Effect of Foliar Spray Application of Zinc Oxide Nanoparticles on Quantitative, Nutritional, and Physiological Parameters of Foxtail Millet (Setaria italica L.) under Field Conditions
It has been shown that the foliar application of inorganic nano-materials on cereal plants during their growth cycle enhances the rate of plant productivity by providing a micro-nutrient source. We therefore studied the effects of foliarly applied ZnO nanoparticles (ZnO NPs) on Setaria italica L. foxtail millet’s quantitative, nutritional, and physiological parameters. Scanning electron microscopy showed that the ZnO NPs have an average particle size under 20 nm and dominant spherically shaped morphology. Energy dispersive X-ray spectrometry then confirmed ZnO NP homogeneity, and X-ray diffraction verified their high crystalline and wurtzite-structure symmetry. Although plant height, thousand grain weight, and grain yield quantitative parameters did not differ statistically between ZnO NP-treated and untreated plants, the ZnO NP-treated plant grains had significantly higher oil and total nitrogen contents and significantly lower crop water stress index (CWSI). This highlights that the slow-releasing nano-fertilizer improves plant physiological properties and various grain nutritional parameters, and its application is therefore especially beneficial for progressive nanomaterial-based industries.
Identification of QTL regions and candidate genes for growth and feed efficiency in broilers
Background Feed accounts for about 70% of the total cost of poultry meat production. Residual feed intake (RFI) has become the preferred measure of feed efficiency because it is phenotypically independent of growth rate and body weight. In this study, our aim was to estimate genetic parameters and identify quantitative trait loci (QTL) for feed efficiency in 3314 purebred broilers using a genome-wide association study. Broilers were genotyped using a custom 55 K single nucleotide polymorphism (SNP) array. Results Estimates of genomic heritability for seven growth and feed efficiency traits, including body weight at 28 days of age (BW28), BW42, average daily feed intake (ADFI), RFI, and RFI adjusted for weight of abdominal fat (RFIa), ranged from 0.12 to 0.26. Eleven genome-wide significant SNPs and 15 suggestively significant SNPs were detected, of which 19 clustered around two genomic regions. A region on chromosome 16 (2.34–2.66 Mb) was associated with both BW28 and BW42, and the most significant SNP in this region, AX_101003762, accounted for 7.6% of the genetic variance of BW28. The other region, on chromosome 1 (91.27–92.43 Mb) was associated with RFI and ADFI, and contains the NSUN3 and EPHA6 as candidate genes. The most significant SNP in this region, AX_172588157, accounted for 4.4% of the genetic variance of RFI. In addition, a genomic region containing the gene AGK on chromosome 1 was found to be associated with RFIa. The NSUN3 and AGK genes were found to be differentially expressed in breast muscle, thigh muscle, and abdominal fat between male broilers with high and low RFI. Conclusions We identified QTL regions for BW28 and BW42 (spanning 0.32 Mb) and RFI (spanning 1.16 Mb). The NSUN3 , EPHA6 , and AGK were identified as the most likely candidate genes for these QTL. These genes are involved in mitochondrial function and behavioral regulation. These results contribute to the identification of candidate genes and variants for growth and feed efficiency in poultry.
