Overview
The mechanisms mediating hematopoietic stem and progenitor cell (HSPC) mobilization are complex. A recent study showed that hematopoietic colony forming capacity/functionality under the autonomic nervous system. Herein we investigated whether norepinephrine (NE) and dopamine (DA) concentration could be responsible for directly influencing granulocyte-macrophage colony forming unit (CFU-GM) functionality. Bone marrow cells were taken from femora of healthy male mice (6- 12 weeks old). The influence of NE and DA on marrow CFU-GM was analyzed in vitro under normal appropriate granulocyte-macrophage colony forming unit (CFU-GM) assays. Specific concentrations (10'8 M) of free NE, DA showed a significant impairment of growing CFU-GM. Thus, NE and DA were thought to trigger CFU-GM mobilization and proliferation capacity suggesting that high concentration of both NE and DA have a direct negative effect on mobilization of marrow cellularity. Furthermore, results also show that beta and alpha -adrenergic antagonists such as propranolol and prazosin could directly counter-act the effect of NE on GM-CFU.
