乌灵胶囊对卒中后抑郁大鼠学习记忆障碍的影响

Overview
目的观察乌灵胶囊对卒中后抑郁大鼠学习记忆功能的影响,探讨其与认知改善及海马脑源性神经营养因子（BDNF）表达变化的关系。方法 40只成年雄性SD大鼠随机均分为对照组、模型组、艾斯西肽普兰治疗组及乌灵胶囊治疗组,每组10只。对照组不作手术及慢性应激处理,模型组用选择性右侧大脑中动脉栓塞结合连续3周慢性不可预见性温和应激方法建立卒中后抑郁大鼠模型,艾斯西肽普兰治疗组及乌灵胶囊治疗组在慢性应激处理期间分别给予艾斯西肽普兰0.2mg/（kg.d）及乌灵胶囊100mg/（kg.d）,每日1次,连续21d。采用糖水偏爱测试和Morris水迷宫试验评价大鼠抑郁程度及学习、记忆功能,采用Western blotting检测大鼠海马BDNF的表达情况。结果模型组大鼠认知功能及BDNF表达均明显低于对照组和治疗组（P〈0.05）。在慢性应激处理的同时,给予艾斯西肽普兰治疗后,大鼠认知功能改善,BDNF表达强度明显提高,与对照组比较无统计学差异（P〉0.05）。给予乌灵胶囊治疗后,大鼠学习记忆障碍改善,但异常的BDNF表达无改变。结论乌灵胶囊能改善卒中后抑郁大鼠的学习记忆障碍,其作用机制与BDNF的表达变化无关。
