Overview
目的探讨丙泊酚麻醉对新生大鼠海马脑源性神经营养因子(BDNF)mRNA、BDNF前体(proBDNF)/成熟型BDNF(m BDNF)表达的影响及其与学习记忆功能的关系。方法 108只新生7d SD大鼠随机分为对照组(C组)、丙泊酚单次麻醉组(P1组)和丙泊酚多次麻醉组(P2组),每组36只。C组腹腔注射生理盐水7.5ml/(kg.d),连续7d;P1组腹腔注射生理盐水7.5ml/(kg.d),连续6d,第7天腹腔注射丙泊酚75mg/kg;P2组腹腔注射丙泊酚75mg/(kg.d),连续7d。各组于第7天注射完毕后15min随机选取6只大鼠,心室穿刺采血,测定血糖及血气分析;于注射完毕后6h、24h、48h、72h及30d,每组随机抽取6只大鼠,分离海马组织,采用RT-PCR检测BDNF mRNA的表达,Western blotting检测m BDNF、proBDNF蛋白的表达;各组取6只大鼠喂养至出生25d进行Morris水迷宫实验,测试空间学习记忆能力。结果 C组大鼠海马BDNF mRNA及m BDNF蛋白表达水平逐渐升高,proBDNF蛋白表达水平及proBDNF/m BDNF比值逐渐降低。与C组比较,P1组各时间点海马BDNF mRNA及mBDNF蛋白表达水平下降(P〈0.05),proBDNF/m BDNF比值升高(P〈0.05),大鼠幼年期逃逸潜伏期延长,空间探索时间缩短(P〈0.05);与C组及P1组比较,P2组各时间点海马BDNF mRNA及m BDNF蛋白表达水平下降,proBDNF蛋白表达及proBDNF/m BDNF比值升高(P〈0.05),大鼠幼年期逃逸潜伏期延长,空间探索时间缩短(P〈0.05)。结论丙泊酚麻醉可导致大鼠幼年期学习记忆功能减退,且多次麻醉的效果强于单次麻醉,其机制可能与BDNF mRNA、proBDNF)/m BDNF表达改变有关。
