Overview
为研究桃果实酯类香气的生物合成,克隆了桃醇酰基转移酶基因AAT,并研究了外源乙烯对该基因mRNA转录水平的影响.根据桃基因组测序结果,采用RACE和反转录PCR技术克隆到1个与醇酰基转移酶基因同源的cDNA,序列全长1 486 bp,CDS区长度为1 353 bp,编码450个氨基酸,命名为PpAA T2.PpAA T2蛋白属于PLN02481超级家族,含有植物转移酶基因2个高度保守结构域HXXXD和D(N) F(V) GWG.基因位于桃第5条染色体,BAC注释为1条包含1个内含子的mRNA序列转录链,CDS区有2处碱基突变,其中,555 bp处碱基由C突变为G,导致丙氨酸突变为缬氨酸.AAT基因响应乙烯表达,0℃冷藏桃施加外源乙烯促进AAT基因表达水平;该基因在叶片、花和成熟果肉中均表达,且在叶片和花中的表达丰度比果实高,推测其在不同的生理过程中均发挥作用.
