Effect of gestation dietary methionine/lysine ratio on placental angiogenesis and reproductive performance of sows1

Abstract
The current study aims to evaluate the effects of different gestation dietary Met/Lys (methionine, Met/lysine) ratios on the production performance of sows. Specifically, it measured the effect of Met on plasma urea and AA concentrations and placental vascular density of pregnant sows. A total of 325 multiparous sows (third parity, Large × White) were randomly allocated to five dietary treatments (n = 65) with five dietary Met/Lys ratios 0.27 (nutrient requirements of swine [NRC] 2012 level), 0.32, 0.37, 0.42, and 0.47). The litter size and weight at birth were measured and recorded. Blood samples were obtained on days 0, 40, 90, and 114 of gestation, and placenta samples were collected at parturition. The effects of different dietary Met/Lys ratios on the reproductive performance were evaluated based on the prolificacy of sows as either high (≥13 total piglets born) or low (<13 total piglets born). The results showed that dietary Met/Lys ratio had no significant effect on the reproductive performance of lower prolificacy sows (P > 0.05). However, for high-prolificacy sows, litter weight of born alive significantly increased in 0.37 Met/Lys ratios group compared with control group (P < 0.05). The gestation dietary Met/Lys ratio showed significant quadratic effects on the litter birth weight and percentage of piglets born with weight <0.9 kg (P < 0.05), and the Met/Lys ratios to achieve the best reproductive performance determined to be 0.37. Furthermore, plasma urea concentrations of sows also changed with Met/Lys ratios quadratically (P < 0.05). Increasing dietary Met/Lys ratios elevated the concentration of most plasma AA. Although the dietary Met/Lys ratio had no significant effect on the placental vascular density (P > 0.05), the gestation dietary Met/Lys ratio showed significant quadratic effects on the placental vascular density (P < 0.05). In addition, the birth weight of piglets of high-prolificacy sows was positively correlated with the placental vascular density (P < 0.01). Taken as a whole, the dietary Met/Lys ratio showed a quadratic curve relation with birth weight performance and placental angiogenesis performance, to which 0.37 ratio contributed to the best performance of high-prolificacy sows.

Porcine sperm binding to oviduct cells and glycans as supplements to traditional laboratory semen analysis1

Abstract
Accurate semen evaluation is necessary to maintain high reproductive efficiency but difficult to accomplish. The objective was to determine if the ability to bind oviduct cells or oviduct glycans are useful supplements to traditional semen analyses. Measuring binding to specific soluble glycans is less laborious than assessing binding to oviduct cell aggregates and more suitable for routine use. Previous work has shown that sperm binding to oviduct cells improves fertility prediction, possibly by estimating the ability of sperm to form an oviduct reservoir. The two oviduct glycan motifs, biantennary 6-sialylated N-acetyllactosamine (bi-SiaLN) and LewisX trisaccharide (LeX), that bind boar spermatozoa with high affinity and specificity were tested. Semen from 30 boars was shipped overnight for laboratory analysis and for inseminations to determine fertility (n = 3 replicates). Oviduct cell binding and traditional sperm analyses including motility and morphology were completed. Additionally, binding to soluble fluoresceinated glycans bi-SiaLN, sulfated LeX (suLeX), and the control lactosamine disaccharide (LacNAc) was measured. Inseminations were at 15 farms (>50 matings per boar) in the Midwest and farrowing data from all matings were used. Pregnancy rate (PR) and litter size (LS) were adjusted to account for different farms, number of services, number of doses inseminated, and sow parity, using the MIXED procedure in SAS 9.4. A fertility index (FI) was generated, consisting of PR × LS, to estimate boar overall fertility. Finally, the GLMSELECT procedure was used to select variables having a significant impact on PR, LS, and FI. The predictive models constructed were further analyzed using the REG procedure and accounted for 58% or more of the variation in PR, LS, and FI [PR (P < 0.001, r2 = 0.60), LS (P < 0.001, r2 = 0.58), and FI (P < 0.001, r2 = 0.63)]. The final model for PR includes oviduct cell binding as well as boar age, % distal droplets, head morphology, tail morphology, beat/cross frequency, and curvilinear velocity. The final model for LS includes boar age, % distal droplets, tail morphology, and overall morphology. Finally, the FI model included boar age, % distal droplets, head morphology, tail morphology, curvilinear velocity, and semen volume per ejaculate. Although binding to intact oviduct cells was impactful as a means to predict PR, binding to specific soluble oviduct glycans was not a useful supplement to traditional semen analysis.

