Overview
Suppressor of Fused (SuFu) is an intracellular mediator of the Hedgehog signaling pathway with diverse biological roles, including a tumour suppressor function. Recent data suggest an essential role for this gene in cell differentiation in various tissues and cancers. Despite its known role as a tumour suppressor in medulloblastoma, a neuroectodermal tumour of cerebellar origin, the functional importance of SuFu in the developing cerebellum remains unknown. In this thesis, using in vivo conditional knockout approaches and in vitro assays, I present evidence for a novel requirement for SuFu during mid-hindbrain specification, cerebellar patterning and cell differentiation.
