摘要:目的研究组蛋白去乙酰化酶抑制剂(SAHA)对胃癌细胞株MGC-803增殖、凋亡及周期的影响。方法MTT比色法观察不同药物浓度SAHA作用于培养胃癌细胞株MGC-803时的增殖变化,初步选取药物作用48h时的合适药物浓度,进一步用流式细胞术检测1、2μmol/L SAHA对MGC-803细胞凋亡及周期的影响,Westernblot检测乙酰化组蛋白3、4及cyclinD1蛋白的表达情况。结果与正常对照组相比,sAHA处理组细胞增殖能力降低,凋亡率升高,细胞周期内G1期比率升高,差异均具有统计学意义(P〈0.05);Westernblot检测结果显示,与正常对照组相比,SAHA处理组的乙酰化组蛋白3、乙酰化组蛋白4升高,而eyclinD1含量降低,差异均具有统计学意义(P〈0.01)。结论组蛋白去乙酰化酶抑制剂SAHA可以抑制MGC-803细胞增殖,通过G1-S期阻滞抑制细胞周期,促进细胞凋亡,可能与促进乙酰化组蛋白3及乙酰化组蛋白4的表达增多有关。
