and phosphoenolpyruvate carboxykinase (PEPCK) activities in the liver were also observed in fish fed 20% lipid diet. However, fish fed 20% lipid diet had a significantly lower superoxide dismutase (SOD) and catalase (CAT) activities and higher MDA contents in liver than those fed the other diets. The higher nitric oxide (NO) contents and inducible nitric oxide synthase (iNOS) activity in liver were recorded in fish fed 10% lipid diet. Moreover, the alkaline phosphatase (ALP), inducible nitric oxide synthase (iNOS) and lysozyme activities, and nitric oxide (NO) contents in plasma were higher in fish fed the 10% diets than the other groups. In conclusion, high dietary lipid levels could suppress growth performance and liver anti-oxidative capacity, and reduce immune responses of largemouth bass.
Multi-criteria assessment of pea protein quality in rats: a comparison between casein, gluten and pea protein alone or supplemented with methionine
The objective of this study was to assess the nutritional quality of pea protein isolate in rats and to evaluate the impact of methionine (Met) supplementation. Several protein diets were studied: pea protein, casein, gluten, pea protein–gluten combination and pea protein supplemented with Met. Study 1: Young male Wistar rats (n 8/group) were fed the test diets ad libitum for 28 d. The protein efficiency ratio (PER) was measured. Study 2: Adult male Wistar rats (n 9/group) were fed the test diets for 10 d. A protein-free diet group was used to determine endogenous losses of N. The rats were placed in metabolism cages for 3 d to assess N balance, true faecal N digestibility and to calculate the Protein Digestible-Corrected Amino Acid Score (PDCAAS). They were then given a calibrated meal and euthanised 6 h later for collection of digestive contents. The true caecal amino acid (AA) digestibility was determined, and the Digestible Indispensable Amino Acid Score (DIAAS) was calculated. Met supplementation increased the PER of pea protein (2·52 v. 1·14, P < 0·001) up to the PER of casein (2·55). Mean true caecal AA digestibility was 94 % for pea protein. The DIAAS was 0·88 for pea protein and 1·10 with Met supplementation, 1·29 for casein and 0·25 for gluten. Pea protein was highly digestible in rats under our experimental conditions, and Met supplementation enabled generation of a mixture that had a protein quality that was not different from that of casein.
Effects of different carbon to nitrogen ratios in the biofloc system on water quality, growth, and body composition of common carp (Cyprinus carpio L.) fingerlings
Effects of different carbon to nitrogen ratios in the biofloc system with sugarcane molasses as organic carbon source on water quality, growth performance, and body composition of common carp (Cyprinus carpio L.) fingerlings were investigated in a 90-day trial. A total of 450 fish (14.17 ± 0.36 g) were fed twice daily with commercial diet at 5% of their body weight under a 12:12 light:dark regime and cultured in 1 control (without extra carbon source) and 4 experimental treatments (C/N: 11:1, 15:1, 19:1, and 23:1) with 3 replicates at a density of 1.06 kg m−3 per tank. Based on the results, the water electrical conductivity (EC) and total dissolved solids (TDS) (measured daily), flocs volume (FV), and total suspended solids (TSS) (measured once a week) were increased from C/N 11:1 to C/N 23:1. The variation patterns of the average levels of total ammonia nitrogen, nitrite, and nitrate (measured once a week) were similar and showed a decreasing trend with an increase in C/N ratio. Total heterotrophic bacteria was also positively affected by increasing C/N to 23:1. The highest fish final weight (59.36 ± 0.24 g), weight gain (45.21 ± 0.47 g), and protein efficiency ratio (PER, 1.77 ± 0.04) were recorded in C/N 19:1. The lowest (1.46 ± 0.03) and highest (1.51 ± 0.05) feed conversion ratio (FCR) and the highest (1.60 ± 0.03%/day) and lowest (1.49 ± 0.01%/day) specific growth rate (SGR) were observed in C/N 19:1 and the control group, respectively (P < 0.05). By increasing the C/N to 23:1, FCR increased and the feed intake, final weight, weight gain, PER, and SGR decreased. The highest crude protein content (77.31 ± 0.43 %DW) and the lowest crude lipid content (12.13 ± 0.32 %DW) of the fish carcass were observed in C/N 19:1 while these values for flocs were not significantly affected by C/N ratio. Overall, the biofloc system with C/N 19:1 improved the water quality and growth performance of common carp while not negatively affecting the carcass proximate analysis.
