植物种子中,油脂主要贮藏在一种分散的、相对稳定的亚细胞微滴“油体”中,油质蛋白Oleosin是油体结构组成和功能调节的最主要蛋白质,在种子成熟、休眠和萌发过程中发挥重要作用.通过油桐种仁EST文库构建与测序,获得了油桐Oleosin (OLE)蛋白编码基因的5个转录本VfOLE1、VfOLE2、VfOLE3、VfOLE4、VfOLE5,完整阅读框长度分别为474、465、414、414、429 bp.通过实时荧光定量PCR技术分析了5个VfOLE转录本在油桐不同组织以及种仁成熟过程中的表达变化.结果表明,5个VfOLE转录本在成熟的种子中特异高效表达,其它组织中微弱表达;5个VfOLE表达趋势与桐酸、桐油积累趋势一致,其中在桐酸快速合成、桐油快速积累时期,VfOLE1、VfOLE2表达迅速上升并且表达量显著高于VfOLE3、VfOLE4、VfOLE5.试验结果为进一步研究VfOLE基因在桐油合成中的生物学功能提供了参考.