Genetic parameters for different measures of feed efficiency and related traits in boars of three pig breeds
Residual feed intake (RFI) is commonly used as a measure of feed efficiency at a given level of production. A total of 16,872 pigs with their pedigree traced back as far as possible was used to estimate genetic parameters for RFI, growth performance, food conversion ratio (FCR), body conformation, and feeding behavior traits in 3 Danish breeds [Duroc (DD), Landrace (LL), and Yorkshire (YY)]. Two measures of RFI were considered: residual feed intake 1 (RFI1) was calculated based on regression of daily feed intake (DFI) from 30 to 100 kg on initial test weight and ADG from 30 to 100 kg (ADG2). Residual feed intake 2 (RFI2) was as RFI1, except it was also regressed with respect to backfat (BF). The estimated heritabilities for RFI1 and RFI2 were 0.34 and 0.38 in DD, 0.34 and 0.36 in LL, and 0.39 and 0.40 in YY, respectively. The heritabilities ranged from 0.32 (DD) to 0.54 (LL) for ADG2, from 0.54 (DD) to 0.67 (LL) for BF, and from 0.13 (DD) to 0.19 (YY) for body conformation. Feeding behavior traits including DFI, number of visits to feeder per day (NVD), total time spent eating per day (TPD), feed intake rate (FR), feed intake per visit (FPV), and time spent eating per visit (TPV) were moderately to highly heritable. Residual feed intake 2 was genetically independent of ADG2 and BF in all breeds, except it had low genetic correlation to ADG2 in YY (0.2). Residual feed intake 1 was also genetically independent of ADG2 in DD and LL. Both RFI traits had strong genetic correlations with DFI (0.85 to 0.96) and FCR (0.76 to 0.99). They had low or no genetic correlations with feeding behavior traits. Unfavorable genetic correlations were found between ADG2 and both BF and DFI. Among feeding behavior traits, DFI had low genetic correlations to other traits in all breeds. High and negative genetic correlations were also found between TPD with FR (-0.79 in YY to -0.88 in DD), NVD, and TPD (-0.91 in DD to -0.94 in YY) and between NVD and FPV (-0.83 in DD to -0.91 in YY) in all breeds. The genetic trend for feed efficiency was favorable in all breeds regardless of the definition of feed efficiency used. In summary, RFI1 and RFI2 were heritable and selection for reduced RFI2 can be performed without adversely affecting ADG and BF and could replace FCR in the selection index for the Danish pig breeds. Selection could also be based on RFI1 for breeds with fewer concerns about a negative effect of BF or for breeds that do not have BF records.
Genetics of residual feed intake in growing pigs: Relationships with production traits, and nitrogen and phosphorus excretion traits
Residual feed intake (RFI) is defined as the difference between the observed ADFI and the ADFI predicted from production and maintenance requirements. The objectives of this study were to evaluate RFI as a selection criterion to improve feed efficiency and its potential to reduce N and P excretion in 4 pig breeds. Data were collected between 2000 and 2009 in French central test stations for 2 dam breeds [French Landrace (LR) and Large White (LWD)], and 2 sire breeds [Large White (LWS) and Piétrain (PP)]. Numbers of recorded pigs were 6407, 10,694, 2342, and 2448 for the LR, LWD, LWS, and PP breeds, respectively. All PP animals were genotyped for the halothane mutation. This data set was used to calculate RFI equations for each of the 4 breeds, and to estimate genetic parameters for RFI together with growth, carcass, and meat quality traits, and N and P excretion during the test period (35 to 110 kg BW). The RFI explained 20.1% in PP, 26.5% in LWS, 27.6% in LWD, and 29.5% in LR of the phenotypic variability of ADFI. The PP breed differed from the others in this respect, probably due to a lower impact of the variation of body composition on ADFI. Heritability estimates of RFI ranged from 0.21 ± 0.03 (LWD) to 0.33 ± 0.06 (PP) depending on the breed. Heritabilities of N and P excretion traits ranged from 0.29 ± 0.06 to 0.40 ± 0.06. The RFI showed positive genetic correlations with feed conversion ratio (FCR) and excretion traits, these correlations being greater in the sire breeds (from 0.57 to 0.86) than in the dam breeds (from 0.38 to 0.53). Compared with FCR, RFI had weaker genetic correlations with carcass composition, growth rate, and excretion traits. Estimates of genetic correlations between FCR and excretion traits were very close to 1 for all breeds. Finally, excretion traits were, at the genetic level, correlated positively with ADFI, negatively with growth rate and carcass leanness, whereas the halothane n mutation in PP was shown to reduce N and P excretion levels. To conclude, new selection indexes including RFI can be envisaged to efficiently disentangle the responses to selection on growth rate and body composition from those on feed efficiency, with favorable impacts on N and P excretions, particularly in sire pig breeds. However, the switch from FCR to RFI in selection indexes should not resolve the genetic antagonism between feed efficiency and meat quality.
