Overview
为深入了解高原鼢鼠(Eosplax baileyi)高原适应性的生物学基础,应用生物信息学相关软件分析高原鼢鼠肉碱棕榈酰基转移酶1A(carnitine palmitoyl transferase 1A,CPT-1A)基因序列的同源性和趋同进化位点,利用qRT-PCR方法测定不同海拔条件下(3 700 m和2 700 m)高原鼢鼠肝脏、骨骼肌和脂肪组织中CPT-1A基因的表达水平。结果表明:高原鼢鼠CPT-1A基因与盲鼹鼠(Nannospalax galili)的同源性最高;高原鼢鼠、盲鼹鼠和裸鼹鼠(Heterocephalus glaber)3种地下鼠CPT-1A存在明显的趋同进化位点;随着海拔的升高,高原鼢鼠背部脂肪组织中CPT-1A基因表达水平显著升高;不同组织之间的比较发现,肝脏组织中CPT-1A基因表达量最高。以上结果提示,高原鼢鼠CPT-1A基因在进化过程中形成了特定的适应性突变,低氧上调高原鼢鼠脂肪组织中CPT-1A基因的表达,这可能与高原鼢鼠适应地下寒冷、低氧和高二氧化碳浓度的洞道生境有关,是高原鼢鼠适应地下洞道生境的分子机制之一。