Dietary valine levels affect growth, protein utilisation, immunity and antioxidant status in juvenile hybrid grouper (Epinephelus fuscoguttatus ♀ × Epinephelus lanceolatus ♂)
A 6-week growth trial was conducted to evaluate the influences of dietary valine (Val) levels on growth, protein utilisation, immunity, antioxidant status and gut micromorphology of juvenile hybrid groupers. Seven isoenergetic, isoproteic and isolipidic diets were formulated to contain graded Val levels (1·21, 1·32, 1·45, 1·58, 1·69, 1·82 and 1·94 %, DM basis). Each experimental diet was hand-fed to triplicate groups of twelve hybrid grouper juveniles. Results showed that weight gain percentage (WG%), protein productive value, protein efficiency ratio, and feed efficiency were increased as dietary Val level increased, reaching a peak value at 1·58 % dietary Val. The quadratic regression analysis of WG% against dietary Val levels indicated that the optimum dietary Val requirement for hybrid groupers was estimated to be 1·56 %. Gut micromorphology and expression of growth hormone in pituitary, insulin-like growth factor 1, target of rapamycin and S6 kinase 1 in liver were significantly affected by dietary Val levels. In serum, fish fed 1·58 % dietary Val had higher superoxide dismutase, catalase, lysozyme activities and IgM concentrations than fish fed other dietary Val levels. Fish fed 1·58 % dietary Val had higher expression of NF-E2-related factor 2 in head kidney than fish fed other dietary Val levels. Generally, the optimum dietary Val requirement for maximal growth of hybrid groupers was estimated to be 1·56 % of DM, corresponding to 3·16 % of dietary protein, and dietary Val levels affected growth, protein utilisation, immunity and antioxidant status in hybrid groupers.
Dietary arginine affects growth, gut morphology, oxidation resistance and immunity of hybrid grouper (Epinephelus fuscoguttatus♀×Epinephelus lanceolatus♂) juveniles
An 8-week growth trial was conducted to evaluate the effects of dietary arginine (Arg) levels on growth, gut morphology, oxidation resistance and immunity of hybrid grouper (Epinephelus fuscoguttatus♀×Epinephelus lanceolatus♂) juveniles. Seven isoenergetic (1465 kJ (350 kcal)/100-g DM), isoproteic (53·5 % of DM) and isolipidic (7 % of DM) experimental diets were formulated to contain graded Arg levels ranging from 1·9 to 4·7 % (dry weight) at approximately 0·5 % increments. Each diet was randomly assigned to triplicate groups of 16 juvenile fish (average initial body weight: 11·7 (sd 0·1) g) and was administered twice daily (08.00 and 16.00 hours). After the growth trial, all remaining fish were fed their prescribed diets for 2 d and then exposed to 4·5 mg Cu2+/l water for 36 h. Results showed that growth performance and feed utilisation of experimental fish were significantly affected by different dietary Arg levels. Weight gain % (WG%) of fish was increased as dietary Arg increased, reaching a peak value at 3·8 % dietary Arg level, and when dietary Arg level increased to 4·7 % WG% was reduced. Fish fed 1·9 and 2·2 % dietary Arg levels had higher daily feed intake compared with fish fed other dietary Arg levels. Feed conversion ratios in fish fed 1·9, 2·2, 2·7 and 4·7 % dietary Arg levels were higher than those in fish fed 3·1, 3·8 and 4·1 % dietary Arg levels. Protein efficiency ratio and protein productive value (PPV) increased with an increase in dietary Arg, up to a peak value at 3·8 % dietary Arg level, above which these parameters declined. On the basis of quadratic regression analysis of weight gain % (WG%) or PPV against dietary Arg levels, the optimal dietary Arg requirement for hybrid grouper was estimated to be 3·65 %. Fish fed 3·8 % dietary Arg had higher whole-body and muscle protein contents compared with fish fed other dietary Arg levels. Fish fed 3·8 and 4·1 % dietary Arg levels had higher levels of mRNA for insulin-like growth factor-I and target of rapamycin in the liver compared with fish fed other dietary Arg levels. Hepatic S6 kinase 1 mRNA expression in fish fed 3·8 % dietary Arg level was higher than that in fish fed any of the other dietary Arg levels. Gut morphology, hepatic antioxidant indices and immune indices in serum and head kidney were significantly influenced by dietary Arg levels. In conclusion, the optimal dietary Arg requirement for hybrid grouper was estimated to be 3·65 %, and suitable dietary Arg supplementations improved gut morphology and oxidation resistance of hybrid grouper.