Glycine treatment enhances developmental potential of porcine oocytes and early embryos by inhibiting apoptosis1

Abstract
Glycine, a component of glutathione (GSH), plays an important role in protection from reactive oxygen species (ROS) and inhibition of apoptosis. The aim of this study was to determine the effect of glycine on in vitro maturation (IVM) of porcine oocytes and their developmental competence after parthenogenetic activation (PA). We examined nuclear maturation, ROS levels, apoptosis, mitochondrial membrane potential (ΔΨm), and ATP concentration, as well as the expression of several genes related to oocyte maturation and development. Our studies found that treatment with glycine in IVM culture medium increased nuclear maturation rate, but varying the concentrations of glycine (0.6, 6, or 12 mM) had no significant effect. Furthermore, 6 mM glycine supported greater blastocyst formation rates and lesser apoptosis after PA than the other concentrations (P < 0.05). All the glycine treatment groups had decreased levels of ROS in both matured oocytes and at the 2-cell stage (P < 0.05). At the 2-cell stage, the 6 mM glycine group had ROS levels that were lesser than the other 2 glycine treatment groups (0.6 and 12 mM). From this, we deemed 6 mM to be the optimal condition, and we then investigated the effects of 6 mM glycine on gene expression. The expression of both FGFR2 and Hsf1 were greater than the control group in mature oocytes. The glycine treatment group had greater levels of expression of an antiapoptotic gene (Bcl2) in mature oocytes and cumulus cells and lesser levels of expression of a proapoptotic gene (Bax) in PA blastocysts (P < 0.05). In addition, mitochondrial ΔΨm and ATP concentration were increased in 6 mM glycine group compared with the control group. In conclusion, our results suggest that glycine plays an important role in oocyte maturation and later development by reducing ROS levels and increasing mitochondrial function to reduce apoptosis.

Abstract
The effects of hyperleptinemia and leptin resistance during gestation are unclear. Leptin, an important neuroendocrine regulator, has anorexic effects, but its interactions with other metabolic hormones during pregnancy are unclear. We examined potential roles of leptin in regulating prolactin (PRL), GH, and melatonin plasma concentrations during pregnancy in Polish Longwool ewes. Twelve estrus-synchronized ewes carrying twins after mating were randomly assigned to receive i.v. injections of saline or recombinant ovine leptin (2.5 or 5.0 µg/kg BW). Blood samples were collected (15-min intervals over 4 h) immediately before the first injection at dusk and kept under red light. Treatments were repeated at 2-wk intervals, starting before mating and continuing from days 30 to 135 of gestation. Concentrations of plasma PRL, GH, and melatonin were determined using a validated RIA. The effects of leptin on hormone plasma concentrations varied depending on pregnancy stage and leptin dose. PRL plasma concentrations were affected at most stages of pregnancy and before gestation. In non-, very early- (day 30), and late- (day 120 and 135) pregnant ewes, exogenous leptin stimulated PRL (P < 0.001) plasma concentrations, while during the second month of gestation, it decreased PRL concentrations (P < 0.01). Leptin affected GH plasma concentrations (P < 0.05) only during the first 2 mo of pregnancy, with no effects during the second part of gestation or before pregnancy. In early-pregnant ewes (day 30 and 45), leptin decreased melatonin plasma concentrations (P < 0.05), but at day 60, leptin stimulated melatonin plasma concentrations at low (P < 0.01) and high doses (P < 0.05), with no effects in ewes after 105 d of gestation. These data indicate specific pregnancy-induced endocrine adaptations to changes in energy homeostasis, supporting the hypothesis that leptin affects PRL, GH, and melatonin release during gestation.
