Comprehensive studies reveal physiological and genetic diversity in traditional rice cultivars for UV-B sensitivity

Acclimation to crop niches for thousands of years has made indigenous rice cultivars better suited for stress-prone environments. Still, their response to UV-B resiliency is unknown. 38 rice landraces were grown in cemented pots in a randomised block design with three replicates under open field conditions in Sambalpur University in the wet season of 2022. Half of the plants in each of the cultivars were administered UV-B radiation at the panicle emergence stage in an adjustable UV-B chamber permitting sunlight, and the effects of the stress on various morpho-physiological features, such as spikelet sterility, flag leaf photosynthetic and flavonoid pigment contents, and lipid peroxidation activities, were estimated for calibration of stress resistance. The experiment identified Swarnaprabha and Lalkain as the most sensitive and resilient to stress respectively, and the differential response between them was further revealed in the expression of genes related to UV-B sensitivity. Subject to the stress, Swarnaprabha exhibited symptoms of injuries, like leaf burns, and a higher loss of various photosynthetic parameters, such as pigment contents, SPAD and Fv/Fm, ETR and qP values, while NPQ increased only in Lalkain. Exposure to UV-B increased the total phenolic and flavonoid contents in Lalkain while depressing them in Swarnaprabha. Such an effect amounted to a higher release of fluorescent energy in the latter. The magnitude of expression of the genes and the impact of the stress on photosynthetic parameters, phenolic compounds and pubescent hair structure at the panicle emergence stage could be valid indicators among indigenous rice for UV-B tolerance.

Molecular diversity analysis of deciduous pubescent oaks was conducted for populations from Calabria, Sicily and Sardinia. The aims of this study were twofold. First, to provide data on the genetic diversity of pubescent oaks from an understudied area which currently exhibits one of the highest concentrations of pubescent oak species in Europe. Second, to verify if these groups of oaks are genetically distinct and if their identification is in accordance with the current taxonomic classification. Molecular analyses of leaf material of 480 trees from seventeen populations belonging to putatively different pubescent oak species (Quercus amplifolia, Q. congesta, Q. dalechampii, Q. ichnusae, Q. leptobalanos, Q. virgiliana) were performed. Twelve gene-based Expressed Sequence Tag-Simple Sequence Repeat markers were selected, and genetic diversity and differentiation were calculated. The results showed relatively high values of allelic richness, heterozygosity and number of private alleles for the populations investigated. A weak but positive correlation between geographical and genetic distance was detected. Genetic assignment (STRUCTURE) and principle coordinate analyses exhibited a weak separation into two genetic groups which, however, did not correspond to the taxonomic, chorological and ecological features of the populations investigated. Sardinian populations formed one group which was separated from the Calabrian and Sicilian populations. In light of the results obtained, the taxonomic classification for the pubescent white oaks currently reported in the major Italian floras and checklists for the study area was not confirmed by molecular analyses.

Quercus pubescens
(Willd.) is an ectomycorrhizal (ECM) tree species that is capable of withstanding occasional drought events, but the response of its ectomycorrhiza to drought is not well known. An experiment with two rain exclusion plots and two natural precipitation regime plots was established in a secondary sub-Mediterranean oak forest. ECM roots were sampled before the experiment and after 11 months of rain exclusion. ECM root tips were divided into vital and non-vital and quantified. Morphoanatomical characterization and molecular identification were performed for vital ectomycorrhizae to obtain diversity indices and perform community analyses. Soil water content (SWC) in rain exclusion plots was reduced by approx. 6 vol.% relative to natural precipitation regime and was devoid of major peaks in SWC after rain events. After 11 months, ECM vitality and species richness were significantly reduced on rain exclusion plots compared to the natural precipitation regime while ECM community was reduced to a small subset of the most frequent morphotypes, with strongly decreased number of the unique morphotypes. The reduction of unique morphotypes as a result of rain exclusion may compromise the functional diversity of ectomycorrhiza in their role of nutrient uptake, while the reduction of ECM vitality may decrease the absorptive surface for water and nutrients.

Adapting forests to climate change: an experiment to promote different tree species in mixed Scots pine forests (Pinus sylvestris L.)