The evolutionary potential of diet-dependent effects on lifespan and fecundity in a multi-parental population of Drosophila melanogaster
The nutritional conditions experienced by a population have a major role in shaping trait evolution in many taxa. Constraints exerted by nutrient limitation or nutrient imbalance can influence the maximal value that fitness components such as reproduction and lifespan attains, and organisms may shift how resources are allocated to different structures and functions in response to changes in nutrition. Whether the phenotypic changes associated with changes in nutrition represent an adaptive response is largely unknown. Further, it is unclear whether the response of fitness components to diet even has the potential to evolve in most systems. In this study, we use an admixed multi-parental population of Drosophila melanogaster reared in three different diet conditions to estimate quantitative genetic parameters for lifespan and fecundity. We find significant genetic variation for both traits in our population and show that lifespan has moderate to high heritabilities within diets. Genetic correlations for lifespan between diets were significantly less than one, demonstrating a strong genotype by diet interaction. These findings demonstrate substantial standing genetic variation in our population that is comparable to natural populations and highlights the potential for adaptation to changing nutritional environments.
Psychometric validation of the household food insecurity access scale among Inuit pregnant women from Northern Quebec
Globally, food insecurity is a major public health concern. In North America, it is particularly prevalent in certain sub-groups, including Indigenous communities. Although many Indigenous and remote communities harvest and share food, most food security assessment tools focus on economic access. This study describes the psychometric evaluation of a modified Household Food Insecurity Access Scale (HFIAS), developed for mixed economies, to assess food insecurity among pregnant Inuit women. The HFIAS was administered to 130 pregnant women in Nunavik (Arctic region of Quebec), Canada. Data were fit to a Rasch Rating Scale Model (RSM) to determine the discrimination ability of the HFIAS. Person parameter (Theta) estimates were calculated based on the RSM to provide a more accurate scoring system of the modified HFIAS for this population. Theta values were compared to known correlates of food insecurity. Comparative fit indices showed preference for a modified version of the HFIAS over the original. Theta values displayed a continuum of severity estimates and those values indicating greater food insecurity were consistently linked to known correlates of food insecurity. Participants living in households with more than 1 hunter (Theta = -.45) or more than 1 fisher (Theta = -.43) experienced less food insecurity than those with no hunters (Theta = .48) or fishers (Theta = .49) in their household. The RSM indicated the scale showed good discriminatory ability. Subsequent analyses indicated that most scale items pertain to the classification of a household as moderately food insecure. The modified HFIAS shows potential for measuring food insecurity among pregnant women in Nunavik. This is an efficient instrument that can inform interventions targeting health conditions impacting groups that obtain food through both monetary and non-monetary means.
Assessing the Influence of Viral Infection on ‘Tribidrag’ Grapevines: Insights from Two Vegetation Seasons
The objective of this study was to investigate the response of the grapevine variety ‘Tribidrag’ to virus infection over two vegetation seasons. Virus-free plants were greenhouse cultivated and green grafted with five different virus inocula composed of grapevine leafroll-associated virus 3 (GLRaV-3) singly or in coinfection with other most economically important grapevine viruses. Changes in nutrient status and photosynthesis-related parameters, along with symptom development, were measured. Using the quantitative PCR method, the relative concentration of five selected Vitis genes was determined. Cluster analysis and ANOVA revealed the reduction in phosphorus concentration (P) and photosynthesis-related parameters in infected plants in both seasons, even in the absence of symptom expression, indicating P and assimilation rate (Photo (A)) as stable markers of virus infection. Plants infected with inoculum Y composed of five different viruses provoked major significant changes in the first season while, in the second, fewer changes were measured. The sucrose synthase 3 gene was upregulated in infected plants confirming disturbed sugar metabolism related to virus-induced stress. This study showed that virus-induced changes in ‘Tribidrag’ plants even in the absence of symptoms are dependent on plant age, as well as on the composition of virus inocula.
