Overview
目的探讨脑源性神经营养因子(BDNF)基因多态性与海洛因依赖的相关性。方法对符合纳入及排除标准的海洛因依赖组308例及健康对照组317例进行临床资料收集与血样采集。采用飞行时间质谱分析方法对BDNF rs16917204、rs6265、rs7103873、rs7103873、rs56164415、rs13306221、rs2030323共7个多态性位点进行分型,数据采用SPSS 20.0和HaploView 4.0软件进行统计学分析。结果 rs6265位点基因型频率分布在海洛因依赖组及健康对照组中存在统计学差异(χ2=15.151,P=0.001)。与健康对照组相比,海洛因依赖组rs6265位点G等位基因频率显著高于健康对照组(χ2=9.864,P=0.002,OR=1.429,95%CI=1.143~1.786)。rs13306221位点基因型频率分布在海洛因依赖组及健康对照组中存在统计学差异(χ2=7.699,P=0.006)。与健康对照组相比,海洛因依赖组G等位基因频率显著高于健康对照组(χ2=7.137,P=0.008,OR=0.539,95%CI=0.340~0.853)。连锁不平衡检验发现,由rs6265与rs71038731(block1)构建的单倍型为强连锁不平衡(D〉0.9;r2〉0.8)。在海洛因依赖组及健康对照组中,G-G单倍型频率分布存在显著性差异,健康对照组中G-G单倍型频率显著高于海洛因依赖组(χ2=4.546,P=0.033)。结论 BDNF基因rs6265及rs13306221多态性位点可能与海洛因依赖有关,携带有rs6265位点G等位基因及rs13306221位点G等位基因的个体更易形成海洛因依赖,携带有G-G单倍型(rs6265-rs71038731)的个体相对不易对海洛因产生依赖。
