Overview
最近几年,表皮生长因子受体（EGFR）已经成为肿瘤学药物研发的一个最重要靶点。抑制EGFR酪氨酸激酶（TK）区的小分子药物已成为治疗非小细胞肺癌（NSCLC）的有力新型武器。基于TK区突变与EGFRTK抑制剂（TKIs）获得持久显著疗效之间具有相关性，已经设计了一系列试验以检验这些药物作为一线治疗的可能性，并且为将来以基因型为指导的靶向治疗开拓了广阔前景。最新对这些药物获得性耐药的生物学基础有了更深入的认识，从而有望进一步提高这类药物的临床疗效。本综述对EGFR治疗NSCLC的生物学机制、预测EGFRTKIs疗效的临床和分子指标、患者特异性分子谱的应用以及未来临床和基础科学的研究方向做一总结。