Key message
We have applied various silvicultural treatments to enhance the adaptation of Scots pine (
Pinus sylvestris
L.) mixed forests to climate change in Montesquiu Castle Park (Catalonia, NE Spain). Some treatments have shown positive effects, such as increased growth, reduced defoliation, and greater resistance of Scots pine to drought. However, other treatments may lead to a shift in vegetation from pine-dominated to oak-dominated forests. Future extreme droughts could increase pine mortality, potentially accelerating this shift. These findings are significant for forest management aimed at adapting these species to climate change in their southern distribution range: forest thinning could improve Scots pine’s ability to cope with stress, while pine removal may promote the growth of pubescent oak.
Context
Forested systems around the globe are being modified and climate change is one of the main drivers. Many regions of Spain, especially in the south and the east, where aridity is predicted to increase, could be some of the most vulnerable places for Scots pine (
Pinus sylvestris
L.) in Western Europe. In some cases, defoliation, mortality, and lack of regeneration of this species have induced a vegetation shift, as has been seen with
Quercus
spp. Adaptive forest management might help adjust the vulnerable forest systems to new climatic conditions.
Aims
This study, carried out in north-eastern Spain, applies silvicultural treatments to promote changes in species composition for improving the adaptation to climate change of a Scots pine mixed forest. The main objective is to evaluate how different silvicultural treatments give rise to more adapted stands in terms of survival, growth, and regeneration.
Methods
Three experimental treatments (and one control) were applied, two of them to reduce competition for Scots pine and a third pursuing the acceleration of replacement of Scots pine by pubescent oak (
Quercus pubescens
Willd.). The response of the stands to the treatments was monitored during 6 years.
Results
Mortality of Scots pine was nil or very low in the different treatments, but defoliation showed significant differences among treatments at the end of the study: 42% in the control treatment (CO), 25% in the understory clearing treatment (C), and 18% in the understory clearing and pine thinning treatment. The increment in the basal area of Scots pine between 2015 and 2021 did not show significant differences among silvicultural treatments (

> 0.05), but that of pubescent oak was higher in the pine logging than in the other treatments. Regeneration of Scots pine and pubescent oak did not differ among silvicultural treatments.
Conclusions
These findings have relevant implications for the use of management to adapt forests to climate change: in its southern distribution range, forest thinning could improve the capacity of Scots pine to cope with stress conditions, whereas pine removal may promote pubescent oak.

Countermovement Jump Performance Is Related to Ankle Flexibility and Knee Extensors Torque in Female Adolescent Volleyball Athletes

Ankle flexibility and isokinetic knee torque/power generating capacity were previously suggested to contribute or to be correlated to the vertical countermovement jump (CMJ) performance. The aim of this study was to investigate the effect of the passive ankle joint dorsi flexion (θPDF) and the knee muscle’s isokinetic torque and power on the CMJ in adolescent female volleyball players. The θPDF at a knee extension angle of 140 degrees were measured for 37 female post-pubertal volleyball players. Then, the players were assigned to either the flexible (n = 10) or inflexible (n = 14) groups according to earlier recommended criteria. Testing included the CMJ with and without an arm swing, and maximal knee extensions and flexions in 3 angular velocities on an isokinetic dynamometer. CMJ height performed with or without an arm swing (r(22) = 0.563, p = 0.040 and r(22) = 0.518, p = 0.009, respectively) and relative power (r(22) = 0.517, p = 0.010 and r(22) = 0.446, p = 0.030, respectively) were positively correlated with the extensors’ torque at 180°/s and were negatively correlated with the flexibility level of the dominant side ankle (r(22) = −0.529, p = 0.008 and r(22) = −0.576, p = 0.030, respectively). A moderate positive correlation was also revealed between the CMJ height with and without an arm swing and the power of the non-dominant knee extensors (r(22) = 0.458, p = 0.024 and r(22) = 0.402, p = 0.049, respectively) and flexors (r(22) = 0.484, p = 0.016 and r(22) = 0.477, p = 0.018, respectively). Results of the 2 × 2 repeated ANOVA measurements revealed that flexible players jumped significantly (p < 0.05) higher during the CMJs, whilst there was a group effect only on the isokinetic knee extensor muscles’ torque. In conclusion, a more flexible ankle joint and a higher isokinetic knee extensor’s torque generating capacity resulted in higher CMJ performance. Therefore, ankle flexibility should be emphasized in training and is suggested to be included in preseason screening tests of youth female volleyball players.

Pubescence is an important phenotypic trait observed in both vegetative and generative plant organs. Pubescent plants demonstrate increased resistance to various environmental stresses such as drought, low temperatures, and pests. It serves as a significant morphological marker and aids in selecting stress-resistant cultivars, particularly in wheat. In wheat, pubescence is visible on leaves, leaf sheath, glumes and nodes. Regarding glumes, the presence of pubescence plays a pivotal role in its classification. It supplements other spike characteristics, aiding in distinguishing between different varieties within the wheat species. The determination of pubescence typically involves visual analysis by an expert. However, methods without the use of binocular loupe tend to be subjective, while employing additional equipment is labor-intensive. This paper proposes an integrated approach to determine glume pubescence presence in spike images captured under laboratory conditions using a digital camera and convolutional neural networks.
Initially, image segmentation is conducted to extract the contour of the spike body, followed by cropping of the spike images to an equal size. These images are then classified based on glume pubescence (pubescent/glabrous) using various convolutional neural network architectures (Resnet-18, EfficientNet-B0, and EfficientNet-B1). The networks were trained and tested on a dataset comprising 9,719 spike images.
For segmentation, the U-Net model with EfficientNet-B1 encoder was chosen, achieving the segmentation accuracy IoU = 0.947 for the spike body and 0.777 for awns. The classification model for glume pubescence with the highest performance utilized the EfficientNet-B1 architecture. On the test sample, the model exhibited prediction accuracy parameters of F1 = 0.85 and AUC = 0.96, while on the holdout sample it showed F1 = 0.84 and AUC = 0.89. Additionally, the study investigated the relationship between image scale, artificial distortions, and model prediction performance, revealing that higher magnification and smaller distortions yielded a more accurate prediction of glume pubescence.