Chromium Supplementation in Diet Enhances Growth and Feed Utilization of Striped Catfish (Pangasianodon hypophthalmus)
Chromium (Cr) is a trace element and plays a significant role in fish nutrition and physiology. An experiment was designed to know the effects of Cr addition in the diets to growth and feed utilization in striped catfish (Pangasianodon hypophthalmus). Four diets with Cr (0, 2, 4, and 8 mg kg−1) were fed to striped catfish in aquaria with triplicate groups for 60 days. Survival, growth parameters (weight gain, WG; %WG; specific growth rate, SGR), and feed utilization (feed intake, FI; feed efficiency, FE; protein efficiency ratio, PER; feed conversion ratio, FCR) were calculated at the end of the feeding trial. Several hemato-biochemical parameters, such as hemoglobin (Hb), red blood cell (RBC), white blood cell (WBC) and glucose level, and frequency of micronucleus (MN) formation in erythrocytes, were analyzed. The growth parameters (WG, %WG, and SGR) and feed utilization (FE and PER) increased significantly in the fish fed with 2 and 4 mg kg−1 Cr supplemented diets. On the other hand, the growth parameters suppressed in the fish fed with 8 mg kg−1 Cr-based diet. The polynomial regression analysis based on WG showed that 2.82 mg kg−1 Cr supplementation in the diet is optimum for the tested fish species. The values of Hb (g/dL), RBC (×106/mm3) and blood glucose (mg/dL) significantly decreased in the fish fed with the highest (8 mg kg−1) Cr-based feed. Conversely, MN frequency was significantly increased in the fish fed with 8 mg kg−1 Cr-based diet. Overall, 2.82 mg Cr kg−1 can be added to the diets of striped catfish for its better growth with maximum utilization of feed.
Determination of the protein quality of cooked Canadian pulses
A study to determine the protein digestibility‐corrected amino acid score and protein efficiency ratio of nine different cooked Canadian pulse classes was conducted in support of the establishment of protein quality claims in Canada and the United States. Split green and yellow pea, whole green lentil, split red lentil, Kabuli chickpea, navy bean, pinto bean, light red kidney bean, and black bean were investigated. Protein digestibility‐corrected amino acid score (PDCAAS) and the protein efficiency ratio (PER) were determined using the appropriate rodent models. All pulses had high digestibility values, >70%, with PDCAAS values greater than 0.5, thereby qualifying as a quality protein in the United States, but only navy beans qualified as a good source of protein. All pulses except whole green lentils, split red lentils, and split green peas would qualify as sources of protein with protein ratings between 20 and 30.4 in Canada. These findings support the use of pulses as protein sources in the regulatory context of both the United States and Canada. Pulses are consumed globally and are high in protein, but have to be cooked prior to consumption. This article describes the effect of cooking on the protein digestibility and amino acid composition of 9 pulse classes.
Effects of bacillus subtilis wb60 and lactococcus lactis on growth, immune responses, histology and gene expression in nile tilapia, oreochromis niloticus
An eight-week feeding trial was conducted to evaluate the effects of a basal control diet (CON), Bacillus subtilis at 107 (BS7) and at 108 CFU/g diet (BS8), Lactococcus lactis at 107 CFU/g (LL7) and at 108 CFU/g diet (LL8), and oxytetracycline (OTC) at 4 g/kg diet on Nile tilapia. Fish with initial body weight of 2.83 ± 0.05 g (mean ± SD) were fed two times a day. Weight gain, specific growth rate, feed efficiency, protein efficiency ratio and lysozyme activity of fish fed BS8, LL8 and LL7 diets were significantly higher than those of fish fed CON diet (p < 0.05). Superoxide dismutase and myeloperoxidase activity of fish fed BS8, LL8, BS7, LL7 and OTC diets were significantly higher than those of fish fed CON diet. Intestinal villi length and muscular layer thickness of fish fed BS8, LL8 and LL7 diets were significantly higher than those of fish fed CON and OTC diets. Also, heat shock protein 70 (HSP70), interleukin (IL-1β), interferon-gamma (IFN-γ) and tumour necrosis factor (TNF-α) gene expression of fish fed BS8 and LL8 diets were significantly higher than those of fish fed CON diet. After 13 days of challenge test, cumulative survival rate of fish fed BS8 and LL8 diets were significantly higher than those of fish fed CON, BS7 and OTC diets. Based on these results, B. subtilis and L. lactis at 108 (CFU/g) could replace antibiotics, and have beneficial effects on growth, immunity, histology, gene expression, and disease resistance in Nile tilapia.
