三种裸鼠胰腺癌建模方法的比较

Overview
目的建立人胰腺癌细胞株BXPC-3裸鼠皮下瘤、腹腔内种植转移瘤以及原位移植瘤模型,探讨模型建立方法及其生物学特性。方法采用直接皮下瘤细胞注射法建立胰腺癌裸鼠皮下瘤模型,开腹后瘤块包埋法建立腹腔内种植转移瘤模型,瘤细胞悬液置入胰腺包膜下建立原位移植模型,观察三种模型接种阳性率及瘤块生长的速度,对移植瘤做病理检查。结果 3种模型构建的阳性率分别为:皮下瘤模型47%(7/15),腹腔种植转移瘤模型100%(14/14),原位移植模型100%(14/14)。后两种模型1月后发生肝转移、腹膜转移、肠系膜转移,3组间肿瘤生长速度比较,原位移植模型最快,腹腔种植转移模型其次,皮下瘤模型最慢,差异具有统计学意义(P<0.01)。病理检查符合胰腺低分化腺癌。结论成功建立了胰腺癌BXPC-3细胞株的皮下移植瘤模型、腹腔种植转移模型及原位移植模型,显示了各种模型的独特优势,为胰腺癌生物学行为的基础研究及治疗提供了实验平台。