Selection for feed efficiency using the social effects animal model in growing Duroc pigs: evaluation by simulation
Background Traits recorded on animals that are raised in groups can be analysed with the social effects animal model (SAM). For multiple traits, this model specifies the genetic correlation structure more completely than the animal model (AM). Our hypothesis was that by using the SAM for genetic evaluation of average daily gain (ADG) and backfat thickness (BF), a high rate of improvement in feed conversion ratio (FCR) might be achieved, since unfavourable genetic correlations between ADG and BF reported in a Duroc pig line could be partially avoided. We estimated genetic and non-genetic correlations between BF, ADG and FCR on 1144 pigs using Bayesian methods considering the SAM; and responses to selection indexes that combine estimates of indirect (IGE) and direct (DGE) genetic effects for ADG and BF by stochastic simulation. Results Estimates of the ratio of the variance of DGE to the phenotypic variance were 0.31, 0.39 and 0.25 and those of the total genetic variance to the phenotypic variance were 0.63, 0.74 and 0.93 for ADG, BF and FCR, respectively. In spite of this, when the SAM was used to generate data and for the genetic evaluations, the average economic response was worse than that obtained when BV predictions from the AM were considered. The achieved economic response was due to a direct reduction in BF and not to an improvement in FCR. Conclusions Our results show that although social genetic effects play an important role in the traits studied, their proper consideration in pig breeding programs to improve FCR indirectly is still difficult. The correlations between IGE and DGE that could help to overcome the unfavourable genetic correlations between DGE did not reach sufficiently high magnitudes; also, the genetic parameters estimates from the SAM have large errors. These two factors penalize the average response under the SAM compared to the AM.
Genetic variability of metabolic characteristics in chickens selected for their ability to digest wheat
Improving feed efficiency remains crucial for poultry production. Birds have previously been selected on their ability to digest their diet, as assessed by AMEn (Apparent ME corrected for 0 nitrogen). Such selection, for either a high (D+) or a low AMEn (D-), affects energy, nitrogen, lipid, and starch digestibility. The aim of this study was to establish whether selection on the digestive ability of birds modified metabolic traits. A total of 630 broiler chickens of the eighth generation of a divergent selection experiment on AMEn were used for this purpose. A balance trial was performed to determine energy, nitrogen, and phosphorus retention. Growth performance was recorded and body protein and lipid deposition assessed by breast and abdominal fat yields. Tibia development and mineralization were also studied and heat production was indirectly assessed through the measurement of body temperature during fasting and feeding. Phenotypic correlations estimated within line showed that an increased efficiency was associated to fatter birds and more solid bones in D- but not in D+ line, whereas increased consumption was associated with more solid bones in D+ but not in D- line. The heritability estimates for metabolic traits were relatively high, except for temperature traits (from 0.08 to 0.12), ranging from 0.28 to 0.56 for body composition, and from 0.38 to 0.77 for bone characteristics. Breast meat yield did not differ between the 2 lines whereas a slight increase in abdominal fat yield was observed in the high-digestion line (D+). The relative dry tibia weights and ash weights were greater in D+ birds (+6.56 and +8.06%, respectively) but the lengths and the diameters of the tibia were lower (-7.89 and -3.77%, respectively). Finally, AMEn was poorly correlated with almost all metabolic traits (ranging from -0.10 to 0.20), indicating that the ability of the animal to digest its diet is genetically independent of post-digestion metabolic traits.
Parent outcome expectancies for purchasing fruit and vegetables: a validation
To validate four scales -- outcome expectancies for purchasing fruit and for purchasing vegetables, and comparative outcome expectancies for purchasing fresh fruit and for purchasing fresh vegetables versus other forms of fruit and vegetables (F&V). Survey instruments were administered twice, separated by 6 weeks. Recruited in front of supermarkets and grocery stores; interviews conducted by telephone. One hundred and sixty-one food shoppers with children (18 years or younger). Single dimension scales were specified for fruit and for vegetable purchasing outcome expectancies, and for comparative (fresh vs. other) fruit and vegetable purchasing outcome expectancies. Item Response Theory parameter estimates revealed easily interpreted patterns in the sequence of items by difficulty of response. Fruit and vegetable purchasing and fresh fruit comparative purchasing outcome expectancy scales were significantly correlated with home F&V availability, after controlling for social desirability of response. Comparative fresh vegetable outcome expectancy scale was significantly bivariately correlated with home vegetable availability, but not after controlling for social desirability. These scales are available to help better understand family F&V purchasing decisions.