Chronological Sequence of Leaf Phenology, Xylem and Phloem Formation and Sap Flow of Quercus pubescens from Abandoned Karst Grasslands

Intra-annual variations in leaf development, radial growth, including the phloem part, and sap flow have rarely been studied in deciduous trees from drought-prone environments. In order to understand better the chronological order and temporal course of these processes, we monitored leaf phenology, xylem and phloem formation and sap flow in
from abandoned karst grasslands in Slovenia during the growing season of 2014. We found that the initial earlywood vessel formation started before bud opening at the beginning of April. Buds started to open in the second half of April and full leaf unfolding occurred by the end of May. LAI values increased correspondingly with leaf development. About 28% of xylem and 22% of phloem annual increment were formed by the time of bud break. Initial earlywood vessels were fully lignified and ready for water transport, indicating that they are essential to provide hydraulic conductivity for axial water flow during leaf development. Sap flow became active and increasing contemporarily with leaf development and LAI values. Similar early spring patterns of xylem sap flow and LAI denoted that water transport in oaks broadly followed canopy leaf area development. In the initial 3 weeks of radial growth, phloem growth preceded that of xylem, indicating its priority over xylem at the beginning of the growing season. This may be related to the fact that after bud break, the developing foliage is a very large sink for carbohydrates but, at the same time, represents a small transpirational area. Whether the interdependence of the chronological sequence of the studied processes is fixed in
needs to be confirmed with more data and several years of analyses, although the 'correct sequence' of processes is essential for synchronized plant performance and response to environmental stress.

Photosynthetic performance and water relations in young pubescent oak (Quercus pubescens) trees during drought stress and recovery

The capability to withstand and to recover from severe summer droughts is becoming an important issue for tree species in central Europe, as dry periods are predicted to occur more frequently over the coming decades. Changes in leaf gas exchange, chlorophyll a fluorescence and leaf compounds related to photoprotection were analysed in young Quercus pubescens trees under field conditions during two summers (2004 and 2005) of progressive drought and subsequent rewatering. Photochemistry was reversibly down-regulated and dissipation of excess energy was enhanced during the stress phase, while contents of leaf pigments and antioxidants were almost unaltered. Plant water status was restored immediately after rewatering. Net photosynthesis (Pn) measured at ambient CO₂ recovered from inhibition by drought within 4 wk. Pn measured at elevated CO₂- to overcome stomatal limitations - was restored after a few days. A network of photoprotective mechanisms acted in preserving the potential functionality of the photosynthetic apparatus during severe drought, leading to a rapid recovery of photosynthetic activity after rewatering. Thus, Q. pubescens seems to be capable of withstanding and surviving extreme drought events.

The mistletoe, Viscum album, living on Scots pine (Pinus sylvestris) has been reported barely to regulate its transpiration and thus heavily to affect the gas exchange of its host. The extent of this mistletoe effect and its underlying mechanism has, so far, only been partially analysed. In this study, pine branches with different mistletoe infestation levels were investigated by sap flow gauges and analysed with a modelling approach to identify the mistletoe-induced stomatal regulation of pine and its consequences for the water and carbon balances of the tree. It was found that Viscum album barely regulates its stomata and that pines consequently compensate for the additional water loss of mistletoes by closing their own stomata. Despite the reduced stomatal aperture of the needles, the total water loss of branches with mistletoes increased. Furthermore, the increasingly closed stomata reduced carbon assimilation for the pine. Such a negative effect of the mistletoes on pine’s stomatal conductance and carbon gain was particularly strong during dry periods. Our study therefore suggests that mistletoe-induced stomatal closure is a successful mechanism against dying from hydraulic failure in the short term but increases the risk of carbon starvation in the long term. With the current conditions in Valais, Switzerland, a tree with more than about 10–20% of its total leaf area attributable to mistletoes is at the threshold of keeping a positive carbon balance. The currently increasing mistletoe abundance, due to increasing mean annual temperatures, is therefore accelerating the ongoing pine decline in many dry inner-Alpine valleys.
